With the rapid rise of new media platforms such as Weibo and Tiktok,communities with science communication characteristics have progressively grown on social networks.These communities pursue essential objectives such as increased visibility and influence.For the success of the public understanding of science in China,case studies of science communication communities on social media are becoming increasingly valuable as a point of reference.The authenticity of user influence plays an important role in the analysis of the final outcome during the process of community detection.By integrating counterfactual reasoning theory into a community detection algorithm,we present a novel paradigm for eliminating influence bias in online communities.We consider the community of Public Science Day of the Chinese Academy of Sciences as a case study to demonstrate the validity of the proposed paradigm.In addition,we examine data on science communication activities,analyze the key elements of activity communication,and provide references for not only augmenting the communication impact of similar types of popular science activities but also advancing science communication in China.Our main finding is that the propagation channel for the science communication experiment exhibits multi-point scattered propagation and lacks a continuous chain in the process of propagation.

During the development of diet-induced obesity， the change of energy matebolism is closely related to the function of the circadian clock in mammals. Luteolin (LU)， one of the most common natural flavonoids riched in many edible plants， can ameliorate obesity by activating adipose tissue browning， but its effect on circadian clock in this process remains poorly understood. Here we found that dietary LU improved circadian misalignment of energy expenditure in high-fat diet (HFD)-fed wild-type (WT) mice. Moreover， dietary LU efficiently elevated UCP1 levels in adipose tissue during the dark period， which was similar to the LU-increased hepatic PER2 expressions. Hepatic PPARα/RXRα/FGF21 pathway was rhythmically elevated by dietary LU in HFD-fed WT mice， whereas the promotion was inhibited in Per2-/-mice. Meanwhile， Per2 deletion abolished the effects of dietary LU on adipose tissue browning in HFD-fed mice. Further， LU treatment directly activated PPARα/RXRα/FGF21 signaling in primary cultured hepatocytes from WT mice rather than Per2-/-mice. Taken together， the deletion of the core clock component Per2 impedes LU-induced adipose tissue browning through weakening PPARα/RXRα/FGF21 pathway in mice， providing a new insight into the interplay of energy metabolism and circadian clock for the anti-obesity activity of LU.

Data exchange between different mode channels is essential in the optical communication network with mode-division multiplexing(MDM).However,there are challenges in realizing mode exchange with low insert loss,low mode crosstalk,and high integration.Here,we designed and fabricated a mode exchange device based on multiplane light conversion(MPLC),which supports the transmission of LP01,LP11a,LP11b,and LP21 modes in the C-band and L-band.The simulated exchanged mode purities are greater than 85%.The phase masks were fabricated on a silicon substrate to facilitate the integration with optical systems,with an insert loss of less than 2.2 dB and mode crosstalk below-21 dB due primarily to machining inaccuracies and alignment errors.We carried out an optical communication experiment with 10 Gbit/s OOK and QPSK data transmission at the wavelength of 1550 nm and obtained excellent performance with the device.It paves the way for flexible data exchange as a building block in MDM optical communication networks.

Objective:AlkB homolog 5(ALKBH5)has been proven to be closely related to tumors.However,the role and molecular mechanism of ALKBH5 in neuroblastomas have rarely been reported.Methods:The potential functional single-nucleotide polymorphisms(SNPs)in ALKBH5 were identified by National Center for Biotechnology Information(NCBI)dbSNP screening and SNPinfo software.TaqMan probes were used for genotyping.A multiple logistic regression model was used to evaluate the effects of different SNP loci on the risk of neuroblastoma.The expression of ALKBH5 in neuroblastoma was evaluated by Western blotting and immunohistochemistry(IHC).Cell counting kit-8(CCK-8),plate colony formation and 5-ethynyl-2'-deoxyuridine(EdU)incorporation assays were used to evaluate cell proliferation.Wound healing and Transwell assays were used to compare cell migration and invasion.Thermodynamic modelling was performed to predict the ability of miRNAs to bind to ALKBH5 with the rs8400 G/A polymorphism.RNA sequencing,N6-methyladenosine(mA)sequencing,mA methylated RNA immunoprecipitation(MeRIP)and a luciferase assay were used to identify the targeting effect of ALKBH5 on SPP1.Results:ALKBH5 was highly expressed in neuroblastoma.Knocking down ALKBH5 inhibited the proliferation,migration and invasion of cancer cells.miR-186-3p negatively regulates the expression of ALKBH5,and this ability is affected by the rs8400 polymorphism.When the G nucleotide was mutated to A,the ability of miR-186-3p to bind to the 3'-UTR of ALKBH5 decreased,resulting in upregulation of ALKBH5.SPPI is the downstream target gene of the ALKBH5 oncogene.Knocking down SPP1 partially restored the inhibitory effect of ALKBH5 downregulation on neuroblastoma.Downregulation of ALKBH5 can improve the therapeutic efficacy of carboplatin and etoposide in neuroblastoma.Conclusions:We first found that the rs8400 G>A polymorphism in the m6A demethylase-encoding gene ALKBH5 increases neuroblastoma susceptibility and determines the related mechanisms.The aberrant regulation of ALKBH5 by miR-186-3

