Hepatitis B Virus Surface Antigen Promotes Stemness of Hepatocellular Carcinoma through Regulating MicroRNA-203a

Background and Aims:Patients with persistent positive hepatitis B surface antigen(HBsAg),even with a low HBVDNA load,have a higher risk of hepatocellular carcinoma(HCC)than those without HBV infection.Given that tumor stemness has a critical role in the occurrence and maintenance of neoplasms,this study aimed to explore whether HBsAg affects biological function and stemness of HCC by regulating microRNA,and to explore underlying mechanisms.Methods:We screened out miR-203a,the most significant down-regulated microRNA in the microarray analysis of HBsAg-positive samples and focused on that miRNA in the ensuing study.In vitro and in vivo functional experiments were performed to assess its regulatory function.The effect of miR-203a on stemness and the possible correlation with BMI1 were analyzed in this study.Results:MiR-203a was significantly down-regulated in HBsAg-positive HCC with the sharpest decrease shown in microarray analysis.The negative correlation between miR-203a and HBsAg expression was confirmed by quantitative real-time PCR after stimulation or overexpression/knockdown of HBsAg in cells.We demonstrated the function of miR-203a in inhibiting HCC cell proliferation,migration,clonogenic capacity,and tumor development in vivo.Furthermore,the overexpression of miR-203a remarkably increases the sensitivity of tumor cells to 5-FU treatment and decreases the proportion of HCC cells with stem markers.In concordance with our study,the survival analysis of both The Cancer Genome Atlas database and samples in our center indicated a worse prognosis in patients with low level of miR-203a.We also found that BMI1,a gene maintains the self-renewal capacity of stem cells,showed a significant negative correlation with miR-203a in HCC specimen(p<0.001).Similarly,opposite BMI1 changes after overexpression/knockdown of miR203a were also confirmed in vitro.Dual luciferase reporting assay suggested that miR-203a may regulate BMI1 expression by direct binding.Conclusions:HBsAg may promote the development of HCC and tumor stem

Evolution of MnO–SiO2–Al_(2)O_(3)–MgO inclusions during heat treatment at 1100℃

To investigate the evolutionary behavior of the MnO–SiO_(2)–Al_(2)O_(3)–MgO inclusions during heat treatment,water quenched samples were isothermally held at 1100°C for 120 min in Ar and air atmosphere,and the obtaining samples were analyzed by X-ray diffraction,scanning electron microscopy and energy dispersive spectrometer.It showed that 3MnO·Al_(2)O_(3)·3SiO_(2)and MnO·SiO_(2)were detected in the 5 wt.%MgO system after isothermal holding in Ar atmosphere,while MgO·Al_(2)O_(3),MnO·SiO_(2)and Mn7O8·SiO4 were detected in air atmosphere.The evolutionary behavior of the 10,15 and 20 wt.%MgO systems after isothermal holding in different atmosphere were consistent.Oxygen affected the solid phase transformation of the low MgO content systems.The calculation results of FactSage 8.1 showed that MgO·Al_(2)O_(3)was formed in the 5 wt.%MgO system with air atmosphere.The solid phase transformation was accompanied by grain coarsening during the isothermal holding process.The differences in the solid phase transformation in different atmosphere of the 5 wt.%MgO system indicated that it was a gas-phase transport grain coarsening mechanism.The enrichment of Al element in the liquid phase region at the grain edges,the homogeneous distribution of Mg element and the disappearance of the liquid phase within the crystal revealed that other MgO content systems were liquid–solid transport grain coarsening mechanism.

P-type Mg_(3)Sb_(2)-based Zintls have attracted considerable interest in the thermoelectric(TE)field due to their environmental friendliness and low cost.However,compared to their n-type counterparts,they show relatively low TE performance,limiting their application in TE devices.In this work,we simultaneously introduce Bi alloying at Sb sites and Ag doping at Mg sites into the Mg_(3)Sb_(2)to coopera-tively optimize the electrical and thermal properties for the first time,acquiring the highest ZT value of∼0.85 at 723 K and a high average ZT of 0.39 in the temperature range of 323-723 K in sample Mg_(2.94)Ag_(0.06)Sb_(1.9)Bi_(0.1).The first-principle calculations show that the codoping of Ag and Bi can shift the Fermi level into the valence band and narrow the band gap,resulting in the increased carrier concentration from 3.50×10^(17)cm^(-3)in the reference Mg 3 Sb 0.9 Bi 0.1 to∼7.88×10^(19)cm^(-3)in sample Mg 2.94 Ag 0.06 Sb 0.9 Bi 0.1.As a result,a remarkable power factor of∼778.9μW m^(-1)K^(-2)at 723 K is achieved in sample Mg 2.94 Ag 0.06 Sb 0.9 Bi 0.1.Meanwhile,a low lattice thermal conductivity of∼0.48 W m^(-1)K^(-1)at 723 K is also obtained with the help of phonon scattering at the distorted lattice,point defects,and nano-precipitates in sample Mg 2.94 Ag 0.06 Sb 0.9 Bi 0.1.The synergistic effect of using the multi-element co-doping/-alloying to optimize electrical properties in Mg_(3)Sb_(2)holds promise for further improving the TE performance of Zintl phase materials or even others.

