Mycosphaerella areola—The Teleomorph of Ramularia areola of Cotton in Brazil, and Its Epidemiological Significance

While Ramularia leaf blight of cotton caused by Ramularia areola is of top most importance for Brazil, information is lacking regarding the survival mechanism of this pathogen during the cotton-free period. The teleomorph of R. areola is expected to belong to the genus Mycosphaerella. In the present study attempts were made to verify occurrence of this teleomorph in the State of Mato Grosso, Brazil. Decaying cotton leaves were collected two months after harvest of 2014 from 44 commercial and experimental fields where aerial fungicidal applications were made or not during the crop cycle to control the Ramularia leaf blight. Examination of the decaying cotton leaves revealed presence of abundant sclerotia, spermagonia and ascoma of Mycosphaerella sp. intermingled with each other during the cotton-free period in most of the leaf samples. Mono-ascospore isolations were obtained from the ascoma and considering their cultural, morphological, pathological and DNA sequence analysis they were identified as Mycosphaerella areola. M. areola and R. areola isolates produced similar symptoms under glasshouse inoculations. Reisolation of the pathogen from the symptoms produced by M. areola isolates yielded R. areola. Some cotton leaves showing such symptoms were kept on the soil surface on plastic trays for two months under natural field condition. After this period the decaying leaves showed abundant perithecia identical to their original M. areola. ITS rDNA sequence analyses revealed identical sequences from M. areola and R. areola isolates. Occurrence and the viability of the perfect stage M. areola during the cotton-free period on the left-over stubble from one season to another were interpreted as the survival mechanism of the pathogen and were considered responsible for the Ramularia blight epidemics in the State of Mato Grosso. Disease management practices for the State of Mato Grosso are discussed. This is the first report about the occurrence of the M. areola in Brazil.

Copper Phytoextraction and Phytostabilization by <i>Brachiaria decumbens</i>Stapf. in Vineyard Soils and a Copper Mining Waste

Brachiaria decumbens is a high biomass plant with great potential for phytoremediation of copper-polluted soils. The current study aimed to evaluate B. decumbens plants for phytoextraction and phytostabilization use in two different copper contaminated vineyard soils and a copper mining waste. Also, the macro and micronutrients uptake were evaluated after plants growth in copper contaminated soils. B. decumbens was cultivated in two vineyard soils (Inceptisol and Mollisol) and a copper mining waste for 47 days of growth in greenhouse. Then, B. decumbens’s nutrient uptake was evaluated, and it’s potential application in phytoremediation techniques for the phytoextraction and phytostabilization of copper contamination. B. decumbens exhibited high levels of biomass production at contaminated soils and no negative effect on macronutrients uptake was found. Copper contaminated soils affected micronutrients uptake by Brachiaria plants. This Brachiaria specie showed high potential on copper phytoextraction with accumulation of copper concentrations in the shoots and roots of 70 and 585 mg·kg-1 of dry mass, respectively, in the vineyard Inceptisol soil, after 47 days of growth. Mollisol soil and copper mining waste also exhibited high copper concentration in the biomass in the entire plant with 371 and 466 mg·kg-1, respectively. Although Brachiaria exhibited low levels of translocation factor for copper, this specie showed high potential for copper phytoextraction on Inceptisol, Mollisol and copper mining waste with 1900, 1156 and 1363 g·ha-1 of copper, respectively. In summary, B. decumbens plants showed high potential for copper phytoextraction and phytostabilization of copper on contaminated vineyard soils and copper mining

