Neurodegeneration of the central nervous system(CNS)underscores many of humanity’s most debilitating diseases,including Alzheimer’s disease,Parkinson’s disease,and multiple sclerosis.Recently,the nitric oxide-guanylate cyclase-cyclic guanosine monophosphate(NOGC-cGmP)signaling pathway has gained traction as a neuroprotective pathway in the CNS.As an extension of the CNS,the retina is also susceptible to neurodegeneration with age.The most prevalent optic neuropathy is glaucoma,the world’s leading cause of irreversible blindness,predicted to affect more than 112 million people worldwide by 2040(Calkins,2021).In glaucoma,vision loss occurs due to the progressive degeneration of retinal ganglion cells(RGCs),the output neurons of the retina,along with their axons which form the optic nerve(Wareham et al.,2022).Degeneration of RGCs leads to a characteristic pattern of scotopic visual field deficiencies that spread from one retinotopic sector to the next(Elze et al.,2015).Visual deficits are linked to increasing age and the sensitivity of RGCs to intraocular pressure(Calkins,2021).

Escaping the control of natural enemies is thought to heavily influence the establishment success and impact of non-native species.Here,we examined how the profitability of alternative prey in combination with the presence of a competitor and predator aggressive behavior explain individual differences in diet specialization and the consumption of the invasive green porcelain crab Petrolisthes armatus by the native mud crab predator Panopeus herbstii.Results from bomb calorimetry estimates show that invasive P.armatus has high caloric value relative to alternative native prey.Laboratory assays indicated that specialization and consumption of invasive P.armatus was mostly exhibited by large,female P.herbstii,but the presence of a competitor and predator aggressiveness did not influence diet and the consumption of P.armatus.Thus,intrinsic factors(e.g.,sex and body size)seem to explain consumption of P.armatus ar\6 dietary specialization in P.herbstii,more generally.Although there are still many predator individuals that do not consume P.armatus,the proportion of individuals that have begun to specialize on P.armatus suggests that for some,it has become more profitable relative to alternative native prey.Given the high caloric value of P.armatus,we suggest that it is likely that differences in the cost of its consumption,including attack,capture,and handling times relative to alternative prey,determine its net profitability to individual predators.

Determining the role that social learning plays in construction behaviors,such as nest building or tool manufacture,could be improved if more experimental control could be gained over the exact public information that is provided by the demonstrator,to the observing individual.Using video playback allows the experimenter to choose what information is provided but will only be useful in determining the role of social learning if observers attend to,and learn from,videos in a manner that is similar to live demonstration.The goal of the current experiment was to test whether live-streamed video presentations of nest building by zebra finches Taeniopygia guttata would lead observers to copy the material choice demonstrated to them.Here,males that had not previously built a nest were given an initial preference test between materials of 2 colors.Those observers then watched live-stream footage of a familiar demonstrator building a nest with material of the color that the observer did not prefer.After this experience,observers were given the chance to build a nest with materials of the 2 colors.Although two-thirds of the observer males preferred material of the demonstrated color after viewing the demonstrator build a nest with material of that color more than they had previously,their preference for the demonstrated material was not as strong as that of observers that had viewed live demonstrator builders in a previous experiment.Our results suggest researchers should proceed with caution before using video demonstration in tests of social learning.

Background Physical activity(PA)and reductions in sedentary behavior(SED)may mitigate cancer-related cognitive impairment.The purpose of this study was to examine(a)the associations between changes in PA,SED,and cognitive function in cancer survivors before and during the coronavirus disease 2019(COVID-19)pandemic;and(b)clinical subgroups that moderate this association.methods A cross-sectional survey was administered online to adult cancer survivors globally between July and November of 2020.This was a secondary analysis of a cross-sectional survey examining changes in self-reported PA and quality of life in cancer survivors before and during the COVID-19 pandemic.Self-reported Questionnaires assessed moderate-to-vigorous PA(mVPA)using the modified Godin Leisure Time Exercise Questionnaire,cognitive function using the Functional Assessment of Cancer Therapy-Cognitive Function(FACT-Cog)scale,and SED using the Domain-specific Sitting Time questionnaire.Cancer survivors were classified into no change in behavior,desirable change(i.e.,increase mVPA to meet PA guidelines or decrease SED by≥60 min/day),and undesirable change(i.e.,decrease mVPA tomin/week or increase SED by≥60 min/day).Analysis of covariance examined differences in FACT-Cog scores across the activity change categories.Planned contrasts compared differences in FACT-Cog scores between cancer survivors with(a)no meaningful change vs.any change,and(b)a desirable change vs.an undesirable change.Results There were no significant differences in FACT-Cog scores across activity-change categories in the full sample of cancer survivors(n=371;age=48.6±15.3 years(mean±SD)).However,cancer survivors who were diagnosed≥5 years ago(t(160)=–2.15,p=0.03)or who received treatment≥5 years ago(t(102)=–2.23,p=0.03)and who had a desirable change in activity reported better perceived cognitive abilities than those who had an undesirable change.Conclusion PA promotion efforts should consider reducing SED in addition to maintaining mVPA in long-term cancer survivors to miti

