The realization of lossless metasurfaces with true chirality crucially requires the fabrication of three-dimensional structures,constraining experimental feasibility and hampering practical implementations.Even though the threedimensional assembly of metallic nanostructures has been demonstrated previously,the resulting plasmonic resonances suffer from high intrinsic and radiative losses.The concept of photonic bound states in the continuum(BICs)is instrumental for tailoring radiative losses in diverse geometries,especially when implemented using lossless dielectrics,but applications have so far been limited to planar structures.Here,we introduce a novel nanofabrication approach to unlock the height of individual resonators within all-dielectric metasurfaces as an accessible parameter for the efficient control of resonance features and nanophotonic functionalities.In particular,we realize out-of-plane symmetry breaking in quasi-BIC metasurfaces and leverage this design degree of freedom to demonstrate an optical all-dielectric quasi-BIC metasurface with maximum intrinsic chirality that responds selectively to light of a particular circular polarization depending on the structural handedness.Our experimental results not only open a new paradigm for all-dielectric BICs and chiral nanophotonics,but also promise advances in the realization of efficient generation of optical angular momentum,holographic metasurfaces,and parity-time symmetry-broken optical systems.

Jet grouting is one of the most popular soil improvement techniques,but its design usually involves great uncertainties that can lead to economic cost overruns in construction projects.The high dispersion in the properties of the improved material leads to designers assuming a conservative,arbitrary and unjustified strength,which is even sometimes subjected to the results of the test fields.The present paper presents an approach for prediction of the uniaxial compressive strength(UCs)of jet grouting columns based on the analysis of several machine learning algorithms on a database of 854 results mainly collected from different research papers.The selected machine learning model(extremely randomized trees)relates the soil type and various parameters of the technique to the value of the compressive strength.despite the complex mechanism that surrounds the jet grouting process,evidenced by the high dispersion and low correlation of the variables studied,the trained model allows to optimally predict the values of compressive strength with a significant improvement with respect to the existing works.Consequently,this work proposes for the first time a reliable and easily applicable approach for estimation of the compressive strength of jet grouting columns.

The confocal microscopy technique was applied for nonlinear optical characterization of single β-barium-borate（β-BBO） nanocrystals. The experimental setup allows measurements of the laser polarization-selective secondharmonic（sH） generation, and the results can be used to determine the nanocrystals＇ c-axis orientation, as well as to obtain information about their second-order susceptibility χ^（2）. The dependence of the sH signal on the laser polarization allowed the discrimination of individual particles from aggregates. The data were fitted using a model that takes into account the BBO properties and the experimental setup characteristics considering（i） the electrostatic approximation,（ii） the effects of the microscope objective used to focus the light on the sample in an epi-geometry configuration, and（iii） the symmetry of χ^（2） for the β-BBO nanocrystals. A signal at the third-harmonic frequency was also detected, but it was too weak to be studied in detail.

Background Tuberous sclerosis complex(TsC)is a genetic disorder caused by inactivating mutations in the TsCl and TsC2 genes,causing overactivation of the mechanistic(previously referred to as mammalian)target of rapamycin(mTOR)signaling pathway in fetal life.The mTOR pathway plays a crucial role in several brain processes leading to TsC-related epilepsy,intellectual disability,and autism spectrum disorder(AsD).Pre-natal or early post-natal diagnosis of TsC is now possible in a growing number of pre-symptomatic infants.Data sources We searched PubMed for peer-reviewed publications published between January 2010 and April 2023 with the terms“tuberous sclerosis",“autism",or“autism spectrum disorder""animal models",“preclinical studies",“neurobiology",and“treatment".Results Prospective studies have highlighted that developmental trajectories in TsC infants who were later diagnosed with AsD already show motor,visual and social communication skills in the first year of life delays.Reliable genetic,cellular,electroencephalography and magnetic resonance imaging biomarkers can identify pre-symptomatic TsC infants at high risk for having autism and epilepsy.Conclusions Preventing epilepsy or improving therapy for seizures associated with prompt and tailored treatment strategies for autism in a sensitive developmental time window could have the potential to mitigate autistic symptoms in infants with TsC.

BACKGROUND Anti-programmed death-1/programmed death-ligand 1(PD-1/PD-L1)immuno-therapy has demonstrated promising results on gastric cancer(GC).However,PD-L1 can express differently between metastatic sites and primary tumors(PT).AIM To compare PD-L1 status in PT and matched lymph node metastases(LNM)of GC patients and to determine the correlation between the PD-L1 status and clinicopathological characteristics.METHODs We retrospectively reviewed 284 GC patients who underwent D2-gastrectomy.PD-L1 was evaluated by immunohistochemistry(clone sP142)using the com-bined positive score.All PD-L1+PT staged as pN+were also tested for PD-L1 expression in their LNM.PD-L1(-)GC with pN+served as the comparison group.REsULTs Among 284 GC patients included,45 had PD-L1+PT and 24 of them had pN+.For comparison,44 PD-L1(-)cases with pN+were included(sample loss of 4 cases).Of the PD-L1+PT,54.2%(13/24 cases)were also PD-L1+in the LNM.Regarding PD-L1(-)PT,9.1%(4/44)had PD-L1+in the LNM.The agreement between PT and LNM had a kappa value of 0.483.Larger tumor size and moderate/severe peritumoral inflammatory response were associated with PD-L1 positivity in both sites.There was no statistical difference in overall survival for PT and LNM according to the PD-L1 status(P=0.166 and P=0.837,respectively).CONCLUsION Intra-patient heterogeneity in PD-L1 expression was observed between the PT and matched LNM.This disagreement in PD-L1 status may emphasize the importance of considering different tumor sites for analyses to select patients for immunotherapy.

