G-Quadruplex(G4) Motifs in the Maize(Zea mays L.) Genome Are Enriched at Specific Locations in Thousands of Genes Coupled to Energy Status,Hypoxia,Low Sugar,and Nutrient Deprivation

The G-quadruplex （G4） elements comprise a class of nucleic acid structures formed by stacking of guanine base quartets in a quadruple helix. This （34 DNA can form within or across single-stranded DNA molecules and is mutually exclusive with duplex B-form DNA. The reversibility and structural diversity of G4s make them highly versatile genetic structures, as demonstrated by their roles in various functions including telomere metabolism, genome maintenance, immunoglobulin gene diversification, transcription, and translation. Sequence motifs capable of forming G4 DNA are typically located in telomere repeat DNA and other non-telomeric genomic loci. To investigate their potential roles in a large-genome model plant species, we computationaily identified 149,988 non-telomeric G4 motifs in maize （Zea mays L., B73 AGPv2）, 29% of which were in non-repetitive genomic regions. G4 motif hotspots exhibited non-random enrichment in genes at two locations on the antisense strand, one in the 5~ UTR and the other at the 5~ end of the first intron. Several genic G4 motifs were shown to adopt sequence-specific and potassium-dependent G4 DNA structures in vitro. The G4 motifs were prevalent in key regulatory genes associated with hypoxia （group VII ERFs）, oxidative stress （D j-1/GATasel）, and energy status （AMPK/ SnRK） pathways. They also showed statistical enrichment for genes in metabolic pathways that function in glycolysis, sugar degradation, inositol metabolism, and base excision repair. Collectively, the maize G4 motifs may represent conditional regulatory elements that can aid in energy status gene responses. Such a network of elements could provide a mechanistic basis for linking energy status signals to gene regulation in maize, a model genetic system and major world crop species for feed, food, and fuel.

Poly(ethylene- co -chlorotrifluoroethylene) with a 1∶1 molar ratio of ethylene and chlorotrifluoroethylene in composition, PECTFE, is a statistically alternating copolymer widely used in cable coating industry. PECTFE has been commonly characterized as a semicrystalline polymer with crystals melting at 238 ℃. Differential scanning calorimetry, thermal mechanical analysis and dynamic mechanical analysis indicated that PECTFE undergoes a reversible solid-solid transition during cooling and heating in the temperature range between 100 ℃ and 200 ℃. Based on detailed structural analysis via wide angle X-ray diffraction and electron diffraction techniques, we proposed that PECTFE possesses a hexagonal phase above 200 ℃( a =0 60 nm, c =0 49 nm) and an orthorhombic phase( a =0 99 nm, b =0 57 nm, c =0 49 nm) at room temperature.

OBjECTIVE Since cyclic AMP(cAMP) signaling cascade has been implicated in mediating behavioral responses to alcohol, key components in this cascade may serve as potential treatment *** 4(PDE4),an enzyme that specifically catalyzes the hydrolysis of cAMP,plays a critical role in regulating its intracellular ***,it was of interest to determine whether PDE4 was involved in the regulation of alcohol use and *** Male Fawn-Hooded(FH/Wjd) rats were tested for 5%(V/V) ethanol and 10%(W/V) sucrose operant oral self-administration following treatment with the selective PDE4 inhibitor rolipram(0.0125,0.025,and 0.05 mg·kg,s.c.);rolipram at higher doses(0.05,0.1,and 0.2 mg·kg,s.c.) was tested to determine its impact on the intake of ethanol,sucrose,or water using the two-botde choice drinking *** open-field testing was performed to evaluate the influence of higher doses of rolipram on locomotor *** Acute administration of rolipram dose-dependently reduced operant self-administration of 5%ethanol,but had no effect on 10%sucrose ***-course assessment revealed a significant anti-drinking effect of rolipram(0.1 and 0.2 mg·kg) in continuous-and intermittent-access to ethanol at 5%or 10%,***,chronic rolipram treatment time-dependently decreased 5%ethanol consumption and preference during treatment days and after the termination of rolipram *** at the highest doses(0.1 and 0.2 mg·kg) did decrease locomotor activity,but the effect lasted only 10 and 20 min,respectively,which did not likely alter long-term ethanol *** These results suggest that PDE4 plays a role in alcohol seeking and consumption *** interfering with PDE4 may be a potential pharmacotherapy for alcohol dependence.

Expanding the US-China cooperation on service programs： New opportunities for both countries

For years, the US and China have cooperated closely on manufacturing programs, which helps China become the world manufacturing center. While they both have gained much from the cooperation, there are also increasing frictions, disputes, complains and dissatisfaction with each other because of the huge trade unbalance problem and other significant issues. The US is eager to expand export to China, but China seems hesitating to decide what to import from the US. This paper presents an analysis about the benefits of the US-China cooperation with a primary focus on the service sector, which remains a large and untapped opportunity for China. The goal of the paper is to explore a new route to relieve the trade balance issues as they separately impact both nations. While focusing on analyzing several immediate opportunities, the paper also investigates several new ideas that rest on technology as well as entrepreneurial development.

＜正＞Alcohol dependence is a complex psychiatric disorder demanding development of novel pharmacotherapy. Since the cyclic AMP（cAMP） signaling cascade has been implicated in mediating the behavioral response to alcohol, it would be of interest to determine whether phosphodiesterase-4（PDE4）,an enzyme that selectively hydrolyzes intracellular cAMP,is involved in the regulation of alcohol ***,the selective PDE4 inhibitor rolipram was used to measure its anti-drinking properties in alcohol-preferring Fawn-Hooded（FH/Wjd） *** showed that acute administration of rolipram（0.05,0.1,and 0.2 mg/kg） selectively and dose-dependently reduced 5%ethanol, but not 10%sucrose,operant self-administration;it also decreased voluntary 5%ethanol intake,but not water consumption,for the initial 30 min in rats showing high stable preference for *** course assessment revealed the significant anti-drinking effect of rolipram（0.1,0.2 mg/kg） lasted for 5 h in both continuous- and intermittentaccess to 5%or 10%***,chronic rolipram treatment decreased 5%ethanol consumption and preference during the drug treatment *** effect lasted even after withdrawal of drug *** locomotor activity was inhibited by rolipram at 0.1 and 0.2 mg/kg,the effect was present only in the initial about 20 min,which did not seem to alter long-term ethanol *** data suggest that PDE4 may play a promising role in ethanol drinking and dependent *** may be a potential treatment target for further development of anti-drinking drugs.

In this article,we present a nanoelectromechanical system(NEMS)designed to detect changes in the Casimir *** Casimir effect is a result of the appearance of quantum fluctuations in an electromagnetic *** experiments have used nano-or microscale parallel plate capacitors to detect the Casimir force by measuring the small attractive force these fluctuations exert between the two *** this new set of experiments,we aim to directly detect the shifts in the Casimir energy in a vacuum due to the presence of the metallic parallel plates,one of which is a superconductor.A change in the Casimir energy of this configuration is predicted to shift the superconducting transition temperature(T_(c))because of the interaction between it and the superconducting condensation *** our experiment,we take a superconducting film,carefully measure its transition temperature,bring a conducting plate close to the film,create a Casimir cavity,and then measure the transition temperature *** expected shifts are smaller than the normal shifts one sees in cycling superconducting films to cryogenic temperatures,so using a NEMS resonator in situ is the only practical way to obtain accurate,reproducible *** a thin Pb film and opposing Au surface,we observe no shift in T_(c)>12µK down to a minimum spacing of~70 nm at zero applied magnetic field.
