Single nucleus/cell RNA-seq of the chicken hypothalamic-pituitary-ovarian axis offers new insights into the molecular regulatory mechanisms of ovarian development

The hypothalamic-pituitary-ovarian (HPO) axis represents a central neuroendocrine network essential for reproductive function. Despite its critical role, the intrinsic heterogeneity within the HPO axis across vertebrates and the complex intercellular interactions remain poorly defined. This study provides the first comprehensive, unbiased, cell type-specific molecular profiling of all three components of the HPO axis in adult Lohmann layers and liangshan yanying chickens. Within the hypothalamus, pituitary, and ovary, seven, 12, and 13 distinct cell types were identified, respectively. Results indicated that the pituitary adenylate cyclase activating polypeptide (PACAP), follicle-stimulating hormone (FSH), and prolactin (PRL) signaling pathways may modulate the synthesis and secretion of gonadotropin-releasing hormone (GnRH), FSH, and luteinizing hormone (LH) within the hypothalamus and pituitary. In the ovary, interactions between granulosa cells and oocytes involved the KIT, CD99, liFR, FN1, and ANGPTL signaling pathways, which collectively regulate follicular maturation. The SEMA4 signaling pathway emerged as a critical mediator across all three tissues of the HPO axis. Additionally, gene expression analysis revealed that relaxin 3 (RLN3), gastrin-releasing peptide (GRP), and cocaine- and amphetamine regulated transcripts (CART, also known as CARTPT) may function as novel endocrine hormones, influencing the HPO axis through autocrine, paracrine, and endocrine pathways. Comparative analyses between Lohmann layers and liangshan yanying chickens demonstrated higher expression levels of GRP, , RLN3, , CARTPT, , LHCGR, , FSHR, , and GRPR in the ovaries of Lohmann layers, potentially contributing to their superior reproductive performance. In conclusion, this study provides a detailed molecular characterization of the HPO axis, offering novel insights into the regulatory mechanisms underlying reproductive biology.

Laparoscopy-assisted D2 radical distal gastrectomy for advanced gastric cancer： initial experience

Background Laparoscopy-assisted radical gastrectomy is gaining acceptance for treating early gastric cancer. However, few reports concerning the effectiveness of laparoscopy-assisted D2 radical distal gastrectomy （LADG） for advanced gastric cancer or data comparing the results obtained after open distal gastrectomy （ODG） are yet available. The aim of this study was to evaluate the method, feasibility and clinical result of LADG for advanced gastric cancer. Methods A retrospective study was performed comparing LADG and ODG for advanced gastric cancer. Seventy-eight patients who underwent LADG were compared with 90 patients who underwent ODG in terms of pathologic findings, operative outcome, and complications. Results There was no conversion to open surgery in the LADG group and no postoperative mortality of any patients. There were no significant differences between LADG and ODG in operative time （（245±35） vs （220±620） minutes）, complication rate （7.7% vs 10.0%）, and number of lymph nodes （23.5±6.0 vs 21.0±7.5）, while the blood loss was less after LADG （（110±25） vs （196±30） ml, P 〈0.05）. The time to postoperative flatus and postoperative hospital stay were shorter after LADG （（73.0±8.5） vs （102.0±10.5） hours, and （8.6±1.2） vs （12.1 ±2.5） days, P 〈0.05, respectively）. Conclusion LADG for advanced gastric cancer is feasible, safe, and minimally invasive.

Construction and expression of eukaryotic plasmids containing lamivudine-resistant or wild-type strains of Hepatitis B Virus genotype C

AIM:To construct eukaryotic expression plasmids of full-length Hepatitis B Virus(HBV) genotype C genome,which contain lamivudine-resistant mutants(YIDD,YVDD) or wild-type strain(YMDD) ,and to observe the expression of HBV DNA and antigens [hepatitis B surface antigen(HBsAg) and hepatitis B e antigen(HBeAg) ] of the recombinant plasmids in HepG2 cells. METHODS:Three HBV full-length genomes were amplified from the plasmids pMD18T-HBV/YIDD,pMD18T-HBV/YVDD and pMD18T-HBV/YMDD,using PCR. Three recombinant plasmids were generated by inserting each of the PCR products into the eukaryotic expression vector pcDNA3.1(+) ,between the EcoRI and HindⅢ sites. After being characterized by restriction endonuclease digestion,and DNA sequence analysis,the recombinant plasmids were transfected into HepG2 cells. At 48 and 72 h post-transfection,the levels of intracellular viral DNA replication were detected by real-time PCR,and the expression of HBsAg and HBeAg in the cell culture supernatant was determined by EliSA. RESULTS:Restriction endonuclease digestion and DNA sequence analysis confirmed that the threerecombinant plasmids were correctly constructed. After transfecting the plasmids into HepG2 cells,high levels of intracellular viral DNA replication were observed,and HBsAg and HBeAg were secreted into the cell culture supernatant. CONCLUSION:Eukaryotic expression plasmids pcDNA3.1(+) -HBV/YIDD,pcDNA3.1(+) -HBV/YVDD or pcDNA3.1(+) -HBV/YMDD,which contained HBV genotype C full-length genome,were successfully constructed. After transfection into HepG2 cells,the recombinant plasmids efficiently expressed HBV DNA,HBsAg and HBeAg. Our results provide an experimental basis for the further study of HBV lamivudine-resistant mutants.