This work describes the use of TiO_(2)nanotubes-based electrodes(TNT)modified with Cu_(2)O nanostructures and gold nanoparticles for the photoelectroreduction of CO_(2)to produce value-added compounds.A thin layer of polydopamine was used as both an adherent agent and an electron transfer mediator,due to itsπ-conjugated electron system.The highest production yield was achieved using a TNT@PDA/Nc/Au40%electrode,with Faradaic efficiencies of 47.4%(110.5μM cm^(-2))and 27.8%(50.4μM cm^(-2))for methanol and methane,respectively.The performance of the photoelectrodes was shown to be Cu_(2)O facet-dependent,with cubic structures leading to greater conversion of CO_(2)to methanol(43%)and methane(27%),compared to the octahedral morphology,while a higher percentage of metallic gold on the nanostructured Cu_(2)O surface was mainly important for CH4production.density functional theory(DFT)calculations supported these findings,attributing the superior photoelectrocatalytic performance of the TNT@PDA/Nc/Au40%electrode for CH4generation to the formation of an OCH3intermediate bonded to Au atoms.studies using isotope-labeling and analysis by gas chromatograph-mass(GC-Ms)demonstrated that13CO_(2)was the source for photoelectrocatalytic generation of13CH3OH and13CH313CH2OH.

Bladder cancer(BC)is the 10th most common cancer worldwide,with about 0.5 million reported new cases and about 0.2 million deaths per year.In this scoping review,we summarize the current evidence regarding the clinical implications of single-cell sequencing for bladder cancer based on PRIsMA guidelines.We searched PubMed,CENTRAL,Embase,and supplemented with manual searches through the scopus,and Web of science for published studies until February 2023.We included original studies that used at least one single-cell technology to study bladder cancer.Forty-one publications were included in the review.Twenty-nine studies showed that this technology can identify cell subtypes in the tumor microenvironment that may predict prognosis or response to immune checkpoint inhibition therapy.Two studies were able to diagnose BC by identifying neoplastic cells through single-cell sequencing urine samples.The remaining studies were mainly a preclinical exploration of tumor microenvironment at single cell level.single-cell sequencing technology can discriminate heterogeneity in bladder tumor cells and determine the key molecular properties that can lead to the discovery of novel perspectives on cancer management.This nascent tool can advance the early diagnosis,prognosis judgment,and targeted therapy of bladder cancer.

In this editorial,we comment on the article by Wang et al.This manuscript explores the potential synergistic effects of combining zanubrutinib,a novel oral inhibitor of Bruton’s tyrosine kinase,with high-dose methotrexate(HD-MTX)as a therapeutic intervention for primary central nervous system lymphoma(PCNsL).The study involves a retrospective analysis of 19 PCNsL patients,highlighting clinicopathological characteristics,treatment outcomes,and genomic biomarkers.The results indicate the combination’s good tolerance and strong antitumor activity,with an 84.2%overall response rate.The authors emphasize the potential of zanubrutinib to modulate key genomic features of PCNsL,particularly mutations in myeloid differentiation primary response 88 and cluster of differentiation 79B.Furthermore,the study investigates the role of circulating tumor DNA in cerebrospinal fluid for disease surveillance and treatment response monitoring.In essence,the study provides valuable insights into the potential of combining zanubrutinib with HD-MTX as a frontline therapeutic regimen for PCNsL.The findings underscore the importance of exploring alternative treatment modalities and monitoring genomic and liquid biopsy markers to optimize patient outcomes.While the findings suggest promise,the study’s limitations should be considered,and further research is needed to establish the clinical relevance of this therapeutic approach for PCNsL.

Antarctic Peninsula is experiencing one of the largest global warming events worldwide.shallow water bodies generated by the melting of snow in summer are numerous,and they might act as sentinels of climate change due to their rapid response and ability to integrate catchment information.shifts in climate can influence the structure of microbial communities which dominate these freshwaters ecosystems.Here,we characterize three ponds at Cierva Point(Antarctic Peninsula)by examining their physico-chemical and morphological characteristics and we explored how different factors modify the structure of the microbial community.We studied the abundance and biomass of heterotrophic bacteria,picocyanobacteria and picoeukaryote algae during January and February of two consecutive summers(2017 and 2018).We found that ponds had different limnological characteristics,due to their location,geomorphological features and presence of the surrounding flora and fauna.Physico-chemical parameters as well as microbial community differed between ponds,months and years.In 2017,most ponds were oligo to mesotrophic states.The larger accumulated rainfall(as a result of environmental changes on the Antarctic Peninsula)during 2018,particularly in February,causes nutrient runoff into water bodies.This affects those ponds with the highest seabird circulation,such as gentoo penguin,increasing eutrophication.As a result,picoplanktonic abundances were higher,and the community structure shifts to a largely heterotrophic bacteria dominated one.These results suggest that these communities could act as sentinels to environmental changes,anticipating a future with mostly hypertrophic ponds.
