Hepatitis b virus prevalence and transmission risk factors in inflammatory bowel disease patients at Clementino Fraga Filho university hospital

AIM: To evaluate the prevalence of hepatitis b virus (HbV) infection in inflammatory bowel disease (IbD) patients that followed up in our hospital and try to identify the possible risk factors involved in this infection transmission. METHODS: This was a cross-sectional study for which 176 patients were selected according to their arrival for the medical interview. All these patients had already IbD diagnosis. The patient was interviewed and a questionnaire was filled out. RESULTS: In the group of 176 patients whom we examined, we found that 17% (30) were anti-Hbc positive. Out of 30 patients with positive anti-Hbc, 2.3% (4) had positive HbsAg and negative HbV-DNA. In an attempt to identify the possible HbV infection transmission risk factors in IbD patients, it was observed that 117 patients had been submitted to some kind of surgical procedure, but only 24 patients had positive anti-Hbc (P = 0.085). It was also observed that surgery to treat IbD complications was not a risk factor for HbV infection transmission, since we did not get a statically significant P value. However, IbDpatients that have been submitted to surgery to treat IbD complications received more blood transfusions then patients submitted to other surgical interventions (P = 0.015). CONCLUSION: There was a high incidence of positive anti-Hbc (17%) and positive HbsAg (2.3%) in IbD patient when compared with the overall population (7.9%).

Neural tissue engineering is premised on the integration of engineered living tissue with the host nervous system to directly restore lost function or to augment regenerative capacity following ner- vous system injury or neurodegenerative disease. Disconnection of axon pathways - the long-distance fibers connecting specialized regions of the central nervous system or relaying peripheral signals - is a common feature of many neurological disorders and injury. However, functional axonal regenera- tion rarely occurs due to extreme distances to targets, absence of directed guidance, and the presence of inhibitory factors in the central nervous system, resulting in devastating effects on cognitive and sensorimotor function. To address this need, we are pursuing multiple strategies using tissue engi- neered ＂living scaffolds＂, which are preformed three-dimensional constructs consisting of living neural cells in a defined, often anisotropic architecture. Living scaffolds are designed to restore function by serving as a living labeled pathway for targeted axonal regeneration - mimicking key developmental mechanisms- or by restoring lost neural circuitry via direct replacement of neurons and axonal tracts. We are currently utilizing preformed living scaffolds consisting of neuronal dusters spanned by long axonal tracts as regenerative bridges to facilitate long-distance axonal regeneration and for targeted neurosurgical reconstruction of local circuits in the brain. Although there are formidable challenges in predinical and clinical advancement, these living tissue engineered constructs represent a promising strategy to facilitate nervous system repair and functional recovery.

AIM:To investigate chronic stress as a susceptibility factor for developing pancreatitis,as well as tumor necrosis factor-α (TNF-α) as a putative ***:Rat pancreatic acini were used to analyze the influence of TNF-α on submaximal (50 pmol/L) cholecystokinin (CCK) *** restraint (4 h every day for 21 d) was used to evaluate the effects of submaximal (0.2 μg/kg per hour) cerulein stimulation on chronically stressed ***:In vitro exposure of pancreatic acini toTNF-α disorganized the actin *** was further increased by TNF-α/CCK treatment,which additionally reduced amylase secretion,and increased trypsin and nuclear factor-κb activities in a protein-kinase-C δ and ε-dependent ***-α/CCK also enhanced caspases' activity and lactate dehydrogenase release,induced ATP loss,and augmented the ADP/ATP *** vivo,rats under chronic restraint exhibited elevated serum and pancreatic TNF-α ***,pancreatic,and lung inflammatory parameters,as well as caspases' activity in pancreatic and lung tissue,were substantially enhanced in stressed/cerulein-treated rats,which also experienced tissues' ATP loss and greater ADP/ATP *** examination revealed that stressed/cerulein-treated animals developed abundant pancreatic and lung edema,hemorrhage and leukocyte infiltrate,and pancreatic *** severity was greatly decreased by treating animals with an anti-TNF-αantibody,which diminished all inflammatory parameters,histopathological scores,and apoptotic/necrotic markers in stressed/cerulein-treated ***:In rats,chronic stress increases susceptibility for developing pancreatitis,which involves TNF-α sensitization of pancreatic acinar cells to undergo injury by physiological cerulein stimulation.

This study deals with the influence of surface roughness on the color of resin *** resin composites (microfilled,hybrid,and microhybrid) were each polished with 500-grit,1 200-grit,2 000-grit,and 4 000-grit SiC *** roughness parameter (Ra) was measured using a Plμ confocal microscope,and field-emission scanning electron microscope (Fe-SEM) images were used to investigate filler *** was measured using a spectroradiometer and a D65 standard illuminant (geometry diffuse/0° specular component excluded (SCE) mode).Surface roughness decreased with grit number and was not influenced by filler size or size distribution.A significant influence of Ra on lightness (L*) was *** increased with decreases in roughness,except for specimens that underwent polishing procedure 4 (PP4;500-grit,1 200-grit,2 000-grit,and 4 000-grit SiC papers consecutively).Generally,it was found that surface roughness influenced the color of resin *** composites that underwent PP1 (500-grit SiC paper) exhibited significant differences in chroma (C*),hue (h°),and lightness (L*) compared to composites that underwent PP3 (500-grit,1 200-grit,and 2 000-grit SiC papers consecutively) and *** difference (E*) between the polishing procedures was within acceptability thresholds in dentistry.

