Gluten immunogenic peptide excretion detects dietary transgressions in treated celiac disease patients

BACKGROUND Life-long removal of gluten from the diet is currently the only way to manage celiac disease(CeD). Until now, no objective test has proven useful to objectively detect ingested gluten in clinical practice. Recently, tests that determine consumption of gluten by assessing excretion of gluten immunogenic peptides(GIP) in stool and urine have been developed. Their utility, in comparison with conventional dietary and analytical follow-up strategies, has not been fully *** To assess the performance of enzyme-linked immunosorbent assay(ELISA) and point-of-care tests(PoCTs) for GIP excretion in CeD patients on gluten-free diet(GfD).METHODS We conducted an observational, prospective, cross-sectional study in patients following a GfD for at least two years. Using the Gastrointestinal Symptom Rating Scale questionnaire, patients were classified at enrollment as asymptomatic or symptomatic. Gluten consumption was assessed twice by 3-d dietary recall and GIP excretion(by ELISA in stool and PoCTs(commercial kits for stool and urine) in two consecutive samples. These samples and dietary reports were obtained 10 day apart one from the other. Patients were encouraged to follow their usual GfD during the study *** forty-four patients were enrolled, of which 19(43.2%) were symptomatic despite being on a GfD. Overall, 83 sets of stool and/or urine samples were *** out of 44 patients(25.0%) had at least one positive GIP test. The occurrence of at least one positive test was 32% in asymptomatic patients compared with 15.8% in symptomatic patients. GIP was concordant with dietary reports in 65.9% of cases(Cohen′s kappa: 0.317). PoCT detected dietary indiscretions. Nonmuscle myosin Ⅱ regulates migration but not contraction in rat hepatic stellate cells

AIM: To identify and characterize the function of non-mu-scle myosin Ⅱ (NMM Ⅱ) isoforms in primary rat hepatic stellate cells (HSCs).METHODS: Primary HSCs were isolated from male Spra-gue-Dawley rats by pronase/collagenase digestion. Total RNA and protein were harvested from quiescent and culture-activated HSCs. NMM Ⅱ isoform (Ⅱ-A, Ⅱ-B and Ⅱ-C) gene and protein expression were measured by RealTime polymerase chain reaction and Western blot analyses respectively. NMM Ⅱ protein localization was visualized in vitro using immunocytochemical analysis. for in vivo assessment, liver tissue was harvested from bile duct-ligated (BDL) rats and NMM Ⅱisoform expression determined by immunohistochemistry. Using a selective myosin Ⅱ inhibitor and siRNA-mediated knockdown of each isoform, NMM Ⅱ functionality inprimary rat HSCs was determined by contraction and migration ***: NMM Ⅱ-A and Ⅱ-B mRNA expression was increased in culture-activated HSCs (Day 14) with sig-niflicant increases seen in all pairwise comparisons (Ⅱ-A: 12.67 ± 0.99 (quiescent) vs 17.36 ± 0.78 (Day 14), P f icant differences were observed in NMM Ⅱ-C mRNA and protein expression between quiescent and activated HSCs. AIM: To investigate the protein expression of phosphatase and tensin homolog (PTEN) in human liver biopsies of patients with alcoholic and non-alcoholic liver ***: PTEN protein expression was assessed by immunohistochemistry in formalin-fixed, paraffin-embedded liver sections of patients with non-alcoholic fatty liver disease (NAfLD) (n = 44) or alcoholic liver disease (ALD) (n = 25). Liver resections obtained from 3 healthy subjects candidate for partial liver donation served as controls. Histological evaluations were performed by two experienced pathologists, and diagnoses established based on international criteria. The intensity of the PTEN staining in nuclei was compared between steatotic and non-steatotic areas of each liver fragment analyzed. for each liver specimen, the antibody-stained sections were examined and scored blindly by three independent observers, who were unaware of the patients’ clinical ***: In healthy individuals, PTEN immunostaining was intense in both the cytoplasm and nuclei of all hepatocytes. However, PTEN was strongly downregulated in both the nucleus and the cytoplasm of hepatocytes from steatotic areas in patients with NAfLD, independently of the disease stage. In contrast, no changes in PTEN protein expression were observed in patients with ALD, regardless of the presence of steatosis or the stage of the disease. The degree of PTEN downregulation in hepatocytes of patients with NAfLD correlated with the percentage of steatosis (r = 0.3061, P = 0.0459) and the BMI (r = 0.4268, P = 0.0043). Vitamin D Is Associated with Severity and Mortality of Non-alcoholic fatty Liver Disease: A US Population-based Study

