Do alternative stable states exist in large shallow Taihu Lake,China?

Regime shifts from submersed macrophyte dominance to phytoplankton dominance have been widely reported in small-to medium-sized shallow lakes.However,alternative stable states in large shallow lakes(surface area>500 km^(2))remain unconfirmed.To understand the alternative stable states and the main influencing factors of submersed macrophytes in large lakes,the ecosystem states from monitoring data from 1959 to 2019 in large shallow Taihu Lake(2338 km^(2)in average depth of 2.12 m)in China were examined.Changes in submersed macrophyte coverage(C_(Mac))and phytoplankton chlorophyll a(Chl a)in the time series and their relationships with environmental factors were analyzed.During the field investigation from August 2018 to May 2019,nutrients and Chl a showed obvious heterogeneity across the lake,being generally higher in the western and northern areas and lower in the southeast area,while C_(Mac)was only observed in the eastern areas,e.g.,East Taihu Lake,Xukou Bay,and Gonghu Bay.During the long-term monitoring from 1959 to 2019 in the Central Region,Meiliang Bay,and East Taihu Lake,Chl a increased significantly in the time series.C_(Mac)varied slightly among different subareas,always at low levels(liang Bay but at relatively high levels in East Taihu Lake(10%–90%).Frequency distributions of response variables had no multimodality except for C_(Mac)in East Taihu Lake,with two peaks between 15%and 20%and between 55%and 60%.A dual relationship was found between Chl a and total phosphorus(TP)in the areas with and without macrophytes,while C_(Mac)showed no relationship with TP,and submersed macrophytes did not flourish in the Central Region and Meiliang Bay even when TP was at very low levels(≈10 mg/m3).Taihu Lake had similar algal turbidity(TurbAlg)as small-to mediumsized lakes but generally presented with higher values of nonalgal turbidity(TurbNonAlg),as did their contribution to total turbidity as a percentage.This study suggested that large shallow Taihu Lake may have no alternative

采用稀释涂布法分离培养2020年6—9月采自山东省威海市乳山牡蛎养殖区的长牡蛎(Crassostrea gigas)胃和鳃部位共附生可培养细菌,通过形态观察和16S rRNA基因测序进行多样性分析,同时筛选副溶血性弧菌的拮抗菌。结果表明:共分离获得细菌422株,删除重复后共获得不同批次细菌258株,主要包括变形菌门、厚壁菌门、放线菌门和拟杆菌门4个类群,分别占比72.2%、12.6%、11.8%和3.4%,共囊括了6个纲,63个属和119个种,并分离到1个潜在新种。分离的不同种类弧菌可能与牡蛎夏季高死亡率有关,并筛选出一株杀鱼假交替单胞菌ML-231对副溶血性弧菌有较强的抑制作用。本研究首次报道了乳山长牡蛎共附生可培养细菌多样性,发现多种致病性弧菌,并获得1株副溶血性弧菌拮抗菌,对今后乳山长牡蛎的生产管理及弧菌防治具有一定参考价值。

随着国内天然气管道的大规模建设,我国长输天然气用管线钢及钢管的生产制造技术也迅速发展并达到世界先进水平。天然气管道仍将朝着高强度、大口径、高压力、大壁厚、低温服役等方向发展,对高钢级管线钢特别是抗大变形管线钢、低温环境用管线钢等提出了更高的要求。本文着重从管线钢的合金成分体系、组织类型、轧制工艺的演变及其与管线钢的性能间的关系等角度,对高钢级管线钢、抗大变形管线钢及低温环境用管线钢的发展现状进行了综述,并结合目前应用中存在的一些问题对我国长输天然气管道用钢的发展趋势进行了探讨。

Orbital and Millennial-Scale Climate Variability over the Past 76 ka in the Western Tarim Basin, Northwest China

Paleoclimate changes during the last glacial in the arid central Asia are not as well understood as the monsoon-dominated areas of Asia. Here we report a 75-m-long sediment core over past 120 kyr based on astronomical tuning combined with the optically stimulated luminescence(OSL) dating in the Kashi depression of the western Tarim Basin, Northwest China. Analysis of grain size and highresolution gamma ray(GR) logs from the KT11 borehole across the last glacial period yields a climate history for the Tarim Basin which reflects the variations of its temperature and the hydrological cycles. Comparison of these records with north hemisphere summer insolation, the Greenland ice core temperature, stalagmites and the loess from the nearby region indicates that the deposits in the fluvio-lacustrine system of the Kashi depression responded to climate change at the Younger Dryas(YD), six Heinrich cooling events and the Dansgaard-Oeschger cycles. Our work indicates that the alternations between warm-humid and cold-dry climates were prevalent in the western Tarim Basin during the last glacial period, showing an in-phase pattern with the climate variations of the East Asian Monsoon, controlled ultimately by precession and North Atlantic Ocean climate variability on orbital-millennial time scales.