Repurposing small molecule drugs and drug candidates is considered as a promising approach to revolutionise the treatment of snakebite *** this study,we investigated the inhibiting effects of the small molecules varespladib(nonspecific phospholipase A2 inhibitor),marimastat(broad spectrum matrix metalloprotease inhibitor)and dimercaprol(metal ion chelator)against coagulopathic toxins found in Crotalinae(pit vipers)snake *** from bothrops asper,bothrops jararaca,Calloselasma rhodostoma and Deinagkistrodon acutus were separated by liquid chromatography,followed by nanofractionation and mass spectrometry identification undertaken in *** of the venom toxins were then subjected to a high-throughput coagulation assay in the presence of different concentrations of the small molecules under *** venom toxins were mostly identified as phospholipases A2,while procoagulant venom activities were mainly associated with snake venom metalloproteinases and snake venom serine *** was found to effectively inhibit most anticoagulant venom effects,and also showed some inhibition against procoagulant ***,marimastat and dimercaprol were both effective inhibitors of procoagulant venom activities but showed little inhibitory capability against anticoagulant *** information obtained from this study aids our understanding of the mechanisms of action of toxin inhibitor drug candidates,and highlights their potential as future snakebite treatments.

In this study, the ambient seismic noise cross-correlation technique is applied to estimate the upper structure of the crust beneath Great Island of Tierra del Fuego（TdF）, Argentina, by the analysis of shortperiod Rayleigh wave group velocities. The island, situated in the southernmost South America, is a key area of investigation among the interaction between the South American and Scotia plates and is considered as a very seismically active one. Through cross-correlating the vertical components of ambient seismic noise registered at four broadband stations in TdF, we were able to extract Rayleigh waves which were used to estimate group velocities in the period band of 2.5-16 s using a timefrequency analysis. Although ambient noise sources are distributed in homogeneously, robust empirical Green＇s functions could be recovered from the cross-correlation of 12 months of ambient noise, The observed group velocities were inverted considering a non-linear iterative damped least-squares inversion procedure and several 1-D shear wave velocity models of the upper crust were *** to the inversion results, the S-wave velocity ranges between 1.75 and 3,7 km/s in the first10 km of crust, depending on the pair of stations considered. These results are in agreement to the major known surface and sub-surface geological and tectonic features known in the area. This study represents the first ambient seismic noise analysis in TdF in order to constraint the upper crust beneath this *** can also be considered as a successful feasibility study for future analyses with a denser station deployment for a more detailed imaging of structure.

Silybin counteracts lipid excess and oxidative stress in cultured steatotic hepatic cells

AIM: To investigate in vitro the therapeutic effect and mechanisms of silybin in a cellular model of hepatic ***: Rat hepatoma FaO cells were loaded with lipids by exposure to 0.75 mmol/L oleate/palmitate for 3 h to mimic liver steatosis. Then, the steatotic cells were incubated for 24 h with different concentrations (25 to 100 μmol/L) of silybin as phytosome complex with vitamin E. The effects of silybin on lipid accumulation and metabolism, and on indices of oxidative stress were evaluated by absorption and fluorescence microscopy, quantitative real-time PCR, Western blot, spectrophotometric and fluorimetric ***: Lipid-loading resulted in intracellular triglyceride (TG) accumulation inside lipid droplets, whose number and size increased. TG accumulation was mediated by increased levels of peroxisome proliferator-activated receptors (PPARs) and sterol regulatory element-binding protein-1c (SREbP-1c). The lipid imbalance was associated with higher production of reactive oxygen species (ROS) resulting in increased lipid peroxidation, stimulation of catalase activity and activation of nuclear factor kappa-b (NF-κb). Incubation of steatotic cells with silybin 50 μmol/L significantly reduced TG accumulation likely by promoting lipid catabolism and by inhibiting lipogenic pathways, as suggested by the changes in carnitine palmitoyltransferase 1 (CPT-1), PPAR and SREbP-1c levels. The reduction in fat accumulation exerted by silybin in the steatotic cells was associated with the improvement of the oxidative imbalance caused by lipid excess as demonstrated by the reduction in ROS content, lipid peroxidation, catalase activity and NF-κb ***: We demonstrated the direct anti-steatotic and anti-oxidant effects of silybin in steatotic cells, thus elucidating at a cellular level the encouraging results demonstrated in clinical and animal studies.

Stainless steel implants are suitable candidates for bone replacement due to their cytocompatibility and mechanical resistance,but they suffer from lack of bioactivity and are prone to bacterial ***,to overcome those limitations,in this study we developed by electrophoretic deposition(EPD),an innovative surface coating made of(i)zein,a natural fibroblast-friendly polymer,(ii)bioactive glass,a pro-osteogenic inorganic material and(iii)copper containing bioactive glass,an antibacterial and pro-angiogenic *** images confirmed that porous,uniform and free of cracks coatings were obtained by EPD;moreover,coatings were resistant to mechanical stress as demonstrated by the tape test,resulting in a 4b classification of adhesion to the *** coatings were cytocompatible as indicated by metabolism evaluation of human fibroblasts,endothelial cells and mature or progenitor osteoblasts cultivated in direct contact with the *** also maintained pro-osteogenic properties towards undifferentiated progenitor cells that expressed osteogenic genes after 15 days of direct *** conferred antibacterial properties as biofilm formation of the joint pathogens Staphylococcus aureus,Staphylococcus epidermidis and Escherichia coli was significantly reduced in comparison with copper-free coatings(pbacteria while the cells viability was preserved when cells and bacteria were cultivated in the same environment by a co-culture ***,copper ability to recruit blood vessels and to inhibit bacterial infection was confirmed in vivo where the growth of *** biofilm was inhibited and the formation of new(blood vessels was observed.
