Retinal damage in the adult zebrafish induces Müller glia reprogramming to produce neuronal progenitor cells that proliferate and differentiate into retinal neurons.Notch signaling,which is a fundamental mechanism known to drive cell-cell communication,is required to maintain Müller glia in a quiescent state in the undamaged retina,and repression of Notch signaling is necessary for Müller glia to reenter the cell cycle.The dynamic regulation of Notch signaling following retinal damage also directs proliferation and neurogenesis of the Müller glia-derived progenitor cells in a robust regeneration response.In contrast,mammalian Müller glia respond to retinal damage by entering a prolonged gliotic state that leads to additional neuronal death and permanent vision loss.Understanding the dynamic regulation of Notch signaling in the zebrafish retina may aid efforts to stimulate Müller glia reprogramming for regeneration of the diseased human retina.Recent findings identified DeltaB and Notch3 as the ligand-receptor pair that serves as the principal regulators of zebrafish Müller glia quiescence.In addition,multi-omics datasets and functional studies indicate that additional Notch receptors,ligands,and target genes regulate cell proliferation and neurogenesis during the regeneration time course.Still,our understanding of Notch signaling during retinal regeneration is limited.To fully appreciate the complex regulation of Notch signaling that is required for successful retinal regeneration,investigation of additional aspects of the pathway,such as post-translational modification of the receptors,ligand endocytosis,and interactions with other fundamental pathways is needed.here we review various modes of Notch signaling regulation in the context of the vertebrate retina to put recent research in perspective and to identify open areas of inquiry.

Brine shrimp Artemia franciscana provide food for many migrating and staging birds that spend summer and fall on Great Salt Lake,Utah,USA.Artemia produce live young and cysts(hard-walled eggs);these cysts are commercially harvested on Great Salt Lake and support a large industry in Utah.It is unclear the impact that millions of hungry birds have on the Artemia population in the lake.To help assess that,this study evaluated cyst viability(percentage of cysts that contain an embryo)and hatchability(percent of cysts that hatch)from cysts that had passed through the digestive tract of eared grebes Podiceps nigricollis and cysts obtained directly from Great Salt Lake at the same site where each grebe was collected.hatchability was significantly higher for cysts collected from the water column(19%)than from the stomach(0.3%)or intestines(3%)of eared grebes.Viability also was significantly different for cysts collected from the water column(29%),stomach(0.7%),and intestines(5%).These results indicate that eared grebes nutritionally benefit from eating cysts and that they may be an important food source for grebes in late fall after the adult population of Artemia dies off due to the water becoming too cold.Also,enough cysts survive their passage through the digestive system that grebes can vector hatchable cysts to other waterbodies.

Ketamine,a noncompetitive N-methyl-D-aspartate(NMDA)receptor antagonist,has been exclusively used as an anesthetic in medicine and has led to new insights into the pathophysiology of neuropsychiatric disorders.Clinical studies have shown that low subanesthetic doses of ketamine produce antidepressant effects for individuals with depression.however,its use as a treatment for psychiatric disorders has been limited due to its reinforcing effects and high potential for diversion and misuse.Preclinical studies have focused on understanding the molecular mechanisms underlying ketamine’s antidepressant effects,but a precise mechanism had yet to be elucidated.here we review different hypotheses for ketamine’s mechanism of action including the direct inhibition and disinhibition of NMDA receptors,aminomethylphosphonic acid receptors(AMPAR)activation,and heightened activation of monoaminergic systems.The proposed mechanisms are not mutually exclusive,and their combined influence may exert the observed structural and functional neural impairments.Long term use of ketamine induces brain structural,functional impairments,and neurodevelopmental effects in both rodents and humans.Its misuse has increased rapidly in the past 20 years and is one of the most common addictive drugs used in Asia.The proposed mechanisms of action and supporting neuroimaging data allow for the development of tools to identify‘biotypes’of ketamine use disorder(KUD)using machine learning approaches,which could inform intervention and treatment.

Encapsulated peracetic acid as a valid broad‑spectrum antimicrobial alternative,leading to beneficial microbiota compositional changes and enhanced performance in broiler chickens

Background Antimicrobial alternatives are urgently needed,including for poultry production systems.In this study,we tested the potential broad-range antimicrobial alternative peracetic acid,delivered in feed via the hydrolysis of encapsulated precursors through a 28-day study using 375 Ross 308 broiler chickens.We tested two peracetic acid concentrations,30 and 80 mg/kg on birds housed on re-used litter,and we evaluated the impact of both levels on gut microbial communities,bacterial concentration,antimicrobial resistance genes relative abundance and growth performance when compared to control birds housed on either clean or re-used litter.Results Body weight gain and feed conversion ratio improved in peracetic acid fed birds.At d 28,birds given 30 mg/kg of peracetic acid had a decreased Firmicutes and an increased Proteobacteria abundance in the jejunum,accompanied by an increase in Bacillus,Flavonifractor and Rombustia in the caeca,and a decreased abundance of tetracycline resistance genes.Chicken given 80 mg/kg of peracetic acid had greater caecal abundance of macrolides lincosamides and streptogramins resistance genes.Growth performance on clean litter was reduced compared to reused litter,which concurred with increased caecal abundance of Blautia,decreased caecal abundance of Escherichia/Shigella,Anaerostipes and Jeotgalicoccus,and greater gene abundance of vancomycin,tetracycline,and macrolides resistance genes.Conclusions Peracetic acid could be used as a safe broad-spectrum antimicrobial alternative in broilers.Encapsulated precursors were able to reduce the bacterial concentration in the jejunum whilst promoting the proliferation of probiotic genera in the caeca,especially at the low peracetic acid concentrations tested,and improve growth performance.Moreover,our findings offer further insights on potential benefits of rearing birds on re-used litter,suggesting that the latter could be associated with better performance and reduced antimicrobial resistance risk compared to clean litter rearing.

