Auranofin and its analogs as prospective agents for the treatment of colorectal cancer

Today colorectal cancer(CRC)is one of the leading causes of cancer death *** disease is poorly chemo-sensitive toward the existing medical treatments so that new and more effective therapeutic agents are urgently needed and intensely *** drugs,oxaliplatin in particular,were reported to produce some significant benefit in CRC treatment,triggering the general interest of medicinal chemists and oncologists for metal-based compounds as candidate anti-CRC *** this frame,gold compounds and,specifically,the established antiarthritic drug auranofin with its analogs,form a novel group of promising anticancer *** to its innovative mechanism of action and its favorable pharmacological profile,auranofin together with its derivatives are proposed here as novel experimental agents for CRC treatment,capable of overcoming resistance to platinum *** encouraging results in this direction have already been obtained.A few recent studies demonstrate that the action of auranofin may be further potentiated through the preparation of suitable pharmaceutical formulations capable of protecting the gold pharmacophore from unselective reactivity or through the design of highly synergic drug *** perspectives of the research in this field are outlined.

The effects of data aggregation on long-term projections of forest stands development

Forest management planning often relies on Airborne Laser Scanning(ALS)-based Forest Management Inventories(FMIs)for sustainable and efficient *** the area-based(ABA)approach,these inventories estimate forest characteristics for grid cell areas(pixels),which are then usually summarized at the stand *** the ALS-based high-resolution Norwegian Forest Resource Maps(16 m×16 m pixel resolution)alongside with stand-level growth and yield models,this study explores the impact of three levels of pixel aggregation(standlevel,stand-level with species strata,and pixel-level)on projected stand *** results indicate significant differences in the projected outputs based on the aggregation ***,the most substantial difference in estimated volume occurred between stand-level and pixel-level aggregation,ranging from-301 to+253 m^(3)·ha^(-1)for single *** differences were,on average,higher for broadleaves than for spruce and pine dominated stands,and for mixed stands and stands with higher variability than for pure and homogenous *** conclusion,this research underscores the critical role of input data resolution in forest planning and management,emphasizing the need for improved data collection practices to ensure sustainable forest management.

Cells,tissues,and organs are constantly subjected to the action of mechanical forces from the extracellular environment-and the nervous system is no ***-intrinsic properties such as membrane lipid composition,abundance of mechanosensors,and cytoskeletal dynamics make cells more or less likely to sense these *** and extrinsic cues are integrated by cells and this combined information determines the rate and dynamics of membrane protrusion growth or retraction(Yamada and Sixt,2019).Cell protrusions are extensions of the plasma membrane that play crucial roles in diverse contexts such as cell migration and neuronal synapse *** the nervous system,neurons are highly dynamic cells that can change the size and number of their pre-and postsynaptic elements(called synaptic boutons and dendritic spines,respectively),in response to changes in the levels of synaptic activity through a process called *** plasticity is a hallmark of the nervous system and is present throughout our lives,being required for functions like memory formation or the learning of new motor skills(Minegishi et al.,2023;Pillai and Franze,2024).

Oxygen(O_(2))-sensing matrices are promising tools for the live monitoring of extracellular O_(2) consumption levels in long-term cell *** this study,ratiometric O_(2)-sensing membranes were prepared by electrospinning,an easy,low-cost,scalable,and robust method for fabricating ***(ε-caprolactone)and poly(dimethyl)siloxane polymers were blended with tris(4,7-diphenyl-1,10-phenanthroline)ruthenium(II)dichloride,which was used as the O_(2)-sensing probe,and rhodamine B isothiocyanate,which was used as the reference *** functionalized scaffolds were morphologically characterized by scanning electron microscopy,and their physicochemical profiles were obtained by Fourier transform infrared spectroscopy,thermogravimetric analysis,and water contact angle *** sensing capabilities were investigated by confocal laser scanning microscopy,performing photobleaching,reversibility,and calibration curve studies toward different dissolved O_(2)(DO)*** sensing nanofibers showed a high response to changes in DO concentrations in the physiological-pathological range from 0.5%to 20%and good stability under ratiometric *** addition,the sensing systems were highly biocompatible for cell growth promoting adhesiveness and growth of three cancer cell lines,namely metastatic melanoma cell line SK-MEL2,breast cancer cell line MCF-7,and pancreatic ductal adenocarcinoma cell line Panc-1,thus recreating a suitable biological environment in *** O_(2)-sensing biomaterials can potentially measure alterations in cell metabolism caused by changes in ambient O_(2)content during drug testing/validation and tissue regeneration processes.

