Prospects for sociogenomics in avian cooperative breeding and parental care

For the last 40 years,the study of cooperative breeding(CB)in birds has proceeded primarily in the context of discovering the ecological,geographical,and behavioral drivers of *** advent of molecular tools in the early 1990s assisted in clarifying the relatedness of helpers to those helped,in some cases,confirming predictions of kin selection *** for genome-wide analysis of sequence variation,gene expression,and epigenetics promise to add new dimensions to our understanding of avian CB,primarily in the area of molecular and developmental correlates of delayed breeding and dispersal,as well as the ontogeny of achieving parental status in ***,we outline key ways in which modern-omics approaches,in particular genome sequencing,transcriptomics,and epigenetic profiling such as ATAC-seq,can be used to add a new level of ana-lysis of avian *** on recent and ongoing studies of avian social behavior and sociogenom-ics,we review how high-throughput sequencing of a focal species or clade can provide a robust foundation for downstream,context-dependent destructive and non-destructive sampling of spe-cific tissues or physiological states in the field for analysis of gene expression and epigenetics.-Omics approaches have the potential to inform not only studies of the diversification of CB over evolutionary time,but real-time analyses of behavioral interactions in the field or *** of birds represents a new branch in the network of methods used to study CB,and can help clarify ways in which the different levels of analysis of CB ultimately interact in novel and unexpected ways.

<i>In Vitro</i>Anticancer Activity of Plant-Derived Cannabidiol on Prostate Cancer Cell Lines

Cannabinoids, the active components of Cannabis sativa Linnaeus, have received renewed interest in recent years due to their diverse pharmacologic activities such as cell growth inhibition, anti-inflammatory effects and tumor regression, but their use in chemotherapy is limited by their psychotropic activity. To date, cannabinoids have been successfully used in the treatment of nausea and vomiting, two common side effects that accompany chemotherapy in cancer patients. Most non-THC plant cannabinoids e.g. cannabidiol and cannabigerol, seem to be devoid of psychotropic properties. However, the precise pathways through which these molecules produce an antitumor effect have not yet been fully characterized. We therefore investigated the antitumor and anti-inflammatory activities of cannabidiol (CBD) in human prostate cancer cell lines LNCaP, DU145, PC3, and assessed whether there is any advantage in using cannabis extracts enriched in cannabidiol and low in THC. Results obtained in a panel of prostate cancer cell lines clearly indicate that cannabidiol is a potent inhibitor of cancer cell growth, with significantly lower potency in non-cancer cells. The mRNA expression level of cannabinoid receptors CB1 and CB2, vascular endothelial growth factor (VeGF), PSA (prostate specific antigen) are significantly higher in human prostate cell lines. Treatment with Cannabis extract containing high CBD down regulates CB1, CB2, VeGF, PSA, pro-inflammatory cytokines/chemokine IL-6/IL-8. Our overall findings support the concept that cannabidiol, which lacks psychotropic activity, may possess anti-inflammatory property and down regulates both cannabinoid receptors, PSA, VeGF, IL-6 and IL-8. High CBD cannabis extracts are cytotoxic to androgen responsive LNCaP cells and may effectively inhibit spheroid formation in cancer stem cells. Participation in cooperative prey capture and the benefits gained from it are associated with individual personality

In animal societies, behavioral idiosyncrasies of the individuals often guide which tasks theyshould perform. Such personality-specific task participation can increase individual task efficiency,thereby improving group performance. While several recent studies have documented group-levelbenefits of within-group behavioral （i.e., personality） diversity, how these benefits are realized atthe individual level is unclear. Here we probe the individual-level benefits of personality-driven taskparticipation in the social spider Stegodyphus dumicola. In S. dumicola, the presence of at leastone highly bold individual catalyzes foraging behavior in shy colony members, and all group con-stituents heavily compete for prey. We assessed boldness by examining how quickly spidersresumed normal movement after a simulated predator attack. We test here whether （1） participantsin collective foraging gain more mass from prey items and （2） whether bold individuals are less re-sistant to starvation than shy spiders, which would motivate the bold individuals to forage ***, we assembled colonies of shy spiders with and without a bold individual, added one preyitem, and then tracked the mass gain of each individual spider after this single feeding event. Wefound that spiders that participated in prey capture （whether bold or shy） gained more mass thannonparticipators, and colonies containing a single bold spider gained more total mass than purelyshy colonies. We also found that bold spiders participated in more collective foraging events andwere more susceptible to starvation than shy spiders, suggesting that the aggressive foraging ofbold individuals may represent a strategy to offset starvation risk. These findings add to the bodyof evidence that animal personality can shape social organization, individual performance, andgroup success.

