MicroRNA Expression Profiles and MiR-10a Target in Anti-benzo[a] pyrene-7,8-diol-9,10-epoxide-transformed Human 16HBE Cells

Objective To screen miRNA profiles of malignantly transformed human bronchial epithelial cells, 16HBE-T, induced by anti-benzo[a]pyrene-trans-7,8-diol-9,10-epoxide （anti-BPDE）, and to analyze putative miR-10a targets in 16HBE-T. Methods A novel microarray platform was employed to screen miRNA profiles of 16HBE-T cells transformed by anti-BPDE. Microarray data for miR-10a and miR-320 were validated using quantitative real time polymerase chain reaction （QRT-PCR）. The expression of a putative target for miR-10a, HOXA1, was analyzed by reverse transcription polymerase chain reaction （RT-PCR） and QRT-PCR. Results In comparison with the vehicle-treated cells （16HBE-N）, 16HBE-T exhibited differential expression of 54 miRNAs, in which, 45 were over-expressed and 9 were down-regulated. The five most highly expressed miRNAs were miR-494, miR-320, miR-498, miR-129, and miR-106a. The lowest expressed miRNAs were miR-10a, miR-493-5p, and miR-363＊. Three members of miR-17-92 cluster, miR-17-5p, miR-20a, and miR-92, showed significantly higher abundance in 16BHE-T as miR-21, miR-141, miR-27a, miR-27b, miR-16 and miRNAs of the let-7 family. The putative target for miR-10a, HOXA1 mRNA was up-regulated 3-9-fold in 16HBE-T, as compared with 16HBE-N. Conclusion The findings of the study provide information on differentially expressed miRNA in malignant 16HBE-T, and also suggest a potential role of these miRNAs in cell transformation induced by anti-BPDE. HOXA1 is similarly up-regulated, suggesting that miR-10a is associated with the process of HOXA 1-mediated transformation.

Spontaneous emission of a moving atom in a waveguide of rectangular cross section

We study the spontaneous emission(SE) of an excited nonrelativistic two-level system(TLS) interacting with the vacuum in a waveguide of rectangular cross section. All TLS’s transitions and the center-of-mass motion of the TLS are taken into account. The SE rate and the carried frequency of the emitted photon for the TLS initially being at rest are obtained, it is found that in the first order of the mass M, the frequency of the emitted photon is smaller than the transition frequency of the TLS and the SE rate is smaller than the SE rate Γfof the TLS fixed in the same waveguide. The SE rate for the TLS initially being moving is obtained in the second order of the mass M. The SE rate is smaller than Γfbut it is dependent not only on the atomic mass but also on the initial momentum. The carried frequency of the emitted photon is decreased when it travels along the direction of the initial momentum, whereas it is increased when it travels in the opposite direction of the initial momentum.

Objective:To investigate the effect of Huangqin Decoction(HQD)on nuclear factor erythroid 2 related-factor 2(Nrf2)/heme oxygenase(HO-1)signaling pathway by inducing the colitis-associated carcinogenesis(CAC)model mice with azoxymethane(AOM)/dextran sodium sulfate(DSS).Methods:The chemical components of HQD were analyzed by liquid chromatography-quadrupole-time-of-flight mass spectrometry(LC-Q-TOF-MS/MS)to determine the molecular constituents of *** 48 C57BL/6J mice were randomly divided into 6 groups by a random number table,including control,model(AOM/DSS),mesalazine(MS),low-,medium-,and high-dose HQD(HQD-L,HQD-M,and HQD-H)groups,8 mice in each *** for the control group,the mice in the other groups were intraperitoneally injected with AOM(10 mg/kg)and administrated with 2.5%DSS orally for 1 week every two weeks(totally 3 rounds of DSS)to construct a colitis-associated carcinogenesis mouse *** mice in the HQD-L,HQD-M and HQD-H groups were given HQD by gavage at doses of 2.925,5.85,and 11.7 g/kg,respectively;the mice in the MS group was given a suspension of MS at a dose of 0.043 g/kg(totally 11 weeks).The serum levels of malondialdehyde(MDA)and superoxide dismutase(SOD)were measured by enzyme-linked immunosorbent *** mRNA and protein expression levels of Nrf2,HO-1,and inhibitory KELCH like ECH-related protein 1(Keap1)in colon tissue were detected by quantitative real-time PCR,immunohistochemistry,and Westem blot,***:LC-Q-TOF-MS/MS analysis revealed that the chemical constituents of HQD include baicalin,paeoniflorin,and glycyrrhizic *** to the control group,significantly higher MDA levels and lower SOD levels were observed in the model group(P<0.05),whereas the expressions of Nrf2 and HO-1 were significantly decreased,and the expression of Keap1 increased(P<0.01).Compared with the model group,serum MDA level was decreased and SOD level was increased in the HQD-M,HQD-H and MS groups(P<0.05).Higher expressions of

