light is a fundamental environmental factor for living organisms on earth—not only as a primary energy source but also as an informational *** fungi,light can be used as an indicator for both time and space to control important physiological and morphological *** cinerea(***)is a devastating phytopathogenic fungus that exploits light cues to optimize virulence and the balance between conidiation and sclerotia development,thereby improving its dispersal and survival in ***,the components and mechanisms underlying these processes remain ***,we identify a novel light-signaling component in ***,BcCfaS,which encodes a putative cyclopropane fatty-acyl-phospholipid *** is strongly induced by light at the transcriptional level and plays a crucial role in regulating *** of BcCfaS results in reduced vegetative growth,altered colony morphology,impaired sclerotial development,and enhanced conidiation in a lightdependent ***,the mutant exhibits serious defects in stress response and virulence on the *** on a lipidomics analysis,a number of previously unknown fungal lipids and many BcCfaS-regulated lipids are identified in ***,including several novel phospholipids and fatty ***,we find that BcCfaS controls conidiation and sclerotial development by positively regulating methyl jasmonate(MeJA)synthesis to activate the transcription of light-signaling components,revealing for the first time the metabolic base of photomorphogenesis in ***,we propose that BcCfaS serves as an integration node for light and lipid metabolism,thereby providing a regulatory mechanism by which fungi adapt their development to a changing light *** new findings provide an important target for antifungal design to prevent and control fungal disease.

白鹤滩水电站尾水系统为超大地下洞室群,洞室类型多、断面大、隧洞长、结构复杂,灌浆工程量巨大。同时存在多洞段同时施工,同一洞段多工序作业。针对工程灌浆量巨大、工期紧、强度高,且与混凝土浇筑施工交叉干扰存在问题,对超大洞室群灌浆施工组织与管理进行了系统分析研究,并将成果成功运用于该项目的灌浆施工组织与管理,有效提高施工进度,节约了成本,产生了较好的经济和社会效益。

Mercury ions are highly toxic and can accumulate along food chains in water,soil,crops and *** detection of mercury ions in various media is of great significance for maintaining the ecological environment and protecting people's *** this work,a mercury ions specific fluorescent probe was developed by a simple one-step reaction of commercial substrates of 4-chloro-7-nitro-2,1,3-benzoxadiazole and 1-(2-aminoethyl)-*** on sensing behavior showed that this probe had high sensitivity and selectivity towards mercury ***,this probe could be used as a tool to track the level of mercury ions in living *** living cells,the probe with green emission emitted a bright red fluorescence when it was bound to mercury *** Arabidopsis thaliana,similar red emission could be detected from the root tip and stalk when *** was grown in culture medium containing mercury *** imaging in zebrafish showed that mercury ions were mainly concentrated in the stomach and head of ***,this probe could be applied in quantitative analysis of mercury ions in tap water,green tea,sea shrimp and *** work provided a practical tool for the detection of mercury ions in living systems and quantitative analysis in real samples.

萍水河湘东段区域受洪水灾害影响较大,历年损失严重。本文使用MIKE FLOOD构建了萍水河湘东段区域洪水演进一二维耦合模型,结合2019年实测水位过程和调查洪痕等资料验证了模型合理性。根据研究区设防情况,确定了20年、50年和100年一遇3种设计洪水计算方案,并分别提取了3种计算方案下的洪水风险特征要素(包括最大淹没水深、最大演进流速和淹没历时等),使用综合风险度法开展了萍水河湘东段区域洪水风险区划研究。结果表明:萍水河湘东段区域洪水风险区划分为极高、高、中和低风险4个等级;其中,中下游无堤防保护区域洪水风险等级为极高或高风险;湘东区县城区域洪水风险等级主要为高风险和中风险,县政府驻地区域属于洪水灾害高风险等级区。本文洪水风险区划成果可为湘东区防洪规划及非工程措施建设提供科学依据。

CpG DNA enhances the im-mune responses elicited by the DNA vaccine against foot-and-mouth disease virus in guinea pigs

