Cholesterol is a critical lipid for all mammalian cells,ensuring proper membrane integrity,fluidity,and biochemical *** evidence indicates that macrophages rapidly and profoundly reprogram their cholesterol metabolism in response to activation signals to support host defense ***,our understanding of the molecular details underlying how and why cholesterol homeostasis is specifically reshaped during immune responses remains less well *** review discusses our current knowledge of cellular cholesterol homeostatic machinery and introduces emerging concepts regarding how plasma membrane cholesterol is partitioned into distinct *** then discuss how proinflammatory signals can markedly reshape the cholesterol metabolism of macrophages,with a focus on the differences between MyD88-dependent pattern recognition receptors and the interferon signaling *** also discuss recent work investigating the capacity of these proinflammatory signals to selectively reshape plasma membrane cholesterol *** examine how these changes in plasma membrane cholesterol metabolism influence sensitivity to a set of microbial pore-forming toxins known as cholesterol-dependent cytolysins that specifically target cholesterol for their effector *** also discuss whether lipid metabolic reprogramming can be leveraged for therapy to mitigate tissue damage mediated by cholesterol-dependent cytolysins in necrotizing fasciitis and other related *** expect that advancing our understanding of the crosstalk between metabolism and innate immunity will help explain how inflammation underlies metabolic diseases and highlight pathways that could be targeted to normalize metabolic homeostasis in disease states.

Amyotrophic lateral sclerosis(ALS)is a fatal neurodegenerative disease that causes paralysis and respiratory failure(Petrov et al.,2017).The driving mechanisms are unknown,and there are no effective treatments(Petrov et al.,2017).Aging and a few gene mutations,a common one being missense mutations in superoxide dismutase-1(SOD1),are risk factors for ALS(Figure 1).The recent Food and Drug Administration approval of edaravone for the treatment of ALS putatively supports a role for oxidative and nitrative stresses in the disease processes(Figure 1A).DNA damage,abnormalities in DNA repair,and other nuclear abnormalities are implicated also in the pathogenesis of human ALS(Bradley and Krasin,1982;Kim et al.,2020).

Somatic variants and how to find them:One major challenge in studying somatic mosaicism is the low abundance of potentially pathogenic somatic variant(s).Unlike inherited germline variants,which are essentially present in all cells,pathogenic somatic variants only exist in a subset of cells and may be present at very low (<1%)variant allele frequency (VAF).Despite the low VAF,these somatic variants are disease-relevant and considered *** the context of focal brain malformations,which constitute a frequent cause of drug-resistant focal epilepsy,a pathogenic somatic mammalian ta rget of rapamycin (MTOR)variant with a VAF of 0.3%has been described(Baldassari et al.,2019).The identification of pathogenic somatic variants has importa nt implications both clinically and biologically.

As the size of satellites scales down, low-power and compact propulsion systems such as the pulsed plasma thruster(PPT) are needed for stabilizing these miniature satellites in orbit. Most PPT systems are operated at 2 j or more of discharge energy. In this work, the performance of a PPT with a side-fed, tongue-flared electrode configuration operated within a lower discharge energy range of 0.5-2.5 j has been investigated. Ablation and charring of the polytetrafluoroethylene propellant surface were analyzed through field-effect scanning electron microscopy imaging and energy-dispersive X-ray spectroscopy. When the discharge energy fell below 2 j, inconsistencies occurred in the specific impulse and the thrust efficiency due to the measurement of the low mass bit. At energy ≥2 j, the performance parameters are compared with other PPT systems of similar configuration and discussed in depth.