Pulmonary In Situ Adenocarcinoma with Mosaic Paving Pattern

CONTEXT: Adenocarcinoma already comprises half the cases of lung cancer. Its insidious clinical evolution contributes to the fact that, in absolute numbers, lung tumor is the cancer with the highest mortality in the world. When still in situ, the adenocarcinoma is even quieter, making its typical presentation on the computerized tomography of an irregular semisolid nodule smaller than 3.0 cm. It is often diagnosed in a finding of examination in an asymptomatic patient. The prevalence of in situ adenocarcinoma (ISA) is less than 5% of pulmonary malignancies and its radiological presentation with a diffuse mosaic paving pattern is even more unusual, mimicking other conditions more frequent to this finding. CASE REPORT: We describe the case of a 44-year-old male patient with a history of chronic smoking admitted to the emergency room at a referral hospital in S?o Paulo on 12/16/2016 with a complaint of progressive dyspnea associated with dry cough for 3 months, intermittent fever and weight loss of 8 kg in 2 months. A chest X-ray and computed tomography showed discrete focal points of peribroncovascular consolidation, predominantly central, areas with frosted glass attenuation associated with smooth thickening of the interlobular septa, sometimes interspersed with areas of preserved parenchyma, giving an aspect of “crazing paving” with diffuse distribution by the pulmonary parenchyma. The patient underwent a biopsy with the anatomicopathological diagnosis of primary Adenocarcinoma in situ of the lung. CONCLUSION: We emphasize that the “crazing paving” of adenocarcinoma in situ pulmonary should be considered and known by the radiologist, because although isolated it is a rare condition, its early distrust in cases of atypical evolution of the most common injuries can avoid a diagnosis in phases more advanced and higher mortality.

Computing a Predictor Set Influence Zone through a Multi-Layer Genetic Network to Explore the Role of Estrogen in Breast Cancer

Modeling inter-relationships of genes over a specific genetic network is one of the most challenging studies in systems biology. Among the families of models proposed one commonly used is the discrete stochastic, based on conditionally independent Markov chains. In practice, this model is estimated from time sequential sampling, usually obtained by microarray experiments. In order to improve the accuracy of the estimation method, we can use biological knowledge. In this paper, we decided to apply this idea to study the role of estrogen in breast cancer proliferation. The n-influence zone of a set S of genes in a given multi-layer genetic network is a set L of genes regulated, directly or indirectly, by genes in S, after at most n-1 layers. In this manuscript we describe a new approach for computing the n-influence zone of S through the estimation of a multi-layer genetic network from gene expression time series, measured by microarrays, and biological knowledge. Using seed genes related to cell proliferation, our method was able to add to the third layer of the network other genes related to this biological function and validated in the literature. Using a set of genes directly influenced by estrogen, we could find a new role for cell adhesion genes estrogen dependent. Our pipeline is user-friendly and does not have high system requirements. We believe this paper could contribute to improve the data mining for biologists in microarray time series.

Sarcopenia and Anemia Are Predictors of Poor Prognostic in Cervical Cancer Patients

Objective: Evaluate pretreatment sarcopenia and anemia as prognostic factors in women undergoing treatment for cervical cancer (CC) with concurrent chemoradiotherapy (CCRT). Methods: 151 women with CC were analysed in this cohort study. Pretreatment computed tomography (CT) images were analysed to assess skeletal muscle index (SMI). Hazard ratios (HR) and multivariate Cox proportional HR were used to analyse association between low SMI, age, body mass index (BMI), haemoglobin levels, histological type, and International Federation of Gynaecology and Obstetrics (FIGO) stage with PFS and OS. Results: A total of 151 patients were included, 53 (35.1%) presented pretreatment sarcopenia;51 (34%) stage I/II and 100 (66%) stage III/IV. Among those patients in advanced stage (III/IV) 37 (70%) (p = 0.28) were sarcopenic at the beginning of treatment. Sarcopenia was associated with worse progression-free survival (PFS) and overall survival (OS) in our cohort [HR 0.97 (p = 0.01)] [HR 0.73 (p = 0.001)], as well as anemia [HR 0.73 (p = 0.001)] [HR 0.78 (p = 0.001)]. Linear regression models indicated that despite showing no association with age, neutrophil or platelet counts, sarcopenia was associated with pretreatment anemia levels (p = 0.01). After a multivariate analysis, only haemoglobin (anemia) and complete CCRT remained associated with PFS and OS. Sarcopenia and anemia were associated with worse PFS and OS in FIGO stage I/II. Conclusion: Pretreatment sarcopenia was significantly associated with low haemoglobin levels. Anemia and incomplete CCRT were independently associated with poor prognosis in women with CC. Pretreatment sarcopenia, as low SMI, was a predictor of poor prognostic in early stages of CC.