Habanero pepper(Capsicum chinense Jacq.)is a crop of economic relevance in the Peninsula of *** fruits have a high level of capsaicinoids compared to peppers grown in other regions of the world,which gives them industrial *** is an important factor that affects pepper development,nutritional quality,and capsaicinoid ***,the effect of soil type on fruit development and capsaicinoid metabolism has been little *** work aimed to compare the effect of soils with contrasting characteristics,black soil(BS)and red soil(RS),on the expression of genes related to the development of fruits,and capsaicinoid synthesis using a transcriptomic analysis of the habanero pepper *** growing in RS had bigger fruits and higher expression of genes related to floral development,fruit abscission,and softening which suggests that RS stimulates fruit development from early stages until maturation *** from plants growing in BS had enrichment in metabolic pathways related to growth,sugars,and ***,these fruits had higher capsaicinoid accumulation at 25 days post-anthesis,and higher expression of genes related to the branched-chain amino acids metabolism(ketol-acid reductisomerase KARI),pentose phosphate pathway and production of NADPH(glucose-6-phosphate-1-dehydrogenase G6PDH),and proteasome and vesicular traffic in cells(26S proteasome regulatory subunit T4 RPT4),which suggest that BS is better in the early stimulation of pathways related to the nutritional quality and capsaicinoid metabolism in the fruits.

Long-term monitoring reveals the effect of precipitation and silviculture on Nothofagus regeneration in Northern Patagonia mixed forests

Background Tree regeneration is a key component of resilience because it promotes post-disturbance recovery of forests. Northwestern Patagonia from Argentina is occupied by Nothofagus alpina（Na）, N. obliqua（No）, and N. dombeyi（Nd） forests that grow along an intense precipitation gradient, managed throughout shelterwood silvicultural system by technicians of the Lanin Natural Reserve. The objective of this study was to evaluate the influence of seeding cuttings over the dynamics of Nothofagus mixed forests across landscape（precipitation gradient） based mainly on the composition and abundance of tree regeneration, permanent sampling plots and generalized linear mixed models. In particular, we analysed:（i） the structure of sexual and asexual regeneration during 20 years after harvest（the dynamics of managed forests）, and（ii） the structure of sexual regeneration in primary and managed forests after > 20 years of harvest（the effect of silviculture）.Results Nd was the most abundant species in the regeneration of managed forests during all periods in both sites despite its lower contribution to the adult cohort. During the 10–20 years period after harvest, the humid site exhibited higher regeneration density than the mesic site(120,000 and 6000 ind ha-1, respectively), and it decreased afterwards. The number of established regeneration（> 2 m height） was lower for Na in the mesic site and for No in the humid site(0 and 57 ind ha-1, respectively). However, in comparison to No, Na showed a higher number of sprouted stumps and sprouts per stump, and a higher sprout height in the mesic site. No exhibited higher sprout mortality in the humid site. Finally, the regeneration of primary forests showed lower density and height, and a more balanced composition than that of managed *** The silvicultural effects on the mixed forest regeneration dynamics was strongly influenced by the condition of sites. Development and screening of(Ca-P-Si-F)-PEO coatings for biodegradability control of Mg-Zn-Ca alloys

PEO coatings were developed on MgZnxCayalloys for biomedical applications using different treatment times and a novel transparent electrolyte containing Ca,P,Si and *** characteristics and chemical composition of studied materials were evaluated using scanning electron microscopy,X-ray diffraction and optical *** investigations were performed for uncoated and coated materials using potentiodynamic polarization,electrochemical impedance spectroscopy and hydrogen evolution measurements in three media(0.9wt.%NaCl,α-MEM and inorganic part ofα-MEM).Findings revealed that Zn-rich intermetallics favour the formation of large voids in the ***,P,F and Si are successfully incorporated into the *** consist of crystalline phases(Mg O,Mg F_(2)and Ca,Si-rich compounds)and amorphous *** and organic components ofα-MEM solution accelerate corrosion of the bare substrates but have no significant influence on the corrosion resistance of coated ***,inorganicα-MEM can be considered as an appropriate medium for fast screening of PEO *** relatively small amount of F content in the barrier layer facilitates early failure of the coatings,which show undercoating ***-rich intermetallic particles in the undercoating crevice induce a more aggressive corrosive environment and enhanced micro-galvanic couple effect with the Mg *** leads to faster degradation of the coated alloys compared to the bare substrates.

Cat scratch disease(CSD),an infection caused by Bartonella henselae,is uncommon in Europe,whereas 24,000 cases per year have been reported in the United States[1,2].The typical manifestation of CSD consists of fever and subacute,solitary or regional,painful and self-limited lymphadenopathy in a patient with a history of previous scratching,biting,or contact with a kitten or cat[3].In 85%of patients,only one regional lymph node is involved,which is located in the axillary and epitrochlear regions in approximately half of the cases[4,5].One or more round,red-brown nontender papules can develop at the inoculation site or near the scratch after a few days and may persist until two to three weeks[6].
