Objective: To describe the clinical outcomes of patients treated by proton beam irradiation for choroidal metastatic tumors. Design: Noncomparative case series. Participants: A retrospective chart review was performed on a series of 63 patients (76 eyes) with choroidal metastases treated with proton beam therapy between December 1989 and September 2000. methods:Patients were treated with 2 fractions of 14 cobalt gray equivalents (CGEs) (CGE=proton Gy x relative biological effectiveness 1.1), each using a nonoperative “ light- field” technique. Ophthalmologic follow- up was available for 46 patients (55 eyes), with a mean follow- up time of 10 months. The medical record or the Social Security Death Index was used to obtain survival status, which was available in 94% of cases. main Outcome measures: Tumor regression, recurrence, treatment associated complications, and visual acuity were evaluated by ophthalmologic examination and ultrasonography. Eye retention and length of survival also were assessed. Results: At the time of ocular diagnosis,49 patients reported a history of a primary cancer. median survival time after ocular diagnosis was 16 months through may 2003. most choroidal metastases were dome shaped (62% ) and located at the posterior pole (95% ). mean tumor height was approximately 3.5 mm, and serous retinal detachment was seen in 63% of cases. Eighty- four percent of treated tumors regressed completely with in 5 months of treatment, and none of these recurred. Retinal detachment resolved in 82% of patients within 3.8 months after treatment, and visual acuity was preserved or improved in 47% of the patients. Complications occurred in 56% of cases and included madarosis, keratitis, dry eye syndrome, cataract, neovascular glaucoma, chorioretinal atrophy, radiation papillopathy, and radiation maculopathy. Chemical forms and extractability of iron in sediments of three contrasting lakes of China and UK

Iron is a limiting factor for the eutrophication of lakes, especially those lakes that are enriched with phosphorus. Extractability of iron in sediments of West Lake and Taihu Lake in China and Lower Lough Erne in Northern Ireland of UK was comparatively investigated on the basis of analysing chemical forms of iron using different extractants. It was shown that extractable iron in sediments of the lakes was greatly different using various extractants. Reactive iron or easily released iron such as “active" iron oxides, total free iron oxide and water-soluble iron was not high, only accounting for 0.01%—0.15% of total iron. The efficiency of the extractants for exchangeable iron was decreased in the sequence 0.1 mol/L HCl DTPA+TEA mixed solution > 1 mol/L NH 4OAc > 0.5 mol/L mgCl 2 = 0.5 mol/L CaCl 2. It seems that the complexion of iron by organic matter was not strong because the concentration of organically bound iron was significantly lower than the concentration it was forecasted. Extractable iron is not entirely consistent with or dependent on total iron in lake sediments. To a certain extent, phosphate can inhibit the release of iron in sediments of the lakes. The selection of extractants is thus the first key step to evaluate bioavailability of iron in lake sediments.

In 2008 structural failures occurred on the asphalt pavement,with a hydraulic blast furnace slag base,of the motorway A32 (constructed in 1986～1988) in the Netherlands. Within a few weeks serious depressions occurred,in which the asphalt pavement was full of cracks and the base had a very low stiffness. In 2008 and 2009 the pavement including the base was rehabilitated over a length of 10 *** Dutch ministry of Public Works has initiated an extensive research into the risks of occurrence of this type of unexpected structural damages on the main road network in the Netherlands. Among many other things this research includes material research into the base material of the A32 to find the cause of the structural *** total 130 cores,divided over 26 cross-sections,were drilled from the A32. The cross-sections were divided over 3 areas,with a high and low base stiffness respectively and also at heaves that had grown into the pavement since 1998. In areas with high base stiffness the cores mainly contained bound base material,in areas with low base stiffness mainly granular material and at the heaves it was a mix thereof. Standard (static) and dynamic crushing tests were done on the granular material. Dynamic and monotonic indirect tensile tests and compression tests were performed on the bound material. In all cases no systematic difference was found between the mechanical properties of the blast furnace slag base material from the heavily loaded right traffic lane and the material from the other *** is concluded that the pavement failure on the A32 is introduced by the occurrence of full depth asphalt fatigue cracks in the pavement that was originally designed for 12 years (but was never strengthened) in areas where the base stiffness always has been low or became low through crushing (that also has resulted in the heaves). Processing Pisum sativum seed storage protein precursors in vitro

The profile of polypeptides separated by SDS-PAGE from seed of major crop species such as pea (Pisum sativum) is complex, resulting from cleavage (processing) of precursors expressed from multiple copies of genes encoding vicilin and legumin, the major storage globulins. Translation in vitro of mRNAs hybridselected from mid-maturation pea seed RNAs by defined vicilin and legumin cDNA clones provided precursor molecules that were cleaved in vitro by a cell-free protease extract obtained from similar stage seed; the derived polypeptides were of comparable sizes to those observed in vivo. The feasibility of transcribing mRNA in vitro from a cDNA clone and cleavage in vitro of the derived translation products was established for a legumin clone,providing a method for determining polypeptide products of an expressed sequence. This approach will also be useful for characterising cleavage site requirements since modifications an readily be introduced at the DNA level.

Selenium supplementation in a population with low basal blood selenium levels has been reported to decrease theincidence of several cancers including prostate *** on the clinical findings,it is likely that the antioxidantfunction of one or more selenoproteins is responsible for the chemopreventive effect,although low molecular weightseleno-compounds have also been posited to selectively induce apoptosis in transformed *** address the effects ofselenium supplementation on selenoprotein expression in prostate cells,we have undertaken an analysis of antioxidantselenoprotein expression as well as selenium toxicity in non-tumorigenic prostate epithelial cells(RWPE-1)and prostatecancer cells(LNCaP and PC-3).Our results show that two of the glutathione peroxidase family members(GPX1 andGPX4)are highly induced by supplemental selenium in prostate cancer cells but only slightly induced in RWPE-1 *** addition,GPX1 levels are dramatically lower in PC-3 cells as compared to RWPE-1 or LNCaP ***2 proteinand mRNA,however,are only detectable in RWPE-1 *** the three selenium compounds tested(sodium selenite,sodium selenate and selenomethionine),only sodium selenite shows toxicity in a physiological range of *** and in contrast to previous studies,RWPE-1 cells were significantly more sensitive to selenitethan either of the prostate cancer cell *** results demonstrate that selenoproteins and selenium metabolism areregulated at multiple levels in prostate cells.

＜正＞ Liver spheroid formation is a process to culture single cells into three-dimensional spherical structures. There are several factors such as gyrotatory speed of the cell shaker, cell density, volume of medium etc that are key criteria in spheroid formation. Traditional approaches have been adopted to undertake experiments to determine optimal conditions for spheroid studies. If the process of spheroid formation can be fitted to a mathematical model, it will be helpful to improve spheroid culture by computerized simulation. This insight provides a strong indication of the necessity for modelling of the spheroid development behaviour, with systematic descriptions of the whole dynamic process. It is considered that this type of computational biology can offer a significant contribution to experimental design, prediction of culture progression, and so on. This is relevant to furthering our understanding of the dynamics of spheroid culture relevant to scale-up of production to meet demand for spheroids in applied research.
