【目的】探究不同生物引发条件对番茄冷胁迫下种子活力和幼苗生理特性的影响,为番茄的反季节露天栽培提供可行性方案。【方法】以番茄种子种都红运为试验材料,哈茨木霉和荧光假单胞菌作为引发剂,设5个处理:未引发处理(CK);哈茨木霉和荧光假单胞菌的1×10~7 CFU/m L的孢子液(T1处理)和发酵液(T2处理),1×10~6 CFU/m L的发酵液(T3处理)及无菌过滤液(T4处理)引发,回干后测定种子萌发指数;幼苗设4℃低温处理,以常温(25℃)为对照,分别测量形态指标和生理指标。【结果】4个引发处理不同程度促进了番茄种子萌发及植株生长,效果显著。通过种子萌发指数发现,引发效果T4>T3>T2>T1,就活力指数而言,分别显著提高90.00%、67.70%、51.30%和50.00%。分析60 d的幼苗形态指标发现,T4处理的株高、茎粗和茎鲜重均达最大值,T2处理的根长和茎鲜重达最大值;分析60 d幼苗的生理指标发现,T2处理的过氧化氢酶(CAT)含量最高。光合色素和可溶性糖含量则依次表现为T4>T3>T2>T1,T4处理的可溶性蛋白和脯氨酸(PRO)含量达最大值,分别显著增加71.74%和46.50%,其他处理的增幅略低于T4处理。4个引发处理的相对电导率(REC)在低温胁迫前后的变化相差不大,CK经低温胁迫后急剧升高。MDA含量在低温胁迫后的增量依次表现为T3>T4>T2>T1。相关分析结果表明,株高、茎粗、茎鲜重与植株CAT含量呈极显著正相关(P<0.01,下同),根鲜重与REC和MDA含量呈极显著负相关,与PRO含量呈极显著正相关。【结论】番茄种子经哈茨木霉和荧光假单胞菌的无菌过滤液等体积混合引发有利于提高种子活力,促进植株生长,增加植株体内的保护酶活性,最终提高番茄植株的低温耐受性。

AIM： To assess the feasibility of using BRAF, K-ras and BAT26 genes as stool-based molecular markers for detection of colorectal adenomas and hyperplastic polyps （HPs）. METHODS： We applied PCR-SSCP and direct sequencing to detect BRAF mutations of polyps and paired stool samples. Primer-mediated restriction fragment length polymorphism （RFLP） analysis and mutant-enriched PCR were used in detection of K-ras mutations of polyp tissues and paired stool samples respectively. BAT26, a microsatellite instability marker was examined by detection of small unstable alleles in a poly （A） repeat. RESULTS： No genetic alterations were detected in the 36 colonoscopically normal patients in either tissues or stools. BRAF, K-ras and BAT26 mutations were found in 4 （16%）, 10 （40%） and 3 （12%） of 25 adenoma tissues and among them, 75%, 80% and 100% of patients were observed to contain the same mutations in their corresponding stool samples. In HPs, mutations of BRAF and K-ras were detected in the tumor DNA of 2 （11.1%） and 8 （33.3%） of 18 patients respectively, all of whom had identical alterations in their stools. Taken together, the three genetic markers detected 15 （60%） of 25 adenomas and 8 （44.4%） of 18 HPs. The sensitivity of stool detection was 80% for adenomas and 100% for HPs with an overall specificity of 92% for adenomas and 100% for HPs. CONCLUSION： BRAF, K-ras and BAT26 genes have the potential to be molecular markers for colorectal adenomas and HPs, and can be used as non-invasive screening markers for colorectal polyps.

大跨径拼装H型钢叠合梁桥成为桥梁建设中的一种新型结构体系。通过拼装连接形成整体梁体,具有结构坚固、施工便捷、体积小、重量轻、拼装快、使用寿命长等优点,被广泛应用于各类公路、铁路、城市轨道交通等工程。然而,由于受产品制造精度和现场施工精度的限制,大跨径拼装H型钢叠合梁桥整体制作的精度往往无法满足设计要求。因此,为了提高该结构体系的制作精度,从桥孔群的制作精度控制方面入手,对其精度控制技术进行了研究和分析。

针对交直流混联系统的稳态特性,提出了一种增强控制策略以提高多端直流输电系统的稳定性。分布式补充控制结构通过调节换流器有功功率参考值来增强系统的角稳定性,基于对换流器公共连接点频率和角度的测量,确保在换流器达到功率限制时保持直流电网的功率平衡。在修改后的IEEE 39节点系统上进行的电磁暂态仿真验证了控制器的有效性。结果表明,使用补充控制策略的系统瞬态稳定性显著提高,临界故障清除时间从不足0.2 s提高到超过0.35 s。

AIM:To explore the therapeutic effect and main molecular mechanisms of acteoside in a glaucoma model in DBA/2J ***:Proteomics was used to compare the differentially expressed proteins of C57 and DBA/2J *** acteoside administration in DBA/2J mice,anterior segment observation,intraocular pressure(IOP)monitoring,electrophysiology examination,and hematoxylin and eosin staining were used to analyze any potential ***(IHC)assays were used to verify the proteomics ***,retinal ganglion cell 5(RGC5)cell proliferation was assessed with cell counting kit-8(CCK-8)*** domain-containing protein 4(Sertad4)mRNA and protein expression levels were measured by qRT-PCR and Western blot analysis,***:Proteomics analysis suggested that Sertad4 was the most significantly differentially expressed *** with the saline group,the acteoside treatment group showed decreased IOP,improved N1-P1 wave amplitudes,thicker retina,and larger numbers of cells in the ganglion cell layer(GCL).The IHC results showed that Sertad4 expression levels in DBA/2J mice treated with acteoside were significantly lower than in the saline *** treatment could improve RGC5 cell survival and reduce the Sertad4 mRNA and protein expression levels after glutamate ***:Sertad4 is differentially expressed in DBA/2J *** can protect RGCs from damage,possibly through the downregulation of Sertad4,and has a potential use in glaucoma treatment.

Climate models are vital for understanding and projecting global climate change and its associated ***,these models suffer from biases that limit their accuracy in historical simulations and the trustworthiness of future *** these challenges requires addressing internal variability,hindering the direct alignment between model simulations and observations,and thwarting conventional supervised learning ***,we employ an unsupervised Cycle-consistent Generative Adversarial Network(CycleGAN),to correct daily Sea Surface Temperature(SST)simulations from the Community Earth System Model 2(CESM2).Our results reveal that the CycleGAN not only corrects climatological biases but also improves the simulation of major dynamic modes including the El Niño-Southern Oscillation(ENSO)and the Indian Ocean Dipole mode,as well as SST ***,it substantially corrects climatological SST biases,decreasing the globally averaged Root-Mean-Square Error(RMSE)by 58%.Intriguingly,the CycleGAN effectively addresses the well-known excessive westward bias in ENSO SST anomalies,a common issue in climate models that traditional methods,like quantile mapping,struggle to ***,it substantially improves the simulation of SST extremes,raising the pattern correlation coefficient(PCC)from 0.56 to 0.88 and lowering the RMSE from 0.5 to *** enhancement is attributed to better representations of interannual,intraseasonal,and synoptic scales *** study offers a novel approach to correct global SST simulations and underscores its effectiveness across different time scales and primary dynamical modes.
