Objective： The complexity, heterogeneity and capacity of malignant neoplastic cells and tumors for rapid change and evolution suggest that living-cell-based biological-systems approaches to cancer treatment are merited. Testing this hypothesis, the tumor marker, metabolic activity, and overall survival（Os） responses, to the use of one such system, implantable macrobeads [RENCA macrobeads（RMBs）], in phase I and IIa clinical trials in advanced,treatment-resistant metastatic colorectal cancer（m CRC） are described ***： Forty-eight m CRC patients（30 females; 18 males）, who had failed all available, approved treatments,underwent RMB implantation（8 RMB/kg body weight） up to 4 times in phase I and phase IIa open-label ***, labs [tumor and inflammation markers, lactate dehydrogenase（LDH）] and positron emission tomography-computed tomography（PET-CT） imaging to measure number/volume and metabolic activity of the tumors were performed pre-and 3-month-post-implantation to evaluate safety and initial efficacy（as defined by biological responses）. PET-CT maximum standard uptake value（sUVmax）（baseline and d 90; sUVmax ≥2.5）, LDH,and carcinoembryonic antigen（CEA） and/or cancer antigen 19-9（CA 19-9） response（baseline, d 30 and/or d 60）were assessed and compared to ***： Responses after implantation were characterized by an at least 20% decrease in CEA and/or CA 19-9 in75% of patients. Fluorodeoxyglucose（FDG）-positive lesions（phase I, 39; 2 a, 82） were detected in 37/48 evaluable patients, with 35% stable volume and stable or decreased sUV（10） plus four with necrosis; 10, increased tumor volume, sUV. LDH levels remained stable and low in Responders（R）（d 0–60, 290.4–333.9）, but increased steadily in Non-responders（NR）（d 0–60, 382.8–1,278.5）（d 60, P=0.050）. Can inhibition of telomerase increase pancreatic cancer cell's susceptibility to chemotherapeutic reagents?

Objectives: To clarify the inhibition of pancreatic can-cer cells by interference with the hTR component of thetelomerase reverse transcriptase enzymatic complexand evaluate susceptibility of antisense hTR pancreaticcancer cells to chemotherapeutic ***: A 593 bp of full length hTR cDNA was sub-cloned into a mammalian expression vector pcDNA3.1(-) in antisense orientation to construct an antisensehTR expression plasmid. The plasmids were introducedinto Pancl cells, a human pancreatic carcinoma cellline, by lipofectin, and G418-resistant stable trans-formants were expanded. Resulting stable clones werescreened for the presence of hTR insert by PCR withT7 and BGH reverse primers located on the flanks ofthe multiclonal site of pcDNA3.1 vector. Cell growth rate,hTR expression, telomerase activity, and anchorage-in-dependent growth property were analyzed. Finally, sus-ceptibility of antisense hTR cells to chemotherapeuticreagents was ***: significant downregulation of endogenous hTRwas evident in the antisense-hTR transformed cells,and telomerase activity was markedly decreased com-pared to control cells in standard TRAP assays. Fur-thermore, the proliferation and the anchorage-inde-pendent growth ability in antisense-hTR expressingcells were significantly decreased compared with thecontrol parental cells. However, no crisis or senescencephenomena was observed. Antisense hTR appears toincrease Pancl cell's susceptibility to chemotherapeuticreagent cDDP, but not to differentiation reagent DM-sO, COX2 inhibitor sulinbac, Ns-398, curcumin, andchemotherapeutic reagent adriamycin(ADM).Conclusions: These data indicate that hTR is probablya critical component of human telomerase activity andthat downregulation of the RNA component of humantelomerase is an effective target for anticancer strategyand antisense hTR can increase Pancl cell's susceptibilityto cDDP.

