Adipose stromal/stem cells in regenerative medicine:Potentials and limitations

This article presents the stem and progenitor cells from subcutaneous adipose tissue,briefly comparing them with their bone marrow counterparts,and discussing their potential for use in regenerative *** adipose tissue differs from other mesenchymal stromal/stem cells(MSCs)sources in that it contains a pre-adipocyte population that dwells in the adventitia of robust blood ***-adipocytes are present both in the stromal-vascular fraction(SVF;freshly isolated cells)and in the adherent fraction of adipose stromal/stem cells(ASCs;in vitro expanded cells),and have an active role on the chronic inflammation environment established in obesity,likely due their monocyticmacrophage lineage *** SVF and ASCs have been explored in cell therapy protocols with relative success,given their paracrine and immunomodulatory ***,the widely explored multipotentiality of ASCs has direct application in bone,cartilage and adipose tissue *** aim of this editorial is to reinforce the peculiarities of the stem and progenitor cells from subcutaneous adipose tissue,revealing the spheroids as a recently described biotechnological tool for cell therapy and tissue *** cell culture techniques,in particular 3D scaffold-free cultures such as spheroids,are now available to increase the potential for regeneration and differentiation of mesenchymal *** are being explored not only as a model for cell differentiation,but also as powerful 3D cell culture tools to maintain the stemness and expand the regenerative and differentiation capacities of mesenchymal cell lineages.

bioaerosols are airborne microorganisms that cause infectious sickness,respiratory and chronic health *** have become a latent threat,particularly in indoor *** is a promising process to inactivate completely bioaerosols from ***,in systems treating a continuous air flow,catalysts can be partially lost in the gaseous *** avoid such phenomenon,supporting materials can be used to fix *** the present work,four photocatalytic systems using Perlite or Poraver glass beads impregnated with ZnO or TiO_(2)were *** inactivation mechanism of bioaerosols and the cytotoxic effect of the catalysts to^bioaeros^ls were *** plug flow photocatalytic reactor treated a bioaerosol flow of 460×10^(6)cells/m^(3)_(air)with a residence time of 5.7 *** Cytometry(FC)was used to quantify and characterize bioaerosols in terms of dead,injured and live *** most efficient system was ZnO/Perlite with 72%inactivation of bioaerosols,maintaining such inactivation during 7.5 h due to the higher water retention capacity of Perlite(2.8 mL/gpcriite)in comparison with Poraver(1.5 mL/gperiite).However,a global balance showed that Ti0_(2)/Poraver system triggered the highest level of cytotoxicity to bioaerosols retained on the support after 96 h with 95%of dead *** and FC analyses showed that the mechanism of inactivation with ZnO was based on membrane damage,morphological cell changes and cell lysis;whereas only membrane damage and cell lysis were involved with Ti0_(2).Overall,results highlighted that photocatalytic technologies can completely inactivate bioaerosols in indoor environments.

The increasing prevalence of obesity is alarming because it is a risk factor for cardiovascular and metabolic diseases(such as type 2 diabetes). The occurrence of these comorbidities in obese patients can arise from white adipose tissue(WAT) dysfunctions, which affect metabolism, insulin sensitivity and promote local and systemic inflammation. In mammals, WAT depots at different anatomical locations(subcutaneous, preperitoneal and visceral) are highly heterogeneous in their morpho-phenotypic profiles and contribute differently to homeostasis and obesity development, depending on their ability to trigger and modulate WAT inflammation. This heterogeneity is likely due to the differential behavior of cells from each depot. Numerous studies suggest that adiposederived stem/stromal cells(ASC; referred to as adipose progenitor cells, in vivo)with depot-specific gene expression profiles and adipogenic and immunomodulatory potentials are keys for the establishment of the morphofunctional heterogeneity between WAT depots, as well as for the development of depot-specific responses to metabolic challenges. In this review, we discuss depot-specific ASC properties and how they can contribute to the pathophysiology of obesity and metabolic disorders, to provide guidance for researchers and clinicians in the development of ASC-based therapeutic approaches.

In this editorial,we comment on the paper by Muthu et al published in the recent issue of the *** editorial review focusses on the use of adipose-derived stem cells(ADSCs)in knee osteoarthritis *** discuss the differences between the stromal vascular fraction and microfragmented adipose tissue and highlight the results of clinical studies comparing both treatments and the use of hyaluronic acid,platelet-rich plasma,and bone marrow aspirate *** use of expanded ADSCs is also discussed;moreover,concerns regarding treatment with ADSCs,particularly the heterogeneity of published studies and the need to standardize protocols to explore clinical potential is explored.

The discovery that adipose tissue represents an interesting source of multipotent stem cells has led to many studies exploring the clinical potential of these cells in cell-based therapies. Recent advances in understanding the secretory capacity of adipose tissue and the role of adipokines in the development of obesity and associated disorders have added a new dimension to the study of adipose tissue biology in normal and diseased states. Subcutaneous adipose tissue forms the interface between the clinical application of regenerative medicine and the establishment of the pathological condition of obesity. These two facets of adipose tissue should be understood as potentially related phenomena. because of the functional characteristics of adipose stem cells, these cells represent a fundamental tool for understanding how these two facets are interconnected and could be important for therapeutic applications. In fact, adipose tissue stem cells have multiple functions in obesity related to adipogenic, angiogenic and secretory capacities. In addition, we have also previously described a predominance of larger blood vessels and an adipogenic memory in the subcutaneous adipose tissue after massive weight loss subsequent to bariatric surgery(ex-obese patients). Understanding the reversibility of the behavior of adipose stem cells in obeses and in weight loss is relevant to both physiological studies and the potential use of these cells in regenerative medicine.

Dimensionality reduction is often used to project time series data from multidimensional to two-dimensional space to generate visual representations of the temporal *** this context,we address the problem of multidimensional time series visualization by presenting a new method to show and handle projection errors introduced by dimensionality reduction techniques on multidimensional temporal *** visualization,subsequent time instances are rendered as dots that are connected by lines or curves to indicate the temporal ***,inevitable projection artifacts may lead to poor visualization quality and misinterpretation of the temporal *** projected data points,inaccurate variations in the distances between projected time instances,and intersections of connecting lines could lead to wrong assumptions about the original *** adapt local and global quality metrics to measure the visual quality along the projected time series,and we introduce a model to assess the projection error at intersecting *** serve as a basis for our new uncertainty visualization techniques that use different visual encodings and interactions to indicate,communicate,and work with the visualization uncertainty from projection errors and artifacts along the timeline of data points,their connections,and *** approach is agnostic to the projection method and works for linear and non-linear dimensionality reduction methods alike.