Animals are increasingly faced with human?induced stressors that vary in space and time, thus we can expect populati on-level diverge nee in behaviors that help animals to cope with en vironme ntal change. However, empirical evidenee of behavioral trait divergence across environmental extremes is lacking. We tested for variation in behavioral traits among 2 populations of an African cichlid fish (Pseudocrenilabrus multicolor victoriae Seegers, 1990) that experience extremes of dissolved oxygen (DO) and turbidity and are known to vary in a number of physiological and life history traits associated with these stressors. Using a comm on garden reari ng experime nt, F1 progeny from wild-caught parents originating from a swamp (low DO, clear) and a river (high DO, turbid) were reared in high DO, clear water. Predator simulation assays were conducted to test for (1) variation in boldness, general activity, and foraging activity between populations,(2) differences in correlations betwee n behaviors with in and across populati ons, and (3) repeatability of behaviors. There was strong evide nee for diverge nee betwee n populati ons, with swamp fish being more bold (i.e.z leaving refuge sooner after a simulated predator attack) and active (i.e., spent more time out of refuge) than river fish. Across populations there were positive correlations between foraging activity and both boldness and general activity;however, within populations, there was only a strong positive relationship between foraging activity and boldness in the river population. Here, we have demonstrated that populations that originate from drastically different environments can produce proge ny that exhibit measurable differences in behaviors and their correlated relati on ships eve n whe n reared un der comm on conditions.

Toll-like receptor 5 （TLR5） signaling in response to flagellin is dispensable for inducing humoral immunity, but alterations of aa 89-96, the TLR5 binding site, significantly reduced the adjuvanticity of flagellin. These observations indicate that the underlying mechanism remains incompletely understood. Here, we found that the native form of Salmonella typhimurium aa 89-96-mutant flagellin extracted from flagella retains some TLR5 recognition activity, indicating that aa 89-96 is the primary, but not the only site that imparts TLR5 activity. Additionally, this mutation impaired the production of IL-lp and IL-18. Using TLR5KO mice, we found that aa 89-96 is critical for the humoral adjuvant effect, but this effect was independent of TLR5 activation triggered by this region of flageUin. In summary, our findings suggest that aa 89-96 of flagellin is not only the crucial site responsible for TLR5 recognition, but is also important for humoral immune adjuvanticity through a TLR5-independent pathway.

The Slit family of axon guidance cues act as repulsive molecules for precise axon pathfinding and neuronal migration during nervous system development through interactions with specific Robo receptors.Although we previously reported that Slit1–3 and their receptors Robo1 and Robo2 are highly expressed in the adult mouse peripheral nervous system,how this expression changes after injury has not been well studied.Herein,we constructed a peripheral nerve injury mouse model by transecting the right sciatic nerve.At 14 days after injury,quantitative real-time polymerase chain reaction was used to detect mRNA expression of Slit1–3 and Robo1–2 in L4–5 spinal cord and dorsal root ganglia,as well as the sciatic nerve.Immunohistochemical analysis was performed to examine Slit1–3,Robo1–2,neurofilament heavy chain,F4/80,and vimentin in L4–5 spinal cord,L4 dorsal root ganglia,and the sciatic nerve.Co-expression of Slit1–3 and Robo1–2 in L4 dorsal root ganglia was detected by in situ hybridization.In addition,Slit1–3 and Robo1–2 protein expression in L4–5 spinal cord,L4 dorsal root ganglia,and sciatic nerve were detected by western blot assay.The results showed no significant changes of Slit1–3 or Robo1–2 mRNA expression in the spinal cord within 14 days after injury.In the dorsal root ganglion,Slit1–3 and Robo1–2 mRNA expression were initially downregulated within 4 days after injury;however,Robo1–2 mRNA expression returned to the control level,while Slit1–3 mRNA expression remained upregulated during regeneration from 4–14 days after injury.In the sciatic nerve,Slit1–3 and their receptors Robo1–2 were all expressed in the proximal nerve stump;however,Slit1,Slit2,and Robo2 were barely detectable in the nerve bridge and distal nerve stump within 14 days after injury.Slit3 was highly ex-pressed in macrophages surrounding the nerve bridge and slightly downregulated in the distal nerve stump within 14 days after injury.Robo1 was upregulated in vimentin-positive cells and migrating Schwann cells inside the nerve bridge.Robo1 was a

Infection of bone tissue,or osteomyelitis,has become a growing concern in modern healthcare due in no small part to a rise in antibiotic resistance among bacteria,notably Staphylococcus aureus.The current standard of care involves aggressive,prolonged antibiotic therapy combined with surgical debridement of infected tissues.While this treatment may be sufficient for resolving a portion of cases,recurrences of the infection and associated risks including toxicity with long-term antibiotic usage have been reported.Therefore,there exists a need to produce safer,more efficacious options of treatment for osteomyelitis.In order to test treatment regimens,animal models that closely mimic the clinical condition and allow for accurate evaluation of therapeutics are necessary.Establishing a model that replicates features of osteomyelitis in humans continues to be a challenge to scientists,as there are many variables involved,including choosing an appropriate species and method to establish infection.This review addresses the refinement of animal models of osteomyelitis to reflect the clinical disease and test prospective therapeutics.The aim of this review is to explore studies regarding the use of animals for osteomyelitis therapeutics research and encourage further development of such animal models for the translation of results from the animal experiment to human medicine.

Sarcopenic obesity in patients awaiting liver transplant: Unique challenges for nutritional recommendations

Sarcopenic obesity increases the risk of mortality in patients with liver disease awaiting liver transplantation and in the post-transplant period.Nutrition recommendations for individuals with sarcopenia differ from recommendations for patients with obesity or sarcopenic obesity.While these nutrition guidelines have been established in non-cirrhotic patients,established guidelines for liver transplant candidates with sarcopenic obesity are lacking.In this paper,we review existing literature on sarcopenic obesity in patients with chronic liver disease and address opportunities to improve nutritional counseling in patients awaiting liver transplantation.
