Effect of an Improved Mechanical Method for Assisted Hatching on the in vitro Development of Mouse Embryos

Objective To evaluate the safety and efficiency of an improved shape of opening for mechanical assisted hatching （AH） on the in virto development of mouse embryos. Methods A total of 622 KM BAI mouse embryos in 2-cell-4-cell stage were randomly divided into group A, group B and control group. A new mechanical AH method by improving the shape of opening in the ZP was used in group A, and a ＂-/ ＂-shaped opening was created. A ＂＋＂ -shaped opening was made in group B, while no opening was made in control group. Comparisons have been made among the three groups with regard to the duration of AH, the blastocyst formation and complete hatching rate, etc. Results The duration of AH in group A （43.25 ±3.46 s） was significantly shorter than that in group B （52.81 ±4.32 s, P 〈0.05）. The blastocyst formation rate on d 5 was not significantly different among the three groups （92.27%, 93.66% and 94.92% respectively, P 〉0.05）. The complete hatching rate of blastocysts on d 6 between group A and group B was no statistical difference （94.09% vs 92.71%, P 〉0.05）, but significantly higher than that in control group （43.32%, P 〈0.001）. No significant difference in the percentage of grade 1 blastocysts was found among the three groups on d 5 （85.22%, 82.81% and 86.63% respectively, P 〉0.05）. Conclusion R could enhance the process of embryo hatching and facilitate the hatching rate of blastocysts by using the improved mechanical AH method, which is of safety and efficiency to mouse embryo in the in vitro development.

The combination of photodynamic therapy（PDT） and chemodynamic therapy（CDT） in tumor treatment has attracted considerable ***,tumor hypoxia and glutathione（GSH） overproduction in the tumor tissue restricted the progress of their ***,a Mn-porphyrinic metal-organic framework（Mn-TCPP） was constructed by the one-pot method and further used for immobilizing glucose oxidase（GOx） to obtain GOx@*** would rapidly exhaust endogenous glucose into hydrogen peroxide（H2O2） and gluconic acid,thus shutting off the energy supply of tumor cells for starvation ***-TCPP catalyzed H2O2to produce oxygen,regulating the hypoxic tumor microenvironment and in turn improving1O2generation under laser ***,Mn-TCPP can reduce reactive oxygen species consumption owing to the redox reaction between Mn3+and GSH,thus greatly enhancing ***,benefiting from GOx-mediated starvation therapy,Mn2+produced by Mn3+reduction can react with sufficient intracellular H2O2to generate ·OH with high cytotoxicity through a Fenton-like *** treatment by GOx@Mn-TCPP under laser irradiation in vitro,the cell viability of 4T1 and A549 tumor cells reached to 20%,reflecting GOx@Mn-TCPP could give full play to the advantages of PDT/CDT/starvation *** results in vivo demonstrated that GOx@Mn-TCPP mediated synergistic cascade therapy could significantly inhibit tumor growth and improve the therapeutic effect.

本文对青岛地区苜蓿(Medicago sativa)田节肢动物群落种类、优势种田间种群动态进行了调查,分析了优势种群的田间动态规律,对主要害虫与优势天敌的相关性进行了研究。结果表明:青岛地区苜蓿田有节肢动物13目49科91种,其中主要害虫有蓟马(thrips)、蚜虫(aphids)、盲蝽(mirids)、苜蓿斑螟(Salebria semirubella)等,主要天敌有瓢虫(ladybirds)、食蚜蝇(syrphids)、寄生蜂(parasitic wasps)、草蛉(lacewings)、蜘蛛(spiders)等。蚜虫每年发生有2个高峰期,分别为5月中旬和10月下旬;蓟马在田间有2个发生高峰期,分别是7月上旬和10月上旬;苜蓿斑螟有1个高峰期是在9月下旬;盲蝽有3个高峰,分别是7月中旬、8月上旬和9月下旬。瓢虫类在田间有2个高峰期,分别是5月中旬和10月中旬;寄生蜂类有2个主要高峰期,分别是4月中旬和5月下旬;食蚜蝇类有3个高峰,分别是4月中旬、6月上旬和10月中旬;蜘蛛有5个发生高峰期,分别是5月中旬、6月下旬、8月上旬、9月上旬和10月上旬。应用灰色关联度分析方法和一元回归法探讨了天敌与害虫之间的关系,初步明确了对每类害虫有控制作用的天敌种类。

A mathematical model for the dynamics of a prey-dependent consumption model concerning integrated pest management is proposed and analyzed. We show that there exists a globally stable pesteradication periodic solution when the impulsive period is less than some critical values. Furthermore, the conditions for the permanence of the system are given. By using bifurcation theory, we show the existence of a nontrival periodic solution if the pest-eradication periodic solution loses its stability. When the unique positive periodic solution loses its stability, numerical simulation shows there is a characteristic sequence of bifurcations, leading to a chaotic dynamics, which implies that dynamical behaviors of prey-dependent consumption concerning integrated pest management are very complex, including period-doubling cascades, chaotic bands with periodic windows, crises, symmetry-breaking bifurcations and supertransients.