Oxygen evolution reaction（OER） catalysts are the key core materials that determine the performance of fuel cells,metal-air batteries,electrolytic water decomposition,and other applications.In this work,a green lignin-based non-precious metal OER catalyst was prepared by a simple strategy.Firstly,c arboxylated lignin was used to complex Ni and Co in situ,and then they were placed with sodium hypophosphite in the same tube furnace for upstream and downstream high-temperature calcination to construct a lignin carbon-based Ni-Co bimetallic OER catalyst（NiCoP@C）.The synthesized catalyst is a porous bimetallic phosphide with a three-dimensional network structure and high-density electrochemical active sites.NiCoP@C exhibited favorable catalytic activity for the oxygen evolution reaction（OER） with overpotential of 280 mV at 10 mA·cm-2and a Tafel slope of 77 mV·dec-1.Additionally,it exhibited remarkable durability during usage.Density functional theory（DFT） calculations revealed that by leveraging the distinctive structure of transition metal phosphide nanoparticles incorporated into a reticulated substrate,the NiCoP@C catalyst offered an increased number of active sites for OER catalysis,significantly enhancing its stability during practical applications.The present study broadens the utilization pathways of biomass to "turn waste into treasure," aligning the development concept of green sustainable development.

Toward tunable shape memory effect of NiTi alloy by grain size engineering:A phase field study

The inelastic deformations of shape memory alloys(SMAs)always show poor controllability due to the avalanche-like martensite transformation,and the effective control for the deformation of precision de-vices has been not yet mature.In this work,the phase field method was used to investigate the shape memory effects(SMEs)of NiTi SMAs undergoing grain size(GS)engineering,to obtain tunable one-way and stress-assisted two-way SMEs(OWSME and SATWSME).The OWSME and SATWSME of the systems with various gradient-nanograin structures and bimodal grain structure,as well as that with geometric gradients were simulated.The simulated results indicate that due to the GS dependences of martensite transformation and reorientation,the occurrence and expansion of martensite reorientation,martensite transformation and its reverse can be efficaciously controlled via the GS engineering.When combining the GS engineering and geometric gradient design,since the effects of GS and stress gradient can be su-perimposed or competing,and the responses of martensite reorientation,martensite transformation and its reverse to this are different,the OWSME and SATWSME of the geometrically graded systems with various nanograin structures can exhibit different improvements in controllability.In short,the reorienta-tion hardening modulus during OWSME is increased and the transformation temperature window during SATWSME is widened by GS engineering,indicating the improved controllability of SMEs.The optimal GS engineering schemes revealed in this work provide the basic reference and guidance for designing tun-able SMEs and producing NiTi-based driving devices catering to desired functional performance in various engineering fields.

Study on the preservation effects of the amputated forelimb by machine perfusion at physiological temperature

Purpose:Ischemia and hypoxia are the main factors limiting limb replantation and transplantation.Static cold storage(SCS),a common preservation method for tissues and organs,can only prolong limb ischemia time to 4-6 h.The normothermic machine perfusion(NMP)is a promising method for the preservation of tissues and organs,which can extend the preservation time in vitro by providing continuous oxygen and nutrients.This study aimed to evaluate the difference in the efficacy of the 2 limb preservation methods.Methods:The 6 forelimbs from beagle dogs were divided into 2 groups.In the SCS group(n=3),the limbs were preserved in a sterile refrigerator at 4°C for 24 h,and in the NMP group(n=3),the perfusate prepared with autologous blood was used for the oxygenated machine perfusion at physiological temperature for 24 h,and the solution was changed every 6 h.The effects of limb storage were evaluated by weight gain,perfusate biochemical analysis,enzyme-linked immunosorbent assay,and histological analysis.All statistical analyses and graphs were performed using GraphPad Prism 9.0 one-way or two-way analysis of variance.The p value of less than 0.05 was considered to indicate statistical significance.Results:In the NMP group,the weight gained percentage was 11.72%±4.06%;the hypoxia-inducible factor-1αcontents showed no significant changes;the shape of muscle fibers was normal;the gap between muscle fibers slightly increased,showing the intercellular distance of(30.19±2.83)μm;and the vascularα-smooth muscle actin(α-SMA)contents were lower than those in the normal blood vessels.The creatine kinase level in the perfusate of the NMP group increased from the beginning of perfusion,decreased after each perfusate change,and remained stable at the end of perfusion showing a peak level of 4097.6 U/L.The lactate dehydrogenase level of the NMP group increased near the end of perfusion and reached the peak level of 374.4 U/L.In the SCS group,the percentage of weight gain was 0.18%±0.10%,and the contents of hypoxia-inducible factor-1αin

