Deletion of IFT20 in early stage T lymphocyte differentiation inhibits the development of col agen-induced arthritis

IFT20 is the smallest member of the intraflagellar transport protein（IFT） complex B. It is involved in cilia formation. Studies of IFT20 have been confined to ciliated cells. Recently, IFT20 was found to be also expressed in non-ciliated T cells and have functions in immune synapse formation and signaling in vitro. However, how IFT20 regulates T-cell development and activation in vivo is still unknown. We deleted the IFT20 gene in early and later stages of T-cell development by crossing IFT20flox/flox（IFT20f/f） mice with Lck-Cre and CD4-Cre transgenic mice, and investigated the role of IFT20 in T-cell maturation and in the development of T cell-mediated collagen-induced arthritis（CIA）. We found that both Lck-Cre/IFT20f/f and CD4-Cre/IFT20f/f mice were indistinguishable from their wild-type littermates in body size, as well as in the morphology and weight of the spleen and thymus. However, the number of CD4- and CD8-positive cells was significantly lower in thymus and spleen in Lck-Cre/IFT20f/f mice. Meanwhile, the incidence and severity of CIA symptoms were significantly decreased, and inflammation in the paw was significantly inhibited in Lck-Cre/IFT20f/f mice compared to Lck-Cre/IFT201/1littermates. Deletion IFT20 in more mature T cells of CD4-Cre/IFT20f/f mice had only mild effects on the development of T cells and CIA. The expression of IL-1b, IL-6 and TGF-b1 were significantly downregulated in the paw of Lck-Cre/IFT20f/f mice, but just slight decreased in CD4-Cre/IFT20f/f mice. These results demonstrate that deletion of IFT20 in the early stage of T-cell development inhibited CIA development through regulating T-cell development and the expression of critical cytokines.

The current study attempts to compare the hybrid artificial intelligence models to forecast the environmental externalities in Saudi *** have used the denoising based artificial intelligence models to construct hybrid *** comparing the denoising techniques,the CSD-based denoising has ***,we have used the CSD-based hybrid ***-ann and CSD-RNN are used for denoising-based artificial intelligence models,whereas CSD-ARIMA is used for denoising-based traditional *** these models are used to check and compare their performance in terms of level and direction of prediction for PM_(10).The results show that the CSD-based ann model has a higher predictability for PM_(10) levels in Saudi Arabia due to low error values and higher Dstat *** comparing original and forecasted data,the superiority of CSD-ann is evident in predicting the PM_(10) in Saudi ***,this hybrid model can predict the environmental externalities for non-linear and highly noised ***,the findings can be useful in achieving the sustainable development goal.

A Comprehensive 3-Steps Methodology for Vibration-Based Fault Detection,Diagnosis and Localization in Rotating Machines

In any industry,it is the requirement to know whether the machine is healthy or not to operate machine *** the machine is not healthy then what is the fault in the machine and then finally its *** paper is proposing a 3-Steps methodology for the machine fault diagnosis to meet the industrial requirements to aid the maintenance *** Step-1 identifies whether machine is healthy or faulty,then Step-2 detect the type of defect and finally its location in *** method is extended further from the earlier study on the 2-Steps method for the rotor defects only to the 3-Steps methodology to both rotor and bearing *** method uses the optimised vibration parameters and a simple Artificial Neural Network(ann)-based Machine Learning(ML)model from the earlier *** model is initially developed,tested and validated on an experimental rotating rig operating at a speed above 1st critical *** proposed method and model are then further validated at 2 different operating speeds,one below 1st critical speed and other above 2nd critical *** machine dynamics are expected to be significantly different at these *** highlights the robustness of the proposed 3-Steps method.

Numerical modeling and optimization of process parameters in continuous extrusion process through response surface methodology, artificial neural network & genetic algorithm

The product of high complex profile,high strength,high productivity and excellent material properties with infinite length can be produced by Continuous Extrusion(CE)*** numerical simulation of Aluminum(AA 1100)feedstock material at different wheel velocities,product diameter,feedstock temperature,die temperature and friction condition has been carried out using 3D simulation tool Design Environment for Forming(DEFORM-3D)in this *** development of mathematical model is carried out to investigate the influence of wheel velocity,extrusion ratio,feedstock temperature,die temperature and friction conditions on total load required for the deformation and extrusion of feedstock material through Response Surface Methodology(RSM).The statistical significance of mathematical model is verified through analysis of variance(ANOVA).The most optimum value of extrusion load has been found to be 136.4 kN through iterative process of Genetic Algorithm(GA)using Artificial Neural Network(ann).The optimized value of input process variables for minimum value of extrusion load obtained has been found to be 13 Revolutions per Minute(RPM)as wheel velocity,5 mm as product diameter,0.95 as friction condition,650◦C as feedstock temperature and 550◦C as die *** paper with proposed methodology will be helpful for industries working in the area of CE in terms of minimizing energy consumption during production process of bus bars,tubes,wires,cables,sheets,plates,strips,etc.

Sum-Based Meta-Analytical Enrichment of Gene Expression Data to Identify Pathway Signatures of Cancers

A new method for analysis of microarray gene expression experiments referred to as Sum-based Meta-analytical Enrichment?(SME) is proposed in this manuscript. SME is a combined enrichment and meta-analytical approach to infer?on the association of gene sets with particular phenotypes. SME allows enrichment to be performed across datasets,?which to our knowledge was not earlier possible. As a proof of concept study, this technique is applied to datasets from?Oncomine, a publicly available cancer microarray database. The genes that are significantly up-/down-regulated?(p-value ≤ 10-4) in various cancer types in Oncomine were listed. These genes were assigned to biological processes?using GO annotations. The SME algorithm was applied to identify a list of GO processes most deregulated in 4 major?cancer types. For validation we examined whether the processes predicted by SME were already documented in *** method identified several known processes for the 4 cancer types and identified several novel processes?which are biologically plausible. Nearly all the pathways identified by SME as common to the 4 cancers were found to?contribute to processes which are widely regarded as cancer hallmarks. SME provides an intuitive yet objective ‘process-centric’ interpretation of the ‘gene-centric’ output of individual microarray comparison studies. The methods described?here should be applicable in the next-generation sequencing based gene expression analysis as well.

Predicting heat-stressed EEG spectra by self-organising feature map and learning vector quantizers——SOFM and LVQ based stress prediction

Self-Organising Feature Map (SOFM) along with learning vector quantizers (LVQ) have been designed to identify the alterations in brain electrical potentials due to exposure to high environmental heat in rats. Three groups of rats were considered—acute heat stressed, chronic heat stressed and control groups. After long EEG recordings following heat exposure, EEG data representing three different vigilance states such as slow wave sleep (SWS), rapid eye movement (REM) sleep and AWAKE were visually selected and further subdivided into 2 seconds long epoch. In order to evaluate the performance of artificial neural network (ann) in recognizing chronic and acute effects of heat stress with respect to the control subjects, unsupervised learning algorithm was applied on EEG data. Mean performance of SOFM with quadratic taper function was found to be better (chronic-92.6%, acute-93.2%) over the other two tapers. The effect of LVQ after the initial SOFM training seems explicit giving rise to considerable improvements in performance in terms of selectivity and sensitivity. The percentage increase in selectivity with uniform taper function is maximum for chronic and its control group (4.01%) and minimum for acute group (1.29%) whereas, with Gaussian it is almost identical (chronic-2.57%, acute-2.03%, control- 2.33%). Quadratic taper function gives rise to an increase of 2.41% for chronic, 1.96% for acute and 2.91% for control patterns.