CpG DNA is DNA sequence that has immune stimulatory effects. Several lines of investigation over the past few years indicate that CpG DNA plays an important role in the induction of immune responses to DNA vaccines. In this study, CpG DNA-containing synthetic oligodeoxynucleotide (CpG-ODN) was cloned into the eukaryotic expression plasmid encoding a fusion protein containing b- galactosidase from E. coli and immunogenic epitopes of foot- and-mouth disease virus (FMDV) type O, and the immune responses induced by the plasmid were assayed. The results showed that guinea pigs immunized with the recombinant plasmid containing CpG-ODN generated a higher level of FMDV-neutralizing antibody and a stronger T cell proliferative response and protection against viral challenge than those receiving the plasmid containing no CpG-ODN. Our study demonstrated that it is an effective route to enhance the efficacy of DNA vaccines by inserting exogenous CpG DNA into the plasmids, and the DNA vaccine developed here is a promising candidate to prevent FMDV infection.

TBAB hydrate formation and growth in a microdevice under static and dynamic conditions

The natural gas hydrate has become one of the most promising future green energy sources on the *** natural gas hydrates mostly exist in the sediments with porous structure, so a solid understanding of the hydrate formation and growth processes in the porous medium is of significance for the exploitation of natural gas hydrate. The micro-packed bed device is one of the efficient microfluidic devices in the engineering field, but it has been rarely used for the hydrate-based research. In this study, a transparent micro-packed bed device filled with glass beads was developed to mimic the porous condition of sediments, and used to in-situ visualize the hydrate formation and growth habits in the pore spaces under both static and dynamic conditions. For the static experiment, two types of hydrate growth patterns in porous medium were observed and identified in the micro-packed bed device, which were the graincoating growth and pore-filling growth. For the dynamic condition, the hydrate formation, growth,distribution habits and hydrate blockage phenomena in the pore spaces were in-situ visually *** impacts of flowrate and subcooling on the pressure variation of the micro-packed bed and the duration of the hydrate growth under dynamic flow condition in pores were in-situ monitored and analyzed. The higher flowrate could result in the faster hydrate growth and more severe blockage in pores, but the effect of subcooling condition might be less significant at the high flowrate.

Highly efficient CO_(2) capture using 2-methylimidazole aqueous solution on laboratory and pilot-scale

To date,the primary industrial carbon capture approach is still absorption using aqueous solutions of ***,to pursue a substitute for the amine-based approach to improve the CO_(2) capture efficiency and decrease the energy cost further,we report a new carbon capture approach using a 2-methylimidazole(mIm)aqueous *** properties and sorption behaviors of this approach have been experimentally *** results show that the mIm solution has higher CO_(2) absorption capacity under relatively higher equilibrium pressure(>130 kPa)and lower desorption heat than the methyldiethanolamine solution.91.6%sorption capacity of mIm solution can be recovered at 353.15 K and 80 *** selectivity for CO_(2)/N_(2) and CO_(2)/CH_(4) can reach an exceptional 7609 and 4324,***,the pilot-scale tests were also performed,and the results demonstrate that more than 98%of CO_(2) in the feed gas could be removed and cyclic absorption capacity can reach 1 mol·L^(-1).This work indicates that mIm is an excellent alternative to alkanolamines for carbon capture in the industry.

The clathrate hydrate memory effect is a fascinating phenomenon with potential applications in carbon capture,utilization and storage(CCUS),gas separation,and gas storage as it can accelerate the secondary formation of clathrate ***,the underlying mechanism of this effect remains *** gain a better understanding of the mechanism,we conducted molecular dynamic simulations to simulate the initial formation and reformation processes of methane *** this work,we showed the evolution process of hydrate residual structures into hydrate *** simulation results indicate that the residual structures are closely related to the existence of hydrate memory effect,and the higher the contribution of hydrate dissociated water to the hydrate nucleation process,the faster the hydrate *** hydrate dissociation,the locally ordered structures still exist after hydrate dissociation and can promote the formation of cluster structures,thus accelerating hydrate ***,the nucleation process of hydrate and the formation process of clusters are *** size of clusters composed of cup-cage structures is critical for hydrate *** residence time at high temperature after hydrate decomposition will affect the strength of the hydrate memory *** simulation results provide microscopic insights into the occurrence of the hydrate memory effect and shed light on the hydrate reformation process at the molecular scale.
