Evidence for colorectal sarcomatoid carcinoma arising from tubulovillous adenoma

Sarcomatoid carcinomas of the colorectum are rare tu- mors that display both malignant epithelial and stromal components. Clinically, they are aggressive tumors with early metastasis. Due to their infrequent occurrence, the pathogenesis is poorly understood. We report a case of a 52-year-old woman who presented with a rectal mass and intermittent hematochezia. Superficial biopsies during colonoscopy revealed a tubulovillous adenoma with high-grade dysplasia. Endoscopic ultra- sonography confirmed an invasive nature of the mass, and deeper biopsies revealed the presence of neoplasm with mixed histological components. The surgically- excised specimen demonstrated the presence of poorly differentiated spindle cells underneath the tubulovillous adenoma and an intermediate stage of invasive adeno- carcinoma. Based on the histological appearance and immunohistochemical studies, a diagnosis of sarcoma- toid carcinoma was made. Only nine cases of sarcoma- toid carcinomas of the colorectum have been reported to date. As a result, the terminology and pathogenesis of sarcomatoid carcinoma remain speculative. To the best of our knowledge, this is the first report of co- existence of sarcomatoid carcinoma and invasive ad- enocarcinoma with tubulovillous adenoma; all stages represented within the same tumor. This observation supports the "monoclonal theory" of pathogenesis with an adenoma-sarcoma progression with or without an intermediate stage of carcinoma.

Factors associated with seeking western or Chinese medical treatment for fertility among women with breast cancer in Taiwan

OBJECTIVE: To examine and describe the use of Western and Chinese Medicine for pregnancy preparation among women with breast cancer after cancer-related treatment in ***: Women of reproductive age(20-49years) women who were treated for breast cancer from January 2011 through June 2015 in a Taiwan Residents city participated in the study. Of 306 eligible study participants, interviews were completed in197(64%). Participants were asked about sociodemographic variables, disease and treatment characteristics, their desire for fertility, and their use of Western or Chinese ***: The proportions of women who planned to use Western or Chinese Medicine to help achieve pregnancy were 17.3% and 14.7%, respectively. The result of binary logistic regression showed that current employment and lack of children were factors predictive of the use of Western Medicine. Younger age and the use of Chinese Medicine before treatment were predictors of using Chinese Medicine. While the desire for fertility preservation was related to the use of Western Medicine,the desire for pregnancy was related to the use of Chinese ***: Various characteristics guided the women's decisions to use Western or Chinese Medicine. Health care providers should learn patients' preferences for fertility preservation and provide appropriate advice, referring them to safe providers of their desired method of fertility treatment.

Development of an industrial Internet of things suite for smart factory towards re-industrialization

Re-industrialization, which supports industrial upgrading and transformation, promotes smart production and high value-added manufacturing processes, and helps to create new momentum for the economic. Under the current situation, industrialists encounter several chal- lenges to achieve re-industrialization. Firstly, the cost and technical thresholds for industrialists to leverage emerging technologies are high. Secondly, there are huge quantities and numerous types of Internet of things （IoT） devices in smart factories, warehouses and offices. The enormous extents of data exchange and communication, manage- ment, monitoring and control of IoT devices as well as the establishment and maintenance of a reliable cloud platform hinder industrialists to implement an integrated smart production management. Therefore, to achieve re-indus- trialization, an industrial Internet of things （IIoT） suite consisting of a micro-services-based IIoT cloud platform and IIoT-based smart hub is proposed, which helps to materialize re-industrialization and to conduct industrial upgrading and transformation to achieve smart production and high value-added manufacturing processes.

Numerical Study on the Suppression of Shock Induced Separation on the Non-Adiabatic Wall

A numerical model is constructed to simulate the interaction of supersonic (M = 2.4 ) oblique shock wave / turbulent boundary layer on a strongly heated wall. The heated wall temperature is two times higher than the adiabatic wall temperature and the shock wave is strong enough to induce boundary layer separation. The turbulence model is Spalart-Allmaras model. The comparison of the wall pressure distribution with the experimental data ensures the validity of this numerical model. The effect of strong wall heating enlarges the separation region upstream and downstream. In order to eliminate the separation, wall bleeding is applied at the shock foot position. As a result of the parametric study, the best position of the bleeding slot is selected. The position of the bleeding is very important for the separation suppression. If the bleeding is applied upstream of shock foot, then separation reoccurs after the bleeding slot. If the bleeding is applied downstream of shock foot, the upstream boundary layer is little influenced and still separated. The bleeding vent width is about same as the upstream boundary layer thickness and suction mass flow is 20 to 80 % of the flow rate in the upstream boundary layer. The bleeding mass flow rate is very sensitive to the bleeding vent position if we fix the vent outlet pressure. The final configuration of the shock reflection pattern approaches to the non-viscous value when wall bleeding is applied at the shock impinging point.

