Biomimetic manganese-based theranostic nanoplatform for cancer multimodal imaging and twofold immunotherapy

Objectives:Describe the dermoscopy and reflectance confocal microscopy(RCM) features of SGH.Methods:Thirty-one patients with SGH diagnosed according to the clinical and histopathological standards were examined using dermoscopy and RCM from March 2018 to January 2022.Results:Dermoscopically,lesions revealed yellowish-red background and faintyellow background in 25 cases(80.65%) and 6 cases(19.35%) respectively.White-yellowish lobulated structures in the center of the lesion were presented in 31 cases(100%) and umbilications in 19 patients(61.29%).Crown vessels in the peripheral of the lesions were observed in 11 cases(35.48%) while irregular linear vessels on the surface of the lesions in 18 cases(58.06%).Under the RCM,all lesions presented a honeycombed pattern in the epidermis and typical morulae-shaped sebaceous lobules in dermis.Dilated follicular infundibulum was observed in 15 cases(48.39%) and dilated vessels in 26 cases(83.87%).Conclusion:Dermoscopy and RCM enable us to describe the imaging features of SGH.Combing the two useful tools together provides a non-invasive basis for the accuracy of clinical diagnosis.

The limited clinical response and serious side effect have been challenging in cancer immunotherapy resulting from immunosuppressive tumor microenvironment(TME)and inferior drug targeting.Herein,an active targeting TME nanoplatform capable of revising the immunosuppressive TME microenvironment is designed.Briefly,gold nanorods(GNRs)are covered with silica dioxide(SiO_(2))and then coated manganese dioxide(MnO_(2))to obtain GNRs@SiO_(2)@MnO_(2)(GSM).Myeloid-derived suppressor cells(MDSCs)membrane is further camouflaged on the surface of GSM to obtain GNRs@SiO_(2)@MnO_(2)@MDSCs(GSMM).In this system,GSMM inherits active targeting TME capacity of MDSCs.The localized surface plasmon resonance of GNRs is developed in near-infraredⅡwindow by MnO_(2)layer coating,realizing NIR-Ⅱwindow photothermal imaging and photoacoustic imaging of GSMM.Based on the release of Mn^(2+)in acidic TME,GSMM can be also used for magnetic resonance imaging.In cancer cells,Mn^(2+)catalyzes H_(2)O_(2)into·OH for(chemodynamic therapy)CDT leading to activate cGAS-STING,but also directly acts on STING inducing secretion of typeⅠinterferons,pro-inflammatory cytokines and chemokines.Additionally,photothermal therapy and CDT-mediated immunogenic cell death of tumor cells can further enhance anti-tumor immunity via exposure of CRT,HMGB1 and ATP.In summary,our nanoplatform realizes multimodal cancer imaging and dual immunotherapy.

Macrophages exist in various tissues,several body cavities,and around mucosal surfaces and are a vital part of the innate immune system for host defense against many pathogens and cancers.Macrophages possess binary M1/M2 macrophage polarization settings,which perform a central role in an array of immune tasks via intrinsic signal cascades and,therefore,must be precisely regulated.Many crucial questions about macrophage signaling and immune modulation are yet to be uncovered.In addition,the clinical importance of tumor-associated macrophages is becoming more widely recognized as significant progress has been made in understanding their biology.Moreover,they are an integral part of the tumor microenvironment,playing a part in the regulation of a wide variety of processes including angiogenesis,extracellular matrix transformation,cancer cell proliferation,metastasis,immunosuppression,and resistance to chemotherapeutic and checkpoint blockade immunotherapies.Herein,we discuss immune regulation in macrophage polarization and signaling,mechanical stresses and modulation,metabolic signaling pathways,mitochondrial and transcriptional,and epigenetic regulation.Furthermore,we have broadly extended the understanding of macrophages in extracellular traps and the essential roles of autophagy and aging in regulating macrophage functions.Moreover,we discussed recent advances in macrophages-mediated immune regulation of autoimmune diseases and tumorigenesis.Lastly,we discussed targeted macrophage therapy to portray prospective targets for therapeutic strategies in health and diseases.

