Signaling transduction pathways involved in basophil adhesion and histamine release

Back.round Little is k.own about basophil with respect to the different signaling transduction pathways involved in spontaneous, cytok.ne or anti-IgE induced adhesion and how this compares to IgE-dependent and IgE-independent mediator secretion. The purpose of the present study was to investigate the roles of β1 and β2 integrins in basophil adhesion as well as hosphatidylinositol 3-k.nase （PI3k., src-k.nases and extracellular signal regulated k.nase （ERk.1/2 in basophil adhesion and histamine release （HR）. Methods Basophils （purity of 10%-50%） were preincubated with anti-CD29 or anti-CD 18 block.ng antibodies before used for adhesion study. Basophils were preincubated with the pharmacological inhibitors wortmannin, PP1, PD98059 before used for adhesion and HR study. Cell adherence to bovine serum albumin （BSA） or fibronectin （Fn） was monitored using cell associated histamine as a basophil mark.r and the histamine was measured by the glass fiber assay. Results Basophil spontaneous adhesion to Fn was inhibited by anti-CD29. Interleuk.n （IL）-3, granulocyte/macrophage colony stimulating factor （GM-CSF） induced adhesion to BSA was inhibited by anti-CD18. Wortmannin at 1 μmol/L and PP1 at 20 μmol/L strongly interfered with, whereas PD98059 at 50μmol/L weak.y inhibited basophil spontaneous adhesion to Fn. One μmol/L wortmannin strongly inhibited IL-3, IL-5, GM-CSF and anti-IgE induced adhesion to BSA. PP1 at 20 μmol/L partly inhibited anti-IgE induced adhesion. Fifty μmol/L PD98059 marginally inhibited IL-5, weak.y inhibited anti-IgE, partly inhibited GM-CSF induced adhesion. Wortmannin, PP1 and PD98059 inhibited anti-IgE （1：100 or 1：1000） induced basophil HR in a dose dependent manner. They inhibited calcium ionophore A23187 （10 μmol/L, 5 μmol/L） induced basophil HR in a dose dependent manner, but to different extend with PP1 being the most efficient. 食物过敏是异质性群体相关疾病，会通过一种或多种方式对多个器官造成影响。其反应的严重程度可从轻度的局部反应到全身性的过敏反应。从机制上来看，食物过敏是Th2细胞驱动的免疫紊乱疾病，食物特异性IgE是这种速发型不良反应的基础。过敏的部位和症状并不一定相同。食物过敏常会与自身免疫性（如乳糜泻）和非免疫性（如乳糖不耐受）的其他食物不良反应相混淆。为了正确诊断食物过敏，需要详细了解患者病史，检测特异性IgE，并进行经口激发试验来确诊。一些辅助因素（如运动、药物和酒精）有助于诱发食物过敏，会进一步增加诊断的复杂性。以食物提取物为基础的诊断试验并不能反映症状的严重程度，而检测单个食物致敏原IgE的新一代分子诊断技术，则能够为临床医生和患者提供可靠的症状严重程度依据。分子诊断还有助于确定食物过敏是直接源于对食物的暴露还是间接源于花粉过敏（交叉反应）。流行病学调查表明，桃过敏在欧洲主要是源于对桃的食用，而在中国则是源于对艾蒿花粉的初级致敏。但这两种情况都是由来自同一家族的致敏原分子介导的。流行病学调查深入揭示了食物过敏的病因、流行性及其所包含的风险因素，为预防食物过敏提供了循证策略。过去的十年中，食物过敏在发达国家盛行。经济发展和城市化进程都被认为会导致食物过敏流行性的增加，而且饮食习惯不同，过敏的食物也不同。目前，分子变态反应学和生物技术正在开发安全的免疫治疗方法，有望能够减轻日益加重的食物过敏负担。一种使用低致敏性突变重组分子的免疫疗法已经开始进行第一阶段临床试验的评估。食物致敏原名单的不断完善为转基因食品致敏性的评估提供了坚实基础。

Evaluating the addition of radar with optical data for vegetation mapping in a montane region in Sri Lank.

The use of freely-available multi-source imagery for mapping vegetation in montane terrain is important for many developing countries that do not have the funding for high-resolution data *** images are also now freely available and include Sentinel-1 in dual polarisation,and *** images can penetrate cloud cover and provide the advantage of acquiring data in a cloudy tropical *** research evaluated whether the addition of radar with optical and topographic data improves classification accuracy in a montane region in Sri *** classification experiments were designed based on different combinations of image data to test whether radar data improved land cover classification accuracy compared with optical data *** forest classifier in the Google Earth Engine has been utilised to classify the tropical montane *** results indicate that radar or optical data alone cannot obtain satisfactory ***,when combining radar with optical data the overall accuracy increased by approximately 5%,and by an additional 2%when topography data were *** highest accuracy(92%)was achieved with multiple imagery,and adding the vegetation indices improved the model slightly by 0.3%.In addition,feature importance analysis showed that radar data mak.s a significant contribution to the *** positive outcomes demonstrate that freely-accessible multi-source remotely-sensed data have impressive capability for vegetation mapping,and support the monitoring and managing of forest ecological resources in tropical montane regions.