A Knowledge-enhanced Two-stage Generative Framework for Medical Dialogue Information Extraction

This paper focuses on term-status pair extraction from medical dialogues(MD-TSPE),which is essential in diagnosis dia-logue systems and the automatic scribe of electronic medical records(EMRs).In the past few years,works on MD-TSPE have attracted increasing research attention,especially after the remarkable progress made by generative methods.However,these generative methods output a whole sequence consisting of term-status pairs in one stage and ignore integrating prior knowledge,which demands a deeper un-derstanding to model the relationship between terms and infer the status of each term.This paper presents a knowledge-enhanced two-stage generative framework(KTGF)to address the above challenges.Using task-specific prompts,we employ a single model to com-plete the MD-TSPE through two phases in a unified generative form:We generate all terms the first and then generate the status of each generated term.In this way,the relationship between terms can be learned more effectively from the sequence containing only terms in the first phase,and our designed knowledge-enhanced prompt in the second phase can leverage the category and status candidates of the generated term for status generation.Furthermore,our proposed special status"not mentioned"makes more terms available and en-riches the training data in the second phase,which is critical in the low-resource setting.The experiments on the Chunyu and CMDD datasets show that the proposed method achieves superior results compared to the state-of-the-art models in the full training and low-re-sourcesettings.

Effect of Texture on the Grain-Size-Dependent Functional Properties of NiTi Shape Memory Alloys and Texture Gradient Design:A Phase Field Study

Texture is inevitably introduced during the manufacturing of most NiTi shape memory alloys(SMAs),and the textured nanocrystalline NiTi has been extensively employed in engineering.However,the effect of texture,and the joint effect of grain size(GS)and texture on the functional properties of NiTi SMAs and the corresponding microscopic mechanisms have not been clearly understood yet.In this work,based on the phase field method,the effect of texture on the GS-dependent functional properties of NiTi SMAs,including super-elasticity(SE),one-way shape memory effect(OWSME),and stress-assisted two-way shape memory effect(SATWSME),is investigated,and the corresponding microscopic mechanisms are revealed.Moreover,the samples with discrete geometrical gradients and/or texture gradients are designed to achieve graded functional properties.The simulation results indicate that the dependence of functional properties on texture is due to the effect of crystallographic orientation on martensite transformation and reorientation,which can lead to different inelastic strains.In the designed samples with texture gradients,the stress–strain responses of sheets with various textures are different,allowing for the coordination of overall deformation of the sample by combining such sheets,with varying inelastic deformation degrees.Thus,the overall response of the sample differs from that without texture gradient,leading to the achievement of graded functional properties.The simulation results and new findings in this work contribute to a deeper understanding of the effects of texture,GS,and their interaction on the functional properties of SMAs,and provide valuable reference for the design and development of SMA-based devices with desired functional properties.

Clinicopathological and Prognostic Value of Programmed Cell Death 1 Expression in Hepatitis B Virus-related Hepatocellular Carcinoma:A Meta-analysis

Background and Aims:The efficacy of targeted programmed cell death 1/programmed death ligand 1(PD-1/PD-L1)monoclonal antibodies(mAbs)has been confirmed in many solid malignant tumors.The overexpression of PD-1/PD-L1 serves as a biomarker to predict prognosis and clinical progression.However,the role of PD-1 in patients with hepatitis B virus-related hepatocellular carcinoma(HBV-HCC)remains indeterminate.Given that HBV is the most important cause for HCC,this study aimed to investigate the prognostic and clinicopathological value of PD-1 in HBV-HCC via a meta-analysis.Methods:We searched PubMed,Embase,Scopus,the Cochrane Library,Web of Science and Google Scholar up to January 2021 for studies on the correlation between clinicopathology/prognosis and PD-1 in patients with HBV-HCC.The pooled hazard ratios(HRs)and 95%confidence intervals(CIs)were calculated to investigate the prognostic significance of PD-1 expression.The odds ratios(ORs)and 95%CIs were determined to explore the association between PD-1 expression and clinico-pathological features.Results:Our analysis included seven studies with 658 patients,which showed that high PD-1 expression was statistically correlated with poorer overall survival(HR=2.188,95%CI:[1.262-3.115],p<0.001)and disease-free survival(HR=2.743,95%CI:[1.980-3.506],p<0.001).PD-1 overexpression was correlated with multi-ple tumors(OR=2.268,95%CI:[1.209-4.257],p=0.011),high level of alpha fetoprotein(AFP;OR=1.495,95%CI:[1.005-2.223],p=0.047)and advanced Barcelona Clinic Liver Cancer(BCLC)stage(OR=3.738,95%CI:[2.101-6.651],p<0.001).Conclusions:Our meta-analysis revealed that the high level of PD-1 expression was associated with multiple tumors,high level of AFP and advanced BCLC stage.It significantly predicted a poor prognosis of HBV-HCC,which suggests that anti-PD-1 therapy for HBV-HCC patients is plausible.