Improved prediction of clay soil expansion using machine learning algorithms and meta-heuristic dichotomous ensemble classifiers

soil swelling-related disaster is considered as one of the most devastating geo-hazards in modern ***,proper determination of a soil’s ability to expand is very vital for achieving a secure and safe ground for ***,this study has provided a novel and intelligent approach that enables an improved estimation of swelling by using kernelised machines(Bayesian linear regression(BLR)&bayes point machine(BPM)support vector machine(sVM)and deep-support vector machine(D-sVM));(multiple linear regressor(REG),logistic regressor(LR)and artificial neural network(ANN)),tree-based algorithms such as decision forest(RDF)&boosted trees(BDT).Also,and for the first time,meta-heuristic classifiers incorporating the techniques of voting(VE)and stacking(sE)were *** independent scenarios of explanatory features’combination that influence soil behaviour in swelling were *** results indicated BLR as possessing the highest amount of deviation from the predictor variable(the actual swell-strain).REG and BLR performed slightly better than ANN while the meta-heuristic learners(VE and sE)produced the best overall performance(greatest R2 value of 0.94 and RMsE of 0.06%exhibited by VE).CEC,plasticity index and moisture content were the features considered to have the highest level of *** binary classifiers(sVM,D-sVM and BPM)gave better accuracy(average accuracy and recall rate of 0.93 and 0.60)compared to ANN,LR and ***-driven diagnostic test indicated that the meta-heuristic models’best performance occurred when ML training was conducted using k-fold validation ***,it is recommended that the concepts developed herein be deployed during the preliminary phases of a geotechnical or geological site characterisation by using the best performing meta-heuristic models via their background coding resource.

As of October 2018,almost half of Us imports of goods from China are subject to new Us tariffs at various rates,mostly at 10%until the year-end of 2018,when the rates were scheduled to be raised to 25%.These tariff rates will prove to be prohibitive for most if not all of the Us imports from *** that Us imports from China will be reduced by half,the initial direct real impact on the Chinese economy may be estimated at a loss of 0.43%of *** indirect effects are included,the full real impact may be estimated at a maximum loss of 1.12%of Chinese *** estimated impacts are relatively small and quite *** is a possibility that the scope of the Us tariffs may be expanded to cover all Us imports from China,in which case the full negative economic impact will be doubled,but still leaving an expected rate of economic growth in excess of 4%.The Renminbi is not likely to be significantly devalued as a result of the trade ***,there are also longer-term underlying forces at work behind the China–Us trade war–the competition for economic and technological dominance and the rise of populism,isolationism,nationalism and *** is important for China–Us relations,and China’s relations with the rest of the world,in particular with the European Union,Association of southeast Asian Nations(AsEAN),japan and Russia,to be carefully managed going forward.

Associations of self-reported and device-assessed physical activity with fatigue,quality of life,and sleep quality in adults living with and beyond cancer

Prospective study of Centurion~？ versus Infiniti~？ phacoemulsification systems： surgical and visual outcomes

AIM： To evaluate surgical outcomes（sOs） and visual outcomes（VOs） in cataract surgery comparing the Centurion？ phacoemulsification system（CPs） with the Infiniti？ phacoemulsification system（IPs）.METHODs： Prospective, consecutive study in a singlesite private practice. Totally 412 patients undergoing cataract surgery with either the CPs using the 30-degree balanced？ tip（n=207） or the IPs using the 30-degree Kelman？ tip（n=205）. Intraoperative and postoperative outcomes were documented prospectively up to one month follow-up. Nuclear sclerosis（Ns） grade, cumulated dissipated energy（CDE）, preoperative corrected distance visual acuity（CDVA）, and CDVA at one month were recorded. REsULTs： CDE was 13.50% less in the whole CPs compared with the whole IPs subcohort. In eyes with Ns grade III or greater, CDE was 28.87% less with CPs（n=70） compared with IPs（n=44）（P=0.010）. surgical complications were not statistically different between the two subcohorts（P=0.083）, but in the one case of vitreous loss using the CPs, CDVA of 6/4 was achieved at one month. The mean CDVAs（VOs） at one month for Ns grade III and above cataracts were-0.17 log MAR（6/4.5） in the CPs and-0.15 log MAR（6/4.5） in the IPs subcohort respectively（P=0.033）.CONCLUsION： CDE is 28.87% less, and VOs are significantly improved, in denser cataracts in the CPs compared with the IPs. The authors recommend the CPs for cases with denser nuclei.