Tiered clustering-based management architecture in mega-satellite networks

Hybrid orbital networking and the proliferation of satellites raise explosive management complexity in mega-satellite networks(MSNs) and prompt the management architecture to the hierarchy. In this regard, we propose a tiered clustering-based management architecture(TCMA) to realize the high-efficient and low-cost management performance. Specifically, based on the constrained satellite coverage, we derive an expression of the mapping relationship between the required control overhead of the controller interaction,the average path length of the proposed TCMA, and the corresponding number of management tiers and cluster size. The expression can determine the optimal number of tiers of the TCMA by minimizing the required flow table size between controllers, while ensuring high transmission efficiency. We then exhibit the elasticity of the hierarchical TCMA by adding the peer connections at the second-highest management tier to reduce maximum redundant paths. Simulation results verify the effectiveness of the proposed TCMA and illustrate that a 3-tier TCMA is sufficient for thousands of satellites.

The source of lithium in Lakkor Co Salt Lake on Qinghai-Tibet Plateau, China: evidence from hydrochemical characteristics and boron isotope

The availability of lithium resources is of great significance for the development of modern technologies, as well as for civil and military industries. The Qinghai-Tibet Plateau is a region known for its abundance of lithium-rich salt lakes. However, the specific origin of lithium in these lakes is still unknown, which hinders the advancement of the lithium resource business in this region. To research this issue, this study involved the collection of 20 samples from Lakkor Co Salt Lake on Qinghai-Tibet Plateau, encompassing samples of surface brine, cold springs, fresh lakes, and recharge rivers. The composition of anions and cations in these samples was determined. Furthermore, the analysis extensivelyutilizedthePiperthree-linediagram,Gibbs model, and ion proportion coefficient. The findings of this study indicate that as the moves from the recharge water system to salt lake, there is a transition in water type from strong carbonate to moderate carbonate and weak carbonate,as well as Na sulfate. This research based on a similar source of both lithium and boron, utilized ion correlation analysis and boron isotope study in the Lakkor Co area, and analyzed the source and transporting process of lithium. The main origin of lithium in Lakkor Co is the dissolution of lithiumrich rocks, recharge water systems, and deep hydrothermal fluids. These findings are highly significant in enhancing the foundational data of lithium-rich brine resources in the Qinghai-Tibet Plateau and are beneficial for assessing the future development of such deposits.

Influence of High-Density Bedding Plane Characteristics on Hydraulic Fracture Propagation in Shale Oil Reservoir

The existence of high-density bedding planes is a typical characteristic of shale oil *** the behavior of hydraulic fracturing in high-density laminated rocks is significant for promoting shale oil *** this study,a hydraulic fracturing model considering tensile failure and frictional slip of the bedding planes is established within the framework of the unified pipe-interface element method(UP-IEM).The model developed for simulating the interaction between the hydraulic fracture and the bedding plane is validated by comparison with experimental *** hydraulic fracturing patterns in sealed and unsealed bedding planes are ***,the effects of differential stress,bedding plane permeability,spacing,and the friction coefficient of the bedding plane are *** results showed that a single main fracture crossing the bedding planes is more likely to form in sealed bedding planes under high differential *** decrease in bedding plane permeability and the increase in the friction coefficient also promote the fracture propagating perpendicular to the bedding *** with high-density bedding planes has a poorer fracturing effect than that with low-density bedding planes,as the hydraulic fracture is prone to initiate and propagate along the bedding ***,higher injection pressure is needed to maintain fracture propagation along the *** increase in bedding density will lead to a smaller fracturing *** fluid seepage into the bedding planes slows shale *** is recommended that increasing the injection flow rate,selecting alternative fracturing fluids,and employing multi-well/multi-cluster fracturing may be efficient methods to improve energy production in shale oil reservoirs.