Application of neoadjuvant chemoradiotherapy and neoadjuvant chemotherapy in curative surgery for esophageal cancer:A metaanalysis

BACKGROUND The effectiveness of neoadjuvant therapy in esophageal cancer(EC)treatment is still a subject of *** To compare the clinical efficacy and toxic side effects between neoadjuvant chemoradiotherapy(nCRT)and neoadjuvant chemotherapy(nCT)for locally advanced EC(LAEC).METHODS A comprehensive search was conducted using multiple databases,including PubMed,EMBASE,MEDLINE,Science Direct,The Cochrane Library,China National Knowledge Infrastructure,Wanfang Database,Chinese Science and Technology Journal Database,and Chinese Biomedical Literature Database *** up to December 2022 comparing nCRT and nCT in patients with EC were *** The analysis revealed significant differences between nCRT and nCT in terms of disease-free *** results indicated that nCRT provided better outcomes in terms of the 3-year overall survival rate(OSR)[odds ratio(OR)=0.95],complete response rate(OR=3.15),and R0 clearance rate(CR)(OR=2.25).However,nCT demonstrated a better 5-year OSR(OR=1.02)than ***,when compared to nCRT,nCT showed reduced risks of cardiac complications(OR=1.15)and pulmonary complications(OR=1.30).CONCLUSION Overall,both nCRT and nCT were effective in terms of survival outcomes for ***,nCT exhibited better performance in terms of postoperative complications.

A Scalable Pillar Layered Metal-Organic Framework for Natural Gas Purification

One-step harvest of high-purity methane(CH_(4))from ternary propane/ethane/methane(C_(3)H_(8)/C_(2)H_(6)/CH_(4))mixtures remains a desirable yet challenging goal for natural gas ***,adsorbents either suffer from high capacity and selectivity,or are caught in a dilemma of scalable ***,we demonstrate a scalable pillar layered metal-organic framework Ni-MOF for highly efficient one-step CH_(4)***-MOF exhibits high C_(2)H_(6)and C_(3)H_(8)uptakes of 83.3 and 86.1 cm^(3)/g at 298 K and 100 kPa and remarkable C_(2)H_(6)/CH_(4)(50/50,volume ratio,21.5)and C_(3)H_(8)/CH_(4)(50/50,volume ratio,212.0)***,high C_(2)H_(6)(42.2 cm^(3)/g at 10 kPa)and C_(3)H_(8)(64.7 cm^(3)/g at 5 kPa)capacities in the low-pressure region at 298 K were realized on Ni-MOF,suggesting the strong affinities of Ni-MOF towards C_(2)H_(6)and C_(3)H_(8).Furthermore,the dynamic breakthrough experiments revealed that purifying CH_(4)from natural gas in one-step can be achieved in Ni-MOF with high-purity(>99.8%)and productivity(346.0 cm^(3)/g).Most significantly,the production of Ni-MOF can be scalably synthesized at room temperature,rendering it promising potential for industrial *** combined advantages of exceptional separation performance,scalability,and cycle stability of Ni-MOF pave the way for one-step CH_(4)purification from natural gas.