Mechanism of C_(4)AF Content and Heat-curing Process on the Abrasion Resistance of High Ferrite Cement

Cement used in severe maritime environments must be attached with exceptional properties such as high sulfate resistance and abrasion strength.A sulfate resistant material typically used in marine engineering is high ferrite cement,which has been utilized in sulfate-rich environments.This study aims at exploring the effect of C_(4)AF and heat-curing on the abrasion resistance of high ferrite cement(HFC,C_(4)AF:14%-22%)in order to have a comprehensive understanding of this mechanism and promote the application of HFC in marine engineering.A new invention was designed for the abrasion resistance device by considering the sea-wave abrasion and seawater erosion in laboratory.The compressive strength and abrasion resistance of HFC were measured.Additionally,advanced analytical methods were used to explore the abrasion resistance mechanism of HFC,including X-ray fluorescence(XRF),X-ray diffraction(XRD),and thermogravimetric(TG)analyses,as well as mercury intrusion porosimetry(MIP).The results showed that HFC had the best abrasion resistance under appropriate heat-curing system that the curing temperature was 50℃and the hosting time was 4 hours,compared with PI(Portland cement)and LHC(low heat cement),meanwhile the abrasion resistance of HFC had a 62.4%increase when C_(4)AF content is increased from 14%to 22%.It can be ascribed that the content of portlandite is decreased due to the increase of C_(4)AF,which can reduce the portlandite assembled in ITZ(interfacial transition zone).It can also be ascribed that the DEF(delayed ettringite formation)is successfully avoided in HFC and the hydration degree of HFC can continue to be boosted under appropriate heat-curing system.

To determine the impact of carbohydrates on the metabolic pathway in alkaliphiles, proteomes were obtained from cultures containing different carbohydrates and were resolved on two-dimensional gel electrophoresis (2-DE). The proteomes were compared to determine differentially expressed proteins. A novel alkaliphilic bacterium (alkaliphilic Bacillus sp. N16-5 isolated from Wudunur Soda Lake, China) was isolated in media with five different carbon sources (glucose, mannose, galactose, arabinose, and xylose). Comparative proteome analysis identified 61 differentially expressed proteins, which were mainly involved in carbohydrate metabolism, amino acid transport, and metabolism, as well as energy production and conversion. The comparison was based on the draft genome sequence of strain N16-5. The abundance of enzymes involved in central metabolism was significantly changed when exposed to various carbohydrates. Notably, catabolite control protein A (CcpA) was up-regulated under all carbon sources compared with glucose. In addition, pentose exhibited a stronger effect than hexose in CcpA-mediated carbon catabolite repression. These results provided a fundamental understanding of carbohydrate metabolism in alkaliphiles.

目的探讨FOCUS-PDCA程序对脑卒中患者延续性护理质量的影响。方法以即将从首都医科大学附属北京康复医院出院的脑卒中患者为研究对象,将2020年5月至2020年9月期间符合要求的50例患者作为对照组,2020年10月至2021年2月期间的50例患者为观察组,对照组进行常规延续性护理方法,观察组患者在常规护理的基础上结合FOCUS-PDCA程序干预模式。比较2组患者的MBI、FMA和满意度。结果干预后,观察组在MBI、FMA、满意度方面的得分均显著高于对照组(P<0.05)。结论将FOCUSPDCA程序应用于脑卒中延续性护理中,可显著改善患者日常生活质量,促进肢体功能恢复,提高满意度。

Solution-processed top-contact electrodes strategy for organic crystalline field-effect transistor arrays

Organic crystals,especially ultra-thin two-dimensional(2D)ones such as monolayer molecular crystals,are fragile and vulnerable to traditional vacuum deposition.Up to now,most of the methods reported for fabricating organic field-effect transistors(OFETs)with top-electrodes on the 2D molecular crystals are based on mechanical-transfer method.Nondestructive method for large scale in-situ electrode deposition is urgent.In this work,the silver mirror reaction(SMR)is introduced to construct top-contact electrodes on 2D organic crystalline thin films.OFETs based on bilayer crystalline films with solution-processed silver electrodes show comparable performance to devices with transferred gold electrodes.In addition to that,OFETs with SMR fabricated silver electrodes show lower contact resistance than the ones with evaporated silver electrodes.Furthermore,the temperature under which SMR electrodes annealed is relatively low(60℃),making this approach applicable to varies of organic semiconductors,such as spin-coated polymer films,vacuum evaporated films,2D and even monolayer crystalline films.Besides,OFETs with sub-micrometer channel width and 25μm channel length are realized which might find practical application in the ultra-small pixel mini/micro-LEDs.
