BACKGROUND Metabolic-dysfunction associated steatotic liver disease(MASLD)is a hepatic manifestation of metabolic *** suggest ornithine aspartate(LOLA)as drug *** To analyze the influence of LOLA intake on gut microbiota using a nutritional model of *** Adult male Sprague Dawley rats were randomized into three groups:Control(10 rats fed with a standard diet),MASLD(10 rats fed with a high-fat and choline-deficient diet),and LOLA(10 rats receiving 200 mg/kg/d LOLA,after the 16th week receiving high-fat and choline-deficient diet).After 28 wk of the experiment,animals were euthanized,and feces present in the intestine were *** fecal DNA extraction,the V4 region of the 16S rRNA gene was amplified followed by sequencing in an Ion S5™*** Alpha and beta diversity metrics were comparable between MASLD and LOLA.3 OTUs were differentially abundant between MASLD and LOLA,which belong to the species Helicobacter rodentium,Parabacteroides goldsteinii,and Parabacteroides *** functional prediction provided two different metabolic profiles between MASLD and *** 9 pathways differentially abundant in MASLD are related to a change in energy source,adenosine/purine nucleotides degradation as well as guanosine and adenosine deoxyribonucleotides *** 14 pathways differentially abundant in LOLA are associated with four major metabolic functions primarily influenced by L-aspartate,including tricarboxylic acid cycle pathways,purine/guanosine nucleotides biosynthesis,pyrimidine ribonucleotides biosynthesis and salvage as well as lipid IVA *** Although LOLA had no influence on alpha and beta diversity in this nutritional model of MASLD,it was associated with changes in specific gut microbes and their related metabolic pathways.

Epigenetic marks are modulated by gender and time of the day in the hippocampi of adolescent rats:a preliminary study

Although the involvement of gender in epigenetic machinery in peripheral tissues during the neonatal period has been suggested, the gender-related epigenetic profile of brain areas during the adolescent period is rarely exploited. Furthermore, the influence of time of day on hippocampal acetylation marks has been demonstrated in young adult and aged rats; however, there are no studies reporting epigenetic changes in the adolescent period. Therefore, this study aimed to investigate the effects of gender on hippocampal DNA methyltransferase 1 content and histone deacetylase（HDAC） activity of adolescent rats at different time points, specifically early morning and afternoon. Both epigenetic markers increased significantly in the hippocampi of female rats compared to the male group, an indicator of reduced transcriptional activity. In addition, HDAC activity during the early morning was higher compared to afternoon groups in both male and female rats, while DNA methyltransferase 1 content was not altered by the time of day. Our findings demonstrate that hippocampal DNA methylation and histone acetylation status can be influenced by gender during the adolescent period, while the time of the day impacts HDAC activity.

Here we present new data on the major and trace element compositions of silicate and oxide minerals from mantle xenoliths brought to the surface by the carolina kimberlite,Pimenta Bueno Kimberlitic Field,which is located on the southwestern border of the Amazonian *** also present Sr-Nd isotopic data of garnet xenocrysts and whole-rocks from the carolina *** xenoliths are mainly clinopyroxenites and *** of the clinopyroxenites were classified as GPP–PP–PKP(garnet-phlogopite peridotite,phlogopite-peridotite,phlogopite-K-richterite peridotite)suites,and two clinopyroxenites(eclogites)and two garnetites are relicts of an ancient subducted *** and pressure estimates yield 855–1102℃ and 3.6–7.0 GPa,*** are enriched in light rare earth elements(LREE)(La_(N)/Yb_(N)=5–62;Ce_(N)/Sm_(N)=1–3;where N=primitive mantle normalized values),they have high Ca/Al ratios(10–410),low to medium Ti/Eu ratios(742–2840),and low Zr/Hf ratios(13–26),which suggest they were formed by metasomatic reactions with CO_(2)-rich silicate *** with high TiO_(2)(>2.0 wt.%),Al_(2)O_(3)(>12.0 wt.%),and FeOt(5.0–13.0 wt.%)resemble those found in the groundmass of kimberlites,lamproites and ***,phlogopite with low TiO_(2)(de)and PIC(phlogopite-ilmenite-clinopyorxene)*** GPP-PP-PKP suite of xenoliths,together with the clinopyroxene and phlogopite major and trace element signatures suggests that an intense proto-kimberlite melt metasomatism occurred in the deep cratonic lithosphere beneath the Amazonian *** Sr-Nd isotopic ratios of pyrope xenocrysts(G3,G9 and G11)from the carolina kimberlite are characterized by high ^(143)Nd/^(144)Nd(0.51287–0.51371)and eNd(+4.55 to+20.85)accompanied with enriched ^(87)Sr/^(86)Sr(0.70405–0.71098).These results suggest intera

This work describes the synthesis, antinociceptive and anti-inflammatory effect of O-prenyl derivatives: O-prenyl derivatives 2-(3-methylbut-2-enyloxy)acetophenone (L1) and 3-methoxy-4-(3-methylbut-2-enyloxy)benzaldehyde (V1). Treatment with L1- or V1-produced antinociceptive effect on classical pain models: acetic acid abdominal contortions and formalin test (first and second phases), and in hot plate or tale flick models in mice without changing the locomotors performance, and anti-oedematogenic or anti-inflammatory effect in vivo: carrageenan-induced paw oedema and peritonitis in mice. In addition, L1 and V1 derivatives reduced nitric oxide production on RAW 264.7 cells stimulated with lipopolysaccharide without changing the cell viability. Taken together, our results show for the first time that L1 and V1 can produce antinociception and modulate inflammatory response when administered in mice.

Cultivated potato is a clonally propagated autotetraploid species with a highly heterogeneous *** assemblies of six cultivars including two chromosome-scale phased genome assemblies revealed extensive allelic diversity,including altered coding and transcript sequences,preferential allele expression,and structural variation that collectively result in a highly complex transcriptome and predicted proteome,which are distributed across the homologous *** species contribute to the extensive allelic diversity in tetraploid cultivars,demonstrating ancestral introgressions predating modern breeding *** a clonally propagated autotetraploid that undergoes limited meiosis,dysfunctional and deleterious alleles are not purged in tetraploid *** a quarter of the loci bore mutations are predicted to have a high negative impact on protein function,complicating breeder’s efforts to reduce genetic *** StCDF1 locus controls maturity,and analysis of six tetraploid genomes revealed that 12 allelic variants of StCDF1 are correlated with maturity in a dosage-dependent *** of the complexity of the tetraploid potato genome with its rampant structural variation and embedded deleterious and dysfunctional alleles will be key not only to implementing precision breeding of tetraploid cultivars but also to the construction of homozygous,diploid potato germplasm containing favorable alleles to capitalize on heterosis in F1 hybrids.