Arabidopsis sepals coordinate flower opening in the morning as ambient temperature rises;however,the underlying molecular mechanisms are poorly understood. Mutation of one heat shock proteinencoding gene, hSP70-16, impaired sepal heat stress responses (hSR), disrupting lipid metabolism,especially sepal cuticular lipids, leading to abnormal flower opening. To further explore, to what extent,lipids play roles in this process, in this study, we compared lipidomic changes in sepals of hsp70-16 andvdac3 (mutant of a voltage-dependent anion channel, VDAC3, an hSP70-16 interactor) grown underboth normal (22 C) and mild heat stress (27 C, mild hS) temperatures. Under normal temperature,neither hsp70-16 nor vdac3 sepals showed significant changes in total lipids;however, vdac3 but nothsp70-16 sepals exhibited significant reductions in the ratios of all detected 11 lipid classes, except themonogalactosyldiacylglycerols (MGDGs). Under mild hS temperature, hsp70-16 but not vdac3 sepalsshowed dramatic reduction in total lipids. In addition, vdac3 sepals exhibited a significant accumulationof plastidic lipids, especially sulfoquinovosyldiacylglycerols (SQDGs) and phosphatidylglycerols (PGs),whereas hsp70-16 sepals had a significant accumulation of triacylglycerols (TAGs) and simultaneousdramatic reductions in SQDGs and phospholipids (PLs), such as phosphatidylcholines (PCs), phosphatidylethanolamines (PEs), and phosphatidylserines (PSs). These findings revealed that the impact ofmild hS on sepal lipidome is influenced by genetic factors, and further, that hSP70-16 and VDAC3differently affect sepal lipidomic responses to mild hS. Our studies provide a lipidomic insight intofunctions of hSP and VDAC proteins in the plant’s hSR, in the context of floral development.

Classical methyl-CpG binding proteins contain the conserved DNA binding motif methyl-cytosine binding domain(MBD), which preferentially binds to methylated CpG dinucleotides. These proteins serve as transcriptional repressors,mediating gene silencing via DNA cytosine methylation. Mutations in methyl-CpG binding protein 2 (MeCP2) have beenlinked to the human mental retardation disorder Rett syndrome, suggesting an important role for methyl-CpG bindingproteins in brain development and function. This mini-review summarizes the recent advances in studying the diversefunctions of MeCP2 as a prototype for other methyl-CpG binding proteins in the development and function of thevertebrate nervous system.

A GPU-Accelerated Mixed-Precision WENO Method for Extremal Black hole and Gravitational Wave Physics Computations

We develop and use a novel mixed-precision weighted essentially non-oscillatory(WENO)method for solving the Teukolsky equation,which arises when modeling perturbations of Kerr black holes.We show that WENO methods outperform higher-order finite-difference methods,standard in the discretization of the Teukolsky equation,due to the need to add dissipation for stability purposes in the latter.In particular,as the WENO scheme uses no additional dissipation,it is well suited for scenarios requiring long-time evolution such as the study of price tails and gravitational wave emission from extreme mass ratio bina-ries.In the mixed-precision approach,the expensive computation of the WENO weights is performed in reduced floating-point precision that results in a significant speedup factor of≈3.3.In addition,we use state-of-the-art Nvidia general-purpose graphics processing units and cluster parallelism to further accelerate the WENO computations.Our optimized WENO solver can be used to quickly generate accurate results of significance in the field of black hole and gravitational wave physics.We apply our solver to study the behavior of the Aretakis charge—a conserved quantity,that if detected by a gravitational wave observatory like LIGO/Virgo would prove the existence of extremal black holes.

UDP-glucuronosyltransferase 1A9(UGT1A9) is a major phase II enzyme responsible for elimination of drugs and endogenous molecules.Clinical data have shown increased elimination of UGT1A9 substrates in pregnant women or oral contraceptive users,but the role of estrogen in the regulation of UGT1A9 expression remains unknown.In this study,we investigated the effect of 17β-estradiol(E2) on UGT1A9 expression and the role of ERα in the transcriptional regulation of UGT1A9.E2 significantly increased UGT1A9 promoter activity in hep G2 cells in the presence of ERα.UGT1A9 induction by E2 was abrogated by antiestrogen ICI182,780 in hep G2 cells that constitutively express ERα.Results from transient transfection of ERα mutants into hep G2 cells demonstrated that mutation at DNA-binding domain of ERα abrogates increased UGT1A9 promoter activity by E2.Deletion and mutation assays of UGT1A9 promoter revealed a putative ERE located within -2262/-1987 region.Examination of healthy human liver tissues revealed significantly higher UGT1A9 expression in women as compared to men.Together,these findings provide a mechanistic basis for the previous clinical reports and may shed a light on identifying sources for inter-individual variability in UGT1A9-mediated drug metabolism.