Seasonal photoperiodic influence of pineal melatonin on hypothalamic-pituitary-adrenal axis-hippocampal-receptor in male rats

Background: Based on the effect of seasonal changes on human visceral function, this study investigated the impact of seasonal photoperiod of the pineal body on hypothalamic-pituitary-adrenal axis-hippocampal-receptor in rats, aiming to reveal the mechanism by which pineal gland melatonin regulates the seasonal secretion of hippocampal neurotransmitters.Methods: Vernal equinox, summer solstice, autumn equinox, and winter solstice were selected as four experimental time points, and rats were randomly divided into normal control group, sham operation group, and pinealectomized group. The seasonal changes in corticotropin-releasing hormone(CRH),adrenocorticotropic hormone(ACTH), corticosterone, hypothalamic melatonin receptor(MTR), and hippocampal corticosterone receptor(CORTR) were examined by enzyme-linked immunosorbent assay.Results: Comparing the same group between different seasons, we showed that in the normal control group, CRH, ACTH, corticosterone, and MTR were higher, while CORTR was lower in autumn and winter than in spring(all P <.05). Compared with the normal control group, the pinealectomized group showed higher levels of corticosterone(P =.01), MTR(P =.01), and CORTR(P =.03) during spring;reduced levels of MTR and CORTR(both P <.001) during summer;higher levels of ACTH(P =.001) and MTR(P <.001),and lower levels of CRH(P =.001), corticosterone(P <.001), and CORTR(P =.003) during autumn;and lower levels of CRH(P <.001) and MTR(P =.004), and higher level of ACTH(P <.001) in winter.Conclusions: Seasonal photoperiod acts on the pineal gland to secrete different levels of melatonin,resulting in seasonal changes in the hypothalamic-pituitary-adrenal axis-hippocampal-receptor, which may be the pathophysiological basis for the onset of seasonal affective disorder.

Lymph node(LN)metastasis is one of the predominant metastatic routes of non-small cell lung cancer(NSCLC)and is considered as a leading cause for the unsatisfactory prognosis of patients.Although lymphangiogenesis is well-recognized as a crucial process in mediating LN metastasis,the regulatory mechanism involving lymphangiogenesis and LN metastasis in NSCLC remains unclear.In this study,we employed high-throughput sequencing to identify a novel circular RNA(circRNA),circTLCD4-RWDD3,which was significantly upregulated in extracellular vesicles(EVs)from LN metastatic NSCLC and was positively associated with deteriorated OS and DFS of patients with NSCLC from multicenter clinical cohort.Downregulating the expression of EV-packaged circTLCD4-RWDD3 inhibited lymphangiogenesis and LN metastasis of NSCLC both in vitro and in vivo.Mechanically,circTLCD4-RWDD3 physically interacted with hnRNPA2B1 and mediated the SUMO2 modification at K108 residue of hnRNPA2B1 by upregulating UBC9.Subsequently,circTLCD4-RWDD3-induced SUMOylated hnRNPA2B1 was recognized by the SUMO interaction motif(SIM)of AliX and activated AliX to recruit ESCRT-III,thereby facilitating the sorting of circTLCD4-RWDD3 into NSCLC cell-derived EVs.Moreover,EV-packaged circTLCD4-RWDD3 was internalized by lymphatic endothelial cells to activate the transcription of PROX1,resulting in the lymphangiogenesis and LN metastasis of NSCLC.Importantly,blocking EV-mediated transmission of circTLCD4-RWDD3 via mutating SIM in AliX or K108 residue of hnRNPA2B1 inhibited the lymphangiogenesis and LN metastasis of NSCLC in vivo.Our findings reveal a precise mechanism underlying SUMOylated hnRNPA2B1-induced EV packaging of circTLCD4-RWDD3 in facilitating LN metastasis of NSCLC,suggesting that EV-packaged circTLCD4-RWDD3 could be a potential therapeutic target against LN metastatic NSCLC.

Adoptive cellular immunotherapy with chimeric antigen receptor(CAR)T cells has emerged as a novel modality for treating relapsed and/or refractory B-cell non-Hodgkin lymphoma(B-NHL).With increasing approval of CAR T-cell products and advances in CAR T cell therapy,CAR T cells are expected to be used in a growing number of cases.However,CAR T-cell-associated toxicities can be severe or even fatal,thus compromising the survival benefit from this therapy.Standardizing and studying the clinical management of these toxicities are imperative.In contrast to other hematological malignancies,such as acute lymphoblastic leukemia and multiple myeloma,anti-CD19 CAR T-cell-associated toxicities in B-NHL have several distinctive features,most notably local cytokine-release syndrome(CRS).However,previously published guidelines have provided few specific recommendations for the grading and management of toxicities associated with CAR T-cell treatment for B-NHL.Consequently,we developed this consensus for the prevention,recognition,and management of these toxicities,on the basis of published literature regarding the management of anti-CD19 CAR T-cell-associated toxicities and the clinical experience of multiple Chinese institutions.This consensus refines a grading system and classification of CRS in B-NHL and corresponding measures for CRS management,and delineates comprehensive principles and exploratory recommendations for managing anti-CD19 CAR T-cell-associated toxicities in addition to CRS.

