Calculations of point defect energetics with Density Functional Theory(DFT)can provide valuable insight into several optoelectronic,thermodynamic,and kinetic *** calculations commonly use methods ranging from semi-local functionals with a-posteriori corrections to more computationally intensive hybrid functional *** applications of DFT-based high-throughput computation for data-driven materials discovery,point defect properties are of interest,yet are currently excluded from available materials *** work presents a benchmark analysis of automated,semi-local point defect calculations with a-posteriori corrections,compared to 245“gold standard”hybrid calculations previously *** consider three different a-posteriori correction sets implemented in an automated workflow,and evaluate the qualitative and quantitative differences among four different categories of defect information:thermodynamic transition levels,formation energies,Fermi levels,and dopability *** highlight qualitative information that can be extracted from high-throughput calculations based on semi-local DFT methods,while also demonstrating the limits of quantitative accuracy.

Metabolic and epigenetic reprogramming play important roles in cancer therapeutic ***,their interplays are poorly *** report here that elevated TIGAR(TP53-induced glycolysis and apoptosis regulator),an antioxidant and glucose metabolic regulator and a target of oncogenic histone methyltransferase NSD2(nuclear receptor binding SET domain protein 2),is mainly localized in the nucleus of therapeutic resistant tumor cells where it stimulates NSD2 expression and elevates global H3K36me2 ***,TIGAR directly interacts with the antioxidant master regulator NRF2 and facilitates chromatin recruitment of NRF2,H3K4me3 methylase MLL1 and elongating Pol-II to stimulate the expression of both new(NSD2)and established(NQO1/2,PRDX1 and GSTM4)targets of NRF2,independent of its enzymatic *** TIGAR confers cancer cell resistance to chemotherapy and hormonal therapy in vitro and in tumors through effective maintenance of redox *** addition,nuclear accumulation of TIGAR is positively associated with NSD2 expression in clinical tumors and strongly correlated with poor *** findings define a nuclear TIGAR-mediated epigenetic autoregulatory loop in redox rebalance for tumor therapeutic resistance.

Influence of Substrate Quality and Moisture Availability on Microbial Communities and Litter Decomposition

The main source of carbon (C) to soil stocks is plant litter, the decomposition of which is controlled by a mixture of physical, chemical, and biological processes. bacteria and fungi are the dominant biota responsible for decomposition, yet we know very little about their respective contributions or how community dynamics may be affected by litter quality. This study sought to gain a better understanding of the variable relationships between organic matter decomposition, litter quality, and microbial community composition, with a specific focus on distinguishing bacterial and fungal dynamics. Experiments were conducted under contrasting hydrological conditions, comparing a wetland with an upland forest environment. Decomposition of native vegetation was monitored in addition to breakdown of a common substrate (Acer rubrum (red maple) leaves) placed in both environments. In situ incubations lasted 16 months, and were sampled at ~3-month intervals. Regardless of site, maple litter decomposition proceeded at a similar rate, though we did observe differences in litter quality over time (C:N, %N, solubility of organic C). For the upland site, native litter decomposed more slowly than the maple did. At the wetland site, both litter types decomposed at a similar rate which, surprisingly, was faster than either litter type at the upland site. This finding could be attributed to water-limitation at the upland site and/or stimulation of decomposition at the wetland site due to allochthonous nutrient inputs or organic matter priming. Substrate induced respiration (SIR) was measured for native litter incubated at each sampling site, and the relative contributions of bacteria and fungi were compared. No consistent major differences were detected across these microbial groups, though we did observe much higher rates of SIR at the wetland site compared to the upland site. The long-term success of hydrogenase uptake negative legume-rhizobia associations, in spite of their apparent inefficiency, may be explained by the positive effects of H2 release to soil. A primary benefit of H2 release to soil is the stimulation of H2-oxidizing, plant growth promoting rhizobacteria (PGPR) [1]. Two such previously isolated strains were tested as seed inoculants for barley and spring wheat;there were significant differences between treatments and controls in tiller and grain head production, supported by data from greenhouse trials. T-RFLP analysis of barley soil samples, supported by DNA sequencing data, successfully distinguished both species inoculated. Successful re-isolation indicates that these isolates can reproduce themselves in soils and can be used as effective inoculants with peat as the standard carrier. This study showed that we are able to achieve some of the beneficial effects of crop rotation without the need to implement actual crop rotation.

以赤子爱胜蚓(Eisenia fetida)为供试生物,草甸棕壤为供试土壤,以蚯蚓微粒体细胞色素P450含量、抗氧化酶系以及谷胱甘肽转移酶活性为指标,进行了土壤中苯并(a)芘[b(a)P]暴露与酶活性的量-效关系研究。结果表明,在接近沈抚灌区实地污染状况b(a)P(0.1～***-1)暴露下,第1、3、7以及第14d取样时,P450和SOD有较好的响应:SOD在第1、3d显著升高,而在第7、14d时降低;P450总体表现为低浓度b(a)P诱导下降低,高浓度下升高的趋势。CAT、POD以及GST的敏感性相对较差。比照其他4个指标,蚯蚓P450的敏感性更为优异,具有较好的应用前景。指标敏感性总体表现为:P450>SOD>CAT/POD>GST。