Allograft loss from acute Page kidney secondary to trauma after kidney transplantation

We report a rare case of allograft loss from acute Page kidney secondary to trauma that occurred 12 years after kidney transplantation. A 67-year-old Caucasian male with a past surgical history of kidney transplant presented to the emergency department at a local hospital with left lower abdominal tenderness. He recalled that his cat, which weighs 15 lbs, jumped on his abdomen 7 d prior. On physical examination, a small tender mass was noticed at the incisional site of the kidney transplant. He was producing a normal amount of urine without hematuria. His serum creatinine level was slightly elevated from his baseline. Computer tomography revealed a large subscapular hematoma around the transplant kidney. The patient was observed to have renal trauma grade Ⅱ at the hospital over a period of three days, and he was finally transferred to a transplant center after his urine output significantly decreased. Doppler ultrasound demonstrated an extensive peri-allograft hypoechoic area and abnormal waveforms with absent arterial diastolic flow and a patent renal vein. Despite surgical decompression, the allograft failed to respond appropriately due to the delay in surgical intervention. This is the third reported case of allograft loss from acute Page kidney following kidney transplantation. This case reinforces that kidney care differs if the kidney is solitary or a transplant. early recognition and aggressive treatments are mandatory, especially in a case with Doppler signs that are suggestive of compression.

JAX-CNV:A Whole-genome Sequencing-based Algorithm for Copy Number Detection at Clinical Grade Level

We aimed to develop a whole-genome sequencing(WGS)-based copy number variant(CNV)calling algorithm with the potential of replacing chromosomal microarray assay(CMA)for clinical ***-CNV is thus developed for CNV detection from WGS *** performance of this CNV calling algorithm was evaluated in a blinded manner on 31 samples and compared to the 112 CNVs reported by clinically validated CMAs for these 31 *** result showed that JAX-CNV recalled 100%of these ***,JAX-CNV identified an average of 30 CNVs per individual,representing an approximately seven-fold increase compared to calls of clinically validated *** validation of 24 randomly selected CNVs showed one false positive,i.e.,a false discovery rate(FDR)of 4.17%.A robustness test on lowercoverage data revealed a 100%sensitivity for CNVs larger than 300 kb(the current threshold for College of American Pathologists)down to 10×*** CNVs larger than 50 kb,sensitivities were 100%for coverages deeper than 20×,97%for 15×,and 95%for 10×.We developed a WGS-based CNV pipeline,including this newly developed CNV caller JAX-CNV,and found it capable of detecting CMA-reported CNVs at a sensitivity of 100%with about a FDR of 4%.We propose that JAX-CNV could be further examined in a multi-institutional study to justify the transition of first-tier genetic testing from CMAs to ***-CNV is available at https://***/TheJacksonLaboratory/JAX-CNV.

Assessment of a Novel Surgical Technique for Acoustic Transmitter Insertion

This study compared three surgical techniques to implant acoustic transmitters in rainbow trout (Oncorhynchus mykiss): 1) acoustic transmitter insertion via a ventral incision closed with two sutures, 2) insertion via a ventral incision with no sutures, 3) insertion via a lateral incision with no sutures. A control group consisting of fish that were only anesthetized and handled was also included. Tag retention, wound healing, inflammation, growth, and survival were recorded over a 12-e="font-family:Verdana;">e="font-family:Verdana;">e="font-family:Verdana;">week period. Tag retention was significantly different among the treatments, at 100% in the unsutured ventral incision group, 89% in the lateral incision group, and 63% in the sutured ventral incision group. Surgical wounds in the unsutured treatment groups showed significantly less inflammation than the sutured treatment. Novel Surgical Technique for Acoustic Transmitter Insertion in Rainbow Trout Reduces the Need for Surgical Training

The implantation of acoustic transmitters into the peritoneal cavity of fish is typically performed by experienced surgeons. This study compared the effectiveness of an experienced and inexperienced surgeon performing two different types of transmitter insertion techniques on rainbow trout (Oncorhynchus mykiss). The methods used were either a traditional sutured ventral incision or a novel unsutured lateral incision. There was no significant difference in surgical duration, tag retention, or fish survival between the experienced and naïve surgeon. The time to complete surgical wound closure was not significantly different between the fish operated on by an experienced or naïve surgeon, and surgeon experience had no significant effect on the duration of inflammation. However, there were significant differences between the two surgical techniques, regardless of surgeon experience. The ventral incision with sutures method took approximately three times longer to complete than the lateral incision-only method. However, complete wound closure was significantly faster in the sutured ventral incision treatment compared to the unsutured lateral treatment. Post-surgery inflammation was over seven times longer in the fish with sutures compared to those only receiving a lateral incision. Tag retention was not significantly different between the two surgical methods. The results of this study indicate that the unsutured lateral surgical technique used in this experiment can be completed with minimal training and surgical experience in contrast to the standard technique requiring sutures. These results also provide additional support to the use of sutureless surgical techniques for acoustic transmitter implantation.

INTRODUCTION Plant exudates are a common,yet often overlooked food source eaten by 94 mammalian species spread across 6 orders(Nash&Burrows 2010;Cabana et al.2018).However,exudates(including gums,resins,and latex,defined following Cabana et al.2018)contain large amounts of non-digestible carbohydrates and plant secondary metabolites,posing challenges for mammals exploiting this resource(Power 2010).Due to the difficulty of gaining nutrition from exudates,they are not considered a high-quality food source,despite the number of species that consume them(Power 2010;Cabana et al.2018).