目的研究中药芸香(Ruta graveolens L.)中香豆素类化学成分,并探讨其抗炎、抗凝活性。方法本实验利用各种色谱分离技术,结合核磁共振波谱及质谱联用技术对分离得到的单体化合物进行结构鉴定。CCK-8法筛选出无明显细胞毒性的化合物,用于对脂多糖诱导小鼠巨噬细胞(RAW 264.7)释放NO的检测,进行抗炎活性筛选,通过凝血酶原时间的测试进行抗凝活性筛选。结果从芸香提取物中共鉴定出10个香豆素类化合物,分别为补骨脂素(1)、花椒毒素(2)、chalepensin(3)、rutamontine(4)、3-(1',1',dimethyl allyl)-herniarin(5)、佛手苷内酯(6)、异莨菪亭(7)、异嗪皮啶(8)、去甲基软木花椒素(9)、Chalepin(10)。对10个化合物的抗炎、抗凝进行活性评价,结果显示,化合物2、4、9在20μmol/L时对脂多糖(lipopolysaccharide,LPS)诱导的NO生成有较好的抑制作用,抑制率分别为54.3%±1.7%、51.2%±5.5%、52.3%±1.7%;10个化合物在浓度为1.0 mmol/L条件下,均未表现出抗凝活性。结论从芸香提取物中分离得到10个香豆素类单体化合物,化合物3~5、7~10为首次从芸香中分离得到。该研究结果丰富了芸香的化学成分和生物活性研究,为芸香抗炎活性的开发和利用提供了理论基础。

Multifunctional,wearable,and durable textiles integrated with smart electronics have attracted tremendous attention.However,it remains a great challenge to balance new functionalities with high-temperature stability.Herein,textile-based pressure sensors with excellent electromagnetic interference(EMI)shielding,Joule heating,and high-temperature resistance were fabricated by constructing graphene/SiC(G/SiC)heterostructures on carbon cloth via laser chemical vapor deposition(LCVD).The resultant textiles exhibited excellent EMI efficiency of 74.2 dB with a thickness of 0.45 mm,Joule heating performance within a low working voltage(V)range of 1-3 V,and fast response time within 20 s.These properties arose from multiple reflections,interfacial polarization,and high conductivity due to the numerous amounts of nanoscale G/SiC heterostructures.More importantly,G/SiC/carbon fibers(CFs)demonstrated well high-temperature resistance with a heat resistance index(THri)of 380.2 C owing to the protection of a coating layer on the CFs upon oxidation.Meanwhile,the G/SiC/CFs presented good pressure-sensing performance with high sensitivity(S)of 52.93 kPal,fast response time of 85 ms,and a wide pressure range of up to 186 kPa.These features imply the potential of the G/SiC/CFs as efficient EMI shielding,electrical heater,and piezoresistive sensor textiles.

Polymers and composites with high thermal conductivity are promising yet challenging for the growing demand of thermal management in electrical and electronic equipment.Guided by the closest packing model,a multiscale filling Al_(2)O_(3) strategy was designed and incorporated with epoxy resin(EP)to form high thermal conductive composites in this work.Epoxy composites with single filler loading were also prepared.The microstructures,thermal,rheological,and dielectric characteristics of the multiscale filling Al_(2)O_(3)/EP composites have been investigated.Compared with single-scale Al_(2)O_(3) filled epoxy based composites,it is found that the multiscale filling Al_(2)O_(3)/EP composites exhibit higher thermal conductivity under the same filler loading of 50 vol%,which is attributed to the efficient heat conduction paths formed by appropriate multiscale fillers.Particularly,a remarkably improved thermal conductivity of 2.707 W m^(-1) K^(-1) was acquired in Al_(2)O_(3)/EP composites at filler loading of 50 vol%(5 mm Al_(2)O_(3)(26.67 vol%),30 mm Al_(2)O_(3)(27.41 vol%)and 70 mm Al_(2)O_(3)(45.92 vol%)),which is about 1300%higher than that of the pure epoxy resin.In addition,the dielectric constant of the Al_(2)O_(3)/EP composites were significantly improved while keeping the dielectric loss almost unchanged.The finite element simulation further verified the effectiveness of improving the thermal conductivity of materials in the heat dissipation of electrical equipment.Therefore,this research provides a simple strategy for manufacturing high thermal conductive composite materials with a wide range of potential applications as packaging materials.