Disruption of Arabidopsis CHY1 Reveals an Important Role of Metabolic Status in Plant Cold Stress Signaling

To study cold signaling, we screened for Arabidopsis mutants with altered cold-induced transcription of a firefly luciferase reporter gene driven by the CBF3 promoter （CBF3-LUC）. One mutant, chy1-10, displayed reduced cold-induction of CBF3-LUC luminescence. RNA gel blot analysis revealed that expression of endogenous CBFs also was reduced in the chyl mutant, chy1-10 mutant plants are more sensitive to freezing treatment than wild-type after cold acclimation. Both the wild-type and chyl mutant plants are sensitive to darkness-induced starvation at warm temperatures, although chyl plants are slightly more sensitive. This dark-sensitivity is suppressed by cold temperature in the wildtype but not in chyl. Constitutive CBF3 expression partially rescues the sensitivity of chyl-10 plants to dark treatment in the cold. The chyl mutant accumulates higher levels of reactive oxygen species, and application of hydrogen peroxide can reduce cold-induction of CBF3-LUC in wild-type. Map-based cloning of the gene defective in the mutant revealed a nonsense mutation in CHY1, which encodes a peroxisomal β-hydroxyisobutyryl （HIBYL）-CoA hydrolase needed for valine catabolism and fatty acid β-oxidation. Our results suggest a role for peroxisomal metabolism in cold stress signaling, and plant tolerance to cold stress and darkness-induced starvation.

Despite numerous efforts to overcome neuropathic pain,various pharmacological drugs often fail to meet the needs and have many side *** is an aluminosilicate mineral that has been reported to have an anti-inflammatory effect,but the efficacy of muscovite for neuropathic pain has not been ***,we assessed whether muscovite nanoparticles can reduce the symptoms of pain by controlling the inflammatory process observed in neuropathic *** analgesic effects of muscovite nanoparticles were explored using partial sciatic nerve ligation model of neuropathic pain,in which one-third to onehalf of the nerve trifurcation of the sciatic nerve was tightly tied to the dorsal *** nanoparticles(4 mg/100μL)was given intramuscularly to evaluate its effects on neuropathic pain(3 days per week for 4 weeks).The results showed that the muscovite nanoparticle injections significantly alleviated partial sciatic nerve ligation-induced mechanical and cold *** the spinal cord,the muscovite nanoparticle injections exhibited inhibitory effects on astrocyte and microglia activation and reduced the expression of pro-inflammatory cytokines,such as interleukin-1β,tumor necrosis factor-α,interleiukin-6 and monocyte chemoattractant protein-1,which were upregulated in the partial sciatic nerve ligation ***,the muscovite nanoparticle injections resulted in a decrease in activating transcription factor 3,a neuronal injury marker,in the sciatic *** results suggest that the analgesic effects of muscovite nanoparticle on partial sciatic nerve ligation-induced neuropathic pain may result from inhibiting activation of astrocytes and microglia as well as pro-inflammatory *** propose that muscovite nanoparticle is a potential anti-nociceptive candidate for neuropathic *** experimental protocols in this study were approved by the Institutional Animal Ethics Committee(IACUC)at Dongguk University,South Korea(approval No.2017-022-1)

The effect of sunblock against oxidative stress in farmers：a pilot study

Farmers are frequently exposed to ultraviolet（UV） radiation which causes various diseases by inducing oxidative *** study aimed to assess the effects of sunblock on oxidative stress in the ***-seven farmers were divided into two groups：those who wore sunblock for five days and those who did *** total antioxidant capacity（TAC） in urine,which is an antioxidant indicator,and 8-hydroxy-2-deoxyguanosine（8-OHdG） levels in urine,an oxidative stress indicator,were *** urinary TAC of sunblock users was significantly higher than that of non-users,but urinary 8-OHdG levels were not significantly *** after adjustment for potential confounders,urinary TAC was found to be markedly increased with sunblock *** results suggest that sunblock is effective in preventing oxidative stress among *** addition,they show that urinary TAC can be used as a good effect marker of oxidative stress caused by UV exposure.

Improved method for synthesis of low molecular weight protamine–siRNA conjugate

RNAi technology has aroused wide public interest due to its high efficiency and specificity to treat multiple types of diseases. However, the effective delivery of siRNA remains a challenge due to its large molecular weight and strong anionic charge. Considering their remarkable functions in vivo and features that are often desired in drug delivery carriers, biomimetic systems for siRNA delivery become an effective and promising strategy. Based on this, covalent attachment of synthetic cell penetrating peptides(CPP) to siRNA has become of great interest. We developed a monomeric covalent conjugate of low molecular weight protamine(LMWP, a well-established CPP) and siRNA via a cytosol-cleavable disulfide linkage using PEG as a crosslinker. Results showed that the conjugates didn't generate coagulation, and exhibited much better RNAi potency and intracellular delivery compared with the conventional charge-complexed CPP/siRNA aggregates. Three different synthetic and purification methods were compared in order to optimize synthesis efficiency and product yield. The methodology using hetero-bifunctional NHS–PEG–OPSS as a crosslinker to synthesize LMWP–siRNA simplified the synthesis and purification process and produced the highest yield. These results pave the way towards siRNA biomimetic delivery and future clinical translation.