目的观察基于超声征象建立的风险模型预测胎盘植入性疾病(PAS)侵袭性的价值。方法回顾性分析133例PAS患者资料,根据PAS有无侵袭性将其分为侵袭组(63例)和非侵袭组(70例)。比较组间12个PAS相关超声征象及胎盘下缘与宫颈内口距离(D值)的差异,以单因素logistic回归及受试者工作特征(ROC)曲线分析选定判断PAS侵袭性超声征象的数目阈值,并建立其二分类变量,通过多因素logistic回归确定其与D值是否为预测PAS侵袭性的独立影响因素,并由此建立预测模型。结果12个相关征象中,胎盘后低回声带中断或消失、子宫肌层变薄、异常胎盘陷窝、胎盘增厚、宫颈受累、膀胱壁中断或消失、胎盘陷窝供应血管、桥血管、胎盘基底部血流增多及子宫膀胱间血管增多在侵袭组中的检出率均高于非侵袭组(P均0.05)。侵袭组多见前壁为主的前置胎盘及多超声征象;多超声征象及D值过小是预测PAS侵袭性的独立危险因素(P均<0.05),所获预测模型为logit(P)=-0.717+1.551×多超声征象-0.216×D值,其曲线下面积(AUC)为0.905。结论PAS多超声征象及胎盘下缘与宫颈内口距离过短均为预测PAS侵袭性的独立危险因素;所获模型用于预测PAS侵袭性风险效能较佳。

Acetaminophen(APAP),the most frequently used mild analgesic and antipyretic drug worldwide,is implicated in causing 46%of all acute liver failures in the USA and between 40%and 70%in Europe.The predominant pharmacological intervention approved for mitigating such overdose is the antioxidant N-acetylcysteine(NAC);however,its efficacy is limited in cases of advanced liver injury or when administered at a late stage.In the current study,we discovered that treatment with a moderate intensity static magnetic field(SMF)notably reduced the mortality rate in mice subjected to high-dose APAP from 40%to 0%,proving effective at both the initial liver injury stage and the subsequent recovery stage.During the early phase of liver injury,SMF markedly reduced APAPinduced oxidative stress,free radicals,and liver damage,resulting in a reduction in multiple oxidative stress markers and an increase in the antioxidant glutathione(GSH).During the later stage of liver recovery,application of vertically downward SMF increased DNA synthesis and hepatocyte proliferation.Moreover,the combination of NAC and SMF significantly mitigated liver damage induced by high-dose APAP and increased liver recovery,even 24 h post overdose,when the effectiveness of NAC alone substantially declines.Overall,this study provides a noninvasive non-pharmaceutical tool that offers dual benefits in the injury and repair stages following APAP overdose.Of note,this tool can work as an alternative to or in combination with NAC to prevent or minimize liver damage induced by APAP,and potentially other toxic overdoses.

[Objectives]To discover novel compounds with significant anti-inflammatory activity in the alcohol extract of Polygonum capitatum.[Methods]Firstly,a new flavonoid glycoside,capitaone B(1),was isolated from the ethyl acetate extract from P.capitatum,and then 27 compounds were screened using NO anti-inflammatory activity model.[Results]Four compounds,such as kaempferol(12),1,2,6-trigalloyl-β-d-glucose(15),catechin(16)andβ-sitosterol(26),could significantly inhibit LPS-induced NO production in macrophages,with IC 50 values of 15.31,8.43,6.92 and 5.72μM,respectively.[Conclusions]The chemical composition and anti-inflammatory activity of P.capitatum were preliminarily studied,and the results provide a theoretical basis for further research on the action mechanism of subsequent anti-inflammatory active compounds of P.capitatum.

To explore the method of evaluating the soothing effect of human skin damage,a human skin damage model was established using UV light induction.Four test areas were set up,namely blank control area,UV damage prevention and soothing area,immediate soothing area after UV damage and soothing area after UV damage.Five skin parameters,including skin melanin,red pigment value,skin pigmentation value,a*value,and skin redness value,were used to characterize skin pigmentation before and after using the sample Changes in properties such as skin erythema and skin pigment.The results showed that the method showed significant changes in the skin condition of volunteers before and after using the sample,and could achieve a soothing effect,which has certain reference significance.

东北典型黑土区面临生态系统服务功能退化、景观格局破碎以及农业与生态空间冲突等问题,科学制定具有指导性的黑土区生态保护红线划定与优化框架,对于保障粮食安全与生态安全具有重要意义。基于生态安全格局视角明晰生态保护红线内涵,在考虑生态保护重要性、农业与生态空间协调、自然与社会系统协调的基础上,提出“点—线—面”多层次空间要素的系统性生态保护红线划定思路,考虑生态系统完整性与连通性识别生态保护红线优化区域。结果表明:(1)在保障不划入优质耕地的前提下(阈值为10%),叠加生态系统服务极重要区、生态环境极敏感区、生态系统服务供需不匹配区域确定生态保护红线初步划定区域69646.00 km^(2),并提取生态源地34760.16 km^(2);(2)基于生态源地与构建的综合生态阻力面识别生态廊道10980.90 km与生态节点77个,其缓冲区与生态保护红线初步划定区域共同构成生态保护红线区域76234.54 km^(2);(3)基于破碎度(0.20)和连通度(80)优化阈值,25.48%的生态保护红线区域亟需优化。研究结果提高了生态保护红线划定与优化的科学性和系统性,为东北典型黑土区国土空间精准管控与生态系统可续性管理提供重要依据,且对土地高度集约化利用、生态空间稀缺的中小尺度区域具有重要指导意义。