Back.round: The clinical manifestations of mercury poisoning vary based on chemical form, dose, and route of administration. In the medical field, many of the cases of mercury exposure have been in mining agriculture, or chemical industrial work.rs via inhalation. Aim: To discuss the diagnosis of parenteral mercury poisoning with radiographic identification and potential sequelae. Case Presentation: We present the case of a young male with a chief complaint of chest pain and intravenous mercury administration found to be intentional as a suicide attempt. The chest radiographs lead to a wide differential diagnosis in a hemodynamically stable patient. Conclusion: While ingestion rarely causes toxicity due to the poor absorption of mercury through the gastrointestinal tract, intravenous elemental mercury can range from producing minimal symptoms to being severely toxic.

CFD-simulation of a circulating fluidized bed riser

In the current work. a model of the fluid mechanics in the riser of a circulating fluidized bed （CFB） has been implemented using computational fluid dynamics （CFD）. The model developed shall be used in future as the basis of 3D-reactor model for the simulation of large scale CFB combustors. The two-fluid model （TFM） approach is used to represent the fluid mechanics involved in the flow. The computational implementation is accomplished by the commercial software FLUENT. Different closure formulations are tested on a simplified geometry. Two different turbulence formulations, namely the swirl modified RNG k.e model and the Realizable k.e model, are tested in combination with two different approaches to solid phase turbulence, namely the dispersion and per phase approach. One focus of the current work.is put on the study of different drag correlations. Besides the drag correlations by Syamlal et al. [Syamlal, M., Rogers, W., ＆ O＇Brien, T. J. （1993）. MFIX documentation theory guide. Technical Report DOE/METC-9411004, U.S. Department of Energy （DOE）. Morgantown Energy Technology Center： Morgantown, WV] and Gidaspow [Gidaspow, D. （1994）. Multiphaseflow andfluidization. New York. Academic Press] the EMMS model has been used to determine the momentum exchange between the two phases. Opportunistic bacteria in apical periodontitis (AP) may pose a risk.for systemic ***-associated invariant T (MAIT) cells are innate-lik. T cells with a broad and potent antimicrobial activity important for gut mucosal *** was recently shown that MAIT cells are present in the oral mucosal tissue,but the involvement of MAIT cells in AP is ***,comparison of surgically resected AP and gingival tissues demonstrated that AP tissues express significantly higher levels of Vα7.2-Jα33,Vα7.2- Jα20,Vα7.2-Jα12,Cα and tumour necrosis factor (TNF),interferon (IFN)-γ and interleuk.n (IL)-17A transcripts,resembling a MAIT cell ***,in AP tissues the MR1-restricted MAIT cells positive for MR1–5-OP-RU tetramer staining appeared to be of similar levels as in peripheral blood but consisted mainly of CD4^+ *** gingival tissues,the AP microbiome was quantitatively impacted by factors lik. fistula and high patient age and had a prominent riboflavin-expressing bacterial *** merged in an integrated view,the examined immune and microbiome data in the sparse partial least squares discriminant analysis could identify bacterial relative abundances that negatively correlated with Vα7.2-Jα33,Cα,and IL-17A transcript expressions in AP,implying that MAIT cells could play a role in the local defence at the oral tissue *** conclusion,we describe the presence of MAIT cells at the oral site where translocation of oral microbiota could tak. *** findings have implications for understanding the immune sensing of polymicrobial-related oral diseases.

Diabetics Have the Same Risk.and Benefits Regarding Postoperative Amiodarone Prophylaxis for Atrial Fibrillation When Undergoing Surgery for Lung Cancer

Back.round: To evaluate if the risk.for developing atrial fibrillation after lung surgery is higher for diabetics than non-diabetic patients and whether diabetic status prolongs the length of in-hospital stay. Objective: To compare the outcome of amiodarone prophylaxis in diabetics and non-diabetics. Design: Subgroup analysis within a randomized, controlled, double-blinded trial. Results: Development of atrial fibrillation was equally frequent among diabetics (18.2%) and non-diabetics (20.5%) (p = 1.00). Atrial fibrillation occurred in 7.1% of prophylactic diabetics and in 9.3% of prophylactic non-diabetics, while 37.5% non-prophylactic diabetics and 31.3% non-prophylactic non-diabetics experienced atrial fibrillation (p = 0.31). Prophylactic amiodarone was equally effective in diabetics as in non-diabetics with a relative risk.of 3.5 (1.8 - 67.0) and the number need to treat of 4.4 (3.3 - 8.3) (p = 0.31). The length of in-hospital stay for diabetics was equal to non-diabetics with an average stay of 7.1 versus 8 days at Aarhus University Hospital (p = 0.61) with similar stays at intermediary and intensive care unit as well as total in-hospital stay of 8.9 versus 10 days (p = 0.60). Conclusions: Diabetics have the same risk.of atrial fibrillation and the same benefits from prophylactic amiodarone as non-diabetics after surgery for lung cancer. Furthermore, diabetics have the same length of stay as non-diabetics. No severe adverse effects were found in either group.
