In an attempt to reconcile wood extraction and forest biodiversity in managed boreal forests,ecosystem-based forest management(EBM)has become the de facto management *** forestry represents one prominent way that EBM is implemented in many parts of the *** patches commonly left after harvesting serve as analogues of fire island remnants,which are patches of unburned forests in the burned forest *** the persistence of retention patches has been questioned,few studies have attempted to quantitatively compare forest attributes in both burned and harvested *** part of a larger program examining multiple aspects of ecosystem function in fire and harvest island remnants,we investigated the impact of disturbance type(fire/harvest)and forest edges on c stock in snags and coarse woody debris(cWD)found in island remnants in mixedwood boreal forests of Alberta,*** c stock(in snags and cWD)was similar between the two disturbance types and edge plots had similar total deadwood c stocks to *** edges of island remnants had about two-fold more snag c stock than their interiors in both disturbance types,but c stock in cWD was unaffected by edge effects and disturbance *** results suggest that deadwood c dynamics in island remnants in fire and harvest disturbed boreal forests were similar,thus lending support for the continued implementation of retention forestry in Alberta.

We analyzed national forest inventory data collected from circa 2000–2018 across 37 states in the eastern United States to better understand the influence of forest health related canopy disturbances on the regeneration and recruitment dynamics of upland oaks(Quercus).We found low levels of oak recruitment across all disturbance types examined but limited evidence of any direct effects from the type of disturbance on the population of regenerating *** general lack of differences in oak regeneration response between forest health disturbances and disturbances caused by harvested or non-disturbed plots does not indicate that the effects of forest health disturbances were benign,***,low level of oak recruitment across all disturbance types highlights the pervasiveness of the trend of shifting composition in once oak-dominated forests where oak is absent or sparse in the regeneration *** results show that oak recruitment was higher when oak was present as advance reproduction prior to disturbance from any cause ***,these results lead us to conclude that the widespread inadequacy of oak advance reproduction in mature oak-dominated forests is the prevailing threat to oak forest health and *** suggest the status of advance reproduction be treated as a co-morbidity when weighing the risk and potential outcomes from other threats to upland oak forests in the eastern United States.

Bioinformatic analysis of large and complex omics datasets has become increasingly useful in modern day biology by providing a great depth of information,with its application to neuroscience termed *** mining of omics datasets has enabled the generation of new hypotheses based on differentially regulated biological molecules associated with disease mechanisms,which can be tested experimentally for improved diagnostic and therapeutic targeting of neurodegenerative ***,integrating multi-omics data using a systems bioinformatics approach will advance the understanding of the layered and interactive network of biological regulation that exchanges systemic knowledge to facilitate the development of a comprehensive human brain *** this review,we first summarize data mining studies utilizing datasets from the individual type of omics analysis,including epigenetics/epigenomics,transcriptomics,proteomics,metabolomics,lipidomics,and spatial omics,pertaining to Alzheimer's disease,Parkinson's disease,and multiple *** then discuss multi-omics integration approaches,including independent biological integration and unsupervised integration methods,for more intuitive and informative interpretation of the biological data obtained across different omics *** further assess studies that integrate multi-omics in data mining which provide convoluted biological insights and offer proof-of-concept proposition towards systems bioinformatics in the reconstruction of brain ***,we recommend a combination of high dimensional bioinformatics analysis with experimental validation to achieve translational neuroscience applications including biomarker discovery,therapeutic development,and elucidation of disease *** conclude by providing future perspectives and opportunities in applying integrative multi-omics and systems bioinformatics to achieve precision phenotyping of neurodegenerative diseases and towards personaliz

Bone metastasis of non-seminomatous germ cell tumors(NSGcT)of the testes is a rare event and even more uncommon at initial ***,bone lesions are discovered in the presence of concurrent retroperitoneal lymph node or visceral ***,in this case,a 37 years old male complaining of a growing testicular mass was found to have isolated bone metastasis with associated caudaequina syndrome without apparent abnormal findings on initial computed tomography(cT)*** neurologic symptoms prompted further evaluation with magnetic resonance imaging(MRI),which demonstrated multiple sites of bone metastasis without evidence of retroperitoneal lymph node or visceral organ *** case represents a rare clinical presentation and disease manifestation of NSGcT.

cRISPR/cas-mediated genome editing in human pluripotent stem cells(hPScs)offers unprecedented opportunities for developing in vitro disease modeling,drug screening and cell-based *** efficiently deliver the cRISPR components,here we developed two all-in-one vectors containing cas9/gRNA and inducible cas13d/gRNA cassettes for robust genome editing and RNA interference *** vectors utilized the PiggyBac transposon system,which allows stable expression of cRISPR components in *** cas9 vector PB-cRISPR exhibited high efficiency(up to 99%)of inducing gene knockout in both protein-coding genes and long non-coding *** other inducible cas13d vector achieved extremely high efficiency in RNA knockdown(98%knockdown for cD90)with optimized gRNA *** together,our PiggyBac cRISPR vectors can serve as powerful toolkits for studying gene functions in hPScs.

The molecular basis for enhanced chemosensitivity of testicular germ cell tumors （GcT） has been an area of great interest, as it could potentially give us therapeutic leads in other resistant malignancies. Thus far, however, the increased sensitivity of c＆T has been variously attributed to multiple factors -- an inability to detoxify cisplatin, a lack of export pumps, an inability to repair the DNA damage, an intact apoptotic cascade and lack of p53 mutation; but a unifying underlying etiology leading to the aforementioned processes and having a translational implication has so far been elusive. Herein, we offer evidence to support a potential significant role for the previously demonstrated low hypoxia inducible factor-la （HIF-la） expression in mediating the general exquisite chemosensitivity of testicular GcT, through the aforementioned processes. This molecular mechanism based hypothesis could have a significant translational implication in platinum refractory GcT as well as other platinum resistant malignancies.
