Nutrient Analysis Critical Control Point (naCCP): Hazelnut as a Prototype of Nutrigenomic Study

The aim of this study is to apply the Nutrient Analysis Critical Control Point (naCCP) process to ensure that the highest nutrient levels in food can determine a beneficial effect on the health of the consumer. The naCCP process involves a sequence of analysis and controls that depart from raw material production to the evaluation of the effect of nutrition on health. It is articulated through the following points: 1) identification of nutrient level in the food;2) identification of critical control points (environmental, genetic data, chemical and physical data, production technology, distribution and administration);3) establishing critical limits that can impoverish and damage the nutrient;4) establishing measures to monitor;5) establishing corrective actions. We selected as biomarkers the total phenolic content (TPC) and total antioxidant capacity (TAC) of a genotyped Italian hazelnut cultivars (Corylus e avellana L.). We performed a clinical study evaluating: a) nutritional status;b) clinical-biochemical parameters;c) low density lipoprotein oxidation (LdL-ox);d) the expression level changes of oxidative stress pathway genes in the blood cell at baseline and after 40 g/die of hazelnut consumption. In this study, we found a significant lowering (p ≤ 0.005) of LdL oxidized proteins, in association with the consumption of 40 g/d of hazelnuts. Also, we found a significant variation (p ≤ 0.005) of gene expression of antioxidant and pro-oxidant genes, between the intake of dietary with and without hazelnuts. This results support the hypothesis that the naCCP process could be applied to obtain significant benefits in terms of primary prevention and for contributing to the amelioration of food management at the consumer level.

Steady-state kinetics of <i>Roystonea regia</i>palm tree peroxidase

Royal palm tree peroxidase (RPTP) has been isolated to homogeneity from leaves of Roystonea regia palm trees. The enzyme purification steps included homogenization, (NH4)SO4 precipitation, extraction of palm leaf colored compounds and consecutive chromatography on Phenyl-Sepharose, TSK-Gel dEAE-5PW and Superdex-200. The novel peroxidase was characterized as having a molecular weight of 48.2 ± 3.0 kda and an isoelectric point pI 5.4 ± 0.1. The enzyme forms dimers in solution with approximate molecular weight of 92 ± 2 kda. Here we investigated the steady-state kinetic mechanism of the H2O2-supported oxidation of different organic substrates by RPTP. The results of the analysis of the initial rates vs. H2O2 and reducing substrate concentrations were seen to be consistent with a substrate-inhibited Ping-Pong Bi-Bi reaction mechanism. The phenomenological approach used expresses the peroxidase Ping-Pong mechanism in the form of the Michaelis-Menten equation and affords an interpretation of the effects in terms of the kinetic parameters KmH2O2, KmAH2, kcat, KSIH2O2, KSIAH2 and of the microscopic rate constants k1 and k3 of the shared three-step peroxidase catalytic cycle. furthermore, the concentration and time-dependences and the mechanism of the suicide inactivation of RPTP by hydrogen peroxide were studied kinetically with guaiacol as co-substrate. The turnover number (r) of H2O2 required to complete the inactivation of the enzyme was 2154 ± 100 and the apparent rate constants of catalysis 185 s–1 and 18 s–1.

Activated Montmorillonite was applied for the sorption of pyridine as a health-toxic substance. fourier-transform Infrared spectroscopy (fTIR), zeta potential analysis, thermogravimetric analysis (TGA), N2 physisorption analysis and X-ray diffraction (XRd) method were used to investigate the structure and composition of Montmorillonite. The batch sorption system was used to study the effect of different parameters on the adsorption. Results had shown that neutral pH had a significant effect on the sorption of pyridine;thermodynamic calculations indicated that the adsorption of pyridine over Montmorillonite was exothermic reaction and the equilibrium capacity was slightly increased by using H2SO4 as activating agent by comparison with that obtained by using HCl and H3PO4. fitting the experimental data to isotherms models indicated that the experimental data were well fitted by fowler Guggenheim isotherm models. finally the results obtained by fTIR and TGA proved that the relative affinity of the pyridine toward the Montmorillonite was related to the electron donor-acceptor complexes that were formed between the basic sites (nitrogen or oxygen) and hydrogens (acidic site).

We present results of a long-term study aimed to optimize Nile tilapia O. niloticus production in a recirculating aquaculture system (RAS) in México City. Three experiments were evaluated: 1) fry production;2) production of masculinized fries;and 3) assessment of growth under three different densities (10, 30 and 75 fish/m3). The broodstock produced on average 2400 ± 600 fry every 15 days during six months, with a total production of 28,800 ± 2000. One hundred percent males were produced in 274-L raceways in an eight-week period. The obtained results in fish production in the RAS system indicated variations related to density. A daily growth rate of 3.6 g/day was registered with a density of 10 fish/m3, 0.9 g/day in 30 fish/m3 and 0.4 g/day in 75 fish/m3. This information could be useful for production purposes in this type of systems.

Effect of nanoparticles on Mechanical Properties of Cement-Sand Mortar Applications

Cement-sand mortar is a construction material widely used;tile adhesive is one of the main applications where the compressive strength is the key performance metric. nanoparticles like MWCNTs and HNTs are currently being used to improve the mechanical performance of the cement mortar due to their properties. The cost of MWCNTs has limited their application in the construction industry. This study focuses on the effect of two different concentrations of MWCNTs and HNTs in cement mortar;HNTs are proposed in this study to compare performance and cost overall benefits. A common problem with MWCNTs is agglomeration;to achieve a proper dispersion an ultrasonic processor was utilized. Mechanical properties of a cement mortar modified with nanoparticles were investigated with compression and bending tests on specimens performed at the age of 7 days. Results showed that concentrations of 0.1% of MWCNTs increase in compression strength by 56% and concentrations of 0.1% of HNTs performance increased by 31%, both compared to plain cement mortar.