Background and Aims:There has been increasing evidence that vitamin D deficiency may increase the risk of metabolic *** metabolic syndrome is a major risk factor for non-alcoholic fatty liver disease (NAfLD),we aimed to investigate the association between vitamin D and the severity and mortality of ***:Data was obtained from the United States Third National Health and Nutrition Examination Survey conducted in 1988-1994,with followup mortality data through *** was defined by ultrasonographic detection of hepatic steatosis in the absence of other liver diseases and categorized as normal,mild,moderate or *** severity of hepatic fibrosis was determined by NAfLD fibrosis score (NfS).ANOVA (f-test) was used to evaluate the association between vitamin D level and degree of NAfLD,and Cox proportional hazards regression analysis was used for survival ***:Vitamin D levels for normal,mild,moderate and severe steatosis were 25.1 ± 0.29 ng/mL,24.7 ± 0.42 ng/mL,23.7 ± 0.37 ng/mL and 23.6 ± 0.60 ng/mL,respectively (trend p for low,intermediate and high NfS categories were 24.7 ± 0.38 ng/mL,23.4 ± 0.42 ng/mL and 21.5 ± 0.57 ng/mL,respectively (trend p fter median-follow up over 19 years,vitamin D deficiency was significantly associated with diabetes-and Alzheimer's diseaserelated mortality (hazard ratio (HR):3.64,95%CI:1.51-8.82 and HR:4.80,95%CI:1.53-15.1,respectively),with a borderline significance in overall mortality (HR:1.16,95%CI:0.99-1.36,p =0.06).Conclusion:Vitamin D level was inversely related to the degree of liver steatosis and ***,vitamin D deficiency was associated with diabetes-and Alzheimer's disease-related mortality in NAfLD patients.

The molecular mechanisms controlling mouse embryonic stem cell(ESC)metastability,*** capacity to fluctuate between different states of pluripotency,are not fully *** developed and used a novel automation platform,the Cellmaker,to screen a library of metabolites on two ESC-based phenotypic assays(*** and colony phenotype)and identified two metabolically related amino acids,namely L-proline(L-Pro)and L-ornithine(L-Orn),as key regulators of ESC *** compounds,but mainly L-Pro,force ESCs toward a novel epiblast stem cell(EpiSC)-like state,in a dose-and time-dependent *** EpiSCs,L-Pro-induced cells(PiCs)contribute to chimeric embryos and rely on leukemia inhibitor factor(LIf)to ***,PiCs revert to ESCs or differentiate randomly upon removal of either L-Pro or LIf,***,PiC generation depends on both L-Pro metabolism(uptake and oxidation)and fgf5 induction,and is strongly counteracted by antioxidants,mainly L-ascorbic acid(vitamin C,Vc).ESCs↔PiCs phenotypic transition thus represents a previously undefined dynamic equilibrium between pluripotent states,which can be unbalanced either toward an EpiSC-like or an ESC phenotype by L-Pro/L-Orn or Vc treatments,*** together,our data provide evidence that ESC metastability can be regulated at a metabolic level.

Rayleigh waves from correlation of seismic noise in Great Island of Tierra del fuego,Argentina:Constraints on upper crustal structure

In this study, the ambient seismic noise cross-correlation technique is applied to estimate the upper structure of the crust beneath Great Island of Tierra del fuego（Tdf）, Argentina, by the analysis of shortperiod Rayleigh wave group velocities. The island, situated in the southernmost South America, is a key area of investigation among the interaction between the South American and Scotia plates and is considered as a very seismically active one. Through cross-correlating the vertical components of ambient seismic noise registered at four broadband stations in Tdf, we were able to extract Rayleigh waves which were used to estimate group velocities in the period band of 2.5-16 s using a timefrequency analysis. Although ambient noise sources are distributed in homogeneously, robust empirical Green＇s functions could be recovered from the cross-correlation of 12 months of ambient noise, The observed group velocities were inverted considering a non-linear iterative damped least-squares inversion procedure and several 1-D shear wave velocity models of the upper crust were *** to the inversion results, the S-wave velocity ranges between 1.75 and 3,7 km/s in the first10 km of crust, depending on the pair of stations considered. These results are in agreement to the major known surface and sub-surface geological and tectonic features known in the area. This study represents the first ambient seismic noise analysis in Tdf in order to constraint the upper crust beneath this *** can also be considered as a successful feasibility study for future analyses with a denser station deployment for a more detailed imaging of structure.

N-acetylcysteine in acute pancreatitis

Premature trypsinogen activation and production of oxygen free radicals (OfR) are early pathogenic events which occur within acinar cells and trigger acute pancreatitis (AP). OfR exert their harmful effects on various cell components causing lipid peroxidation, disturbances in calcium homeostasis and DNA damage, which lead to increased cell injury and eventually cell death. This review presents the most recent data concerning the effects of N-Acetylcysteine (NAC), in the treatment of AP. NAC is an antioxidant capable of restoring the levels of Glutathione, the most important cellular antioxidant. Studies show the benef icial effects of NAC treatment in preventing OfR production and therefore attenuating oxidative damage. Additionally, NAC treatment has been shown to prevent the increase in cytosolic Ca2+ concentration and reduce the accumulation of enzymes in acinar cells during AP. The prevention, by NAC, of these pathological events occurring within acinar would contribute to reducing the severity of AP. NAC is also capable of reducing the activation of transcription factors especially sensitive to the cellular redox state, such as Nuclear factor-κB, signal transducer and activator of transcription-3 and mitogenactivated protein kinase. This leads to a down-regulation of cytokines, adhesion molecules and chemokine expression in various cell types during AP. These f indingspoint to NAC as a powerful therapeutic treatment, attenuating oxidative-stress-induced cell injury and other pathological events at early stages of AP, and potentially contributing to reducion in the severity of disease.