By using a self-made carbon/polytetrafluoroethylene （C/PTFE） O2-fed as cathode and Ti/IrO2/RuO2 as anode, the effects of electrochemical oxidation of phenol and the coal-gas wastewater containing phenol were studied. The terylene diaphragm which kept pH〉12 in cathodic compartment and pH〈1 in the anodic compartment was selected in the experiment in comparison with the other types of diaphragm. Furthermore, hydroxyl radical （HO·） was determined in the cathodic compartment of the diaphragm cell by electron spin resonance spectrum（ESR） and the fluorescence spectra. Compared with pH, the accumulated HzO2 and the COD removal of the no-diaphragm cell, the mechanism of electrochemical oxidation in the terylene diaphragm cell was supposed. The degradation of phenol was supposed to be cooperative oxidation by direct or indirect electrochemical oxidation at the anode and H2O2, HO· produced by oxygen reduction at the cathode. The mineralization of phenol in the diaphragm cell was better than that in the no-diaphragm cell. When the coal-gas wastewater was treated by the electrolysis system with terylene diaphragm, the average removal efficiency of the volatile phenol and COD were 100% and 79.6%, respectively.

Segment Routing(SR)is a new routing paradigm based on source routing and provide traffic engineering(TE)capabilities in IP *** extending interior gateway protocol(IGP),SR can be easily applied to IP ***,upgrading current IP network to a full SR one can be costly and *** IP/SR network will last for some *** at the low flexibility problem of static TE policies in the current SR networks,this paper proposes a Deep Reinforcement Learning(DRL)based TE *** proposed scheme employs multi-path transmission and use DRL to dynamically adjust the traffic splitting ratio among different paths based on the network traffic *** a result,the network congestion can be mitigated and the performance of the network is *** results show that our proposed scheme can improve the throughput of the network by up to 9%than existing schemes.

Heterogeneous computing is one effective method of high performance computing with many advantages. Task scheduling is a critical issue in heterogeneous environments as well as in homogeneous environments. A number of task scheduling algorithms for homogeneous environments have been proposed, whereas, a few for heterogeneous environments can be found in the literature. A novel task scheduling algorithm for heterogeneous environments, called the heterogeneous critical task （HCT） scheduling algorithm is presented. By means of the directed acyclic graph and the gantt graph, the HCT algorithm defines the critical task and the idle time slot. After determining the critical tasks of a given task, the HCT algorithm tentatively duplicates the critical tasks onto the processor that has the given task in the idle time slot, to reduce the start time of the given task. To compare the performance of the HCT algorithm with several recently proposed algorithms, a large set of randomly generated applications and the Gaussian elimination application are randomly generated. The experimental result has shown that the HCT algorithm outperforms the other algorithm.

Estimating distribution of water uptake with depth of winter wheat by hydrogen and oxygen stable isotopes under different irrigation depths

Crop root system plays an important role in the water cycle of the soil-plant-atmosphere continuum. In this study, com- bined isotope techniques, root length density and root cell activity analysis were used to investigate the root water uptake mechanisms of winter wheat （Triticum aesfivum L.） under different irrigation depths in the North China Plain. Both direct inference approach and multisource linear mixing model were applied to estimate the distribution of water uptake with depth in six growing stages. Results showed that winter wheat under land surface irrigation treatment （Ts） mainly absorbed water from 10-20 cm soil layers in the wintering and green stages （66.9 and 72.0%, respectively）; 0-20 cm （57.0%） in the jointing stage; 0-40 （15.3%） and 80-180 cm （58.1%） in the heading stage; 60-80 （13.2%） and 180-220 cm （35.5%） in the filling stage; and 0-40 （46.8%） and 80-100 cm （31.0%） in the ripening stage. Winter wheat under whole soil layers irrigation treatment （Tw） absorbed more water from deep soil layer than Ts in heading, filling and ripening stages. Moreover, root cell activity and root length density of winter wheat under TW were significantly greater than that of Ts in the three stages. We concluded that distribution of water uptake with depth was affected by the availability of water sources, the root length density and root cell activity. Implementation of the whole soil layers irrigation method can affect root system distribution and thereby increase water use from deeper soil and enhance water use efficiency.

Soybean cyst nematode （SCN） is one of the most devastating pathogen for soybean. Therefore, identiifcation of resistant germplasm resources and resistant genes is needed to improve SCN resistance for soybean. Soybean varieties Huipizhiheidou and Wuzhaiheidou were distributed in China and exhibited broad spectrums of resistance to various SCN races. In this study, these two resistant varieties, combined with standard susceptible varieties （Lee and Essex）, were utilized to identify the differentially expressed transcripts after infection with SCN race 4 between resistant and susceptible reactions by using the Affymetrix Soybean Genome GeneChip. Comparative analyses indicated that 21 common genes changed signiifcantly in the resistant group, of which 16 increased and 5 decreased. However, 12 common genes changed signiifcantly in the susceptible group, of which 9 increased and 3 decreased. Additionally, 27 genes were found in common between resistant and susceptible reactions. The 21 signiifcantly changed genes in resistant reaction were associated with disease and defense, cell structure, transcription, metabolism, and signal transduction. The fold induction of 4 from the 21 genes was conifrmed by quantitative RT-PCR （qRT-PCR） analysis. Moreover, the gene ontology （GO） enrichment analyses demonstrated the serine family amino acid metabolic process and arginine metabolic process may play important roles in SCN resistance. This study provided a new insight on the genetic basis of soybean resistance to SCN race 4, and the identiifed resistant or resistant-related genes are potentially useful for SCN-resistance breeding in soybean.
