RCAN family member 3 deficiency contributes to noncompaction of the ventricular myocardium

Noncompaction of the ventricular myocardium(NVM),the third most diagnosed cardiomyopathy,is characterized by prominent trabeculae and intratrabecular recesses.However,the genetic etiology of 40%–60%of NVM cases remains unknown.Here,we identify two infants with NVM,in a nonconsanguineous family,with a typical clinical presentation of persistent bradycardia since the prenatal period.A homozygous missense variant(R223L)of RCAN family member 3(RCAN3)is detected in both infants using whole-exome sequencing.In the zebrafish model,marked cardiac dysfunction is detected in rcan3 deficiency(MO-rcan3ATG-injected)and rcan−/−embryos.Developmental dysplasia of both endocardial and myocardial layers is also detected in rcan3-deficient embryos.RCAN3 R223L variant mRNAs can not rescue heart defects caused by rcan3 knockdown or knockout;however,hRCAN3 mRNAs rescue these phenotypes.RNA-seq experiments show that several genes involved in cardiomyopathies are significantly regulated through multiple signaling pathways in the rcan3-knockdown zebrafish model.In human cardiomyocytes,RCAN3 deficiency results in reduced proliferation and increased apoptosis,together with an abnormal mitochondrial ultrastructure.Thus,we suggest that RCAN3 is a susceptibility gene for cardiomyopathies,especially NVM and that the R223L mutation is a potential loss-of-function variant.

Lipase and photodecarboxylase coexpression: A potential strategy for alkane-based biodiesel production from natural triglycerides

Alkane-based biodiesel is considered the next generation of biodiesel owing to its potential environmental benefits and the fact that it exhibits much higher specific caloric values than traditional biodiesel.However,the formidable obstacle impeding the commercialization of this cutting-edge fuel alternative lies in the cost associated with its production.In this study,an engineered strain Escherichia coli(E.coli)showcasing harmonized coexpression of a lipase(from Thermomyces lanuginosus lipase,TLL)and a fatty acid photodecarboxylase(from Chlorella variabilis,CvFAP)was first constructed to transform triglycerides into alkanes.The potential of E.coli BL21(DE3)/pRSFDuet-1-TLL-CvFAP for alkane synthesis was evaluated with tripalmitin as a model substrate under various process conditions.Following a comprehensive examination of the reaction parameters,the scope of the biotransformation was expanded to‘real’substrates(vegetable oils).The results showed that bioderived oils can be transformed into alkanes with high yields(0.80-10.20 mmol·L^(-1))under mild conditions(35℃,pH 8.0,and 36 h)and blue light illumination.The selected processes were performed on an increased lab scale(up to 100 ml)with up to 24.77 mmol·L^(-1) tripalmitin,leading to a yield of 18.89 mmol·L^(-1) pentadecane.With the employment of a method for efficiently producing alkanes under mild conditions and a simple procedure to isolate alkanes from the reaction system,the utilization of sustainable biomass as a fundamental feedstock emerges as the primary solution to lower the cost of alkane-based biodiesel.Thus,this study proposes a readily implementable and highly effective approach for alkane-based biodiesel production.

A CombustionModel for Explosive Charge Affected by a Bottom Gap in the Launch Environment

Launch safety of explosive charges has become an urgent problem to be solved by all countries in the world aslaunch situation of ammunition becomes consistentlyworse.However, the existing numericalmodels have differentdefects. This paper formulates an efficient computational model of the combustion of an explosive charge affectedby a bottom gap in the launch environment in the context of the material point method. The current temperatureis computed accurately from the heat balance equation, and different physical states of the explosive charges areconsidered through various equations of state. Microcracks in the explosive charges are described with respectto the viscoelastic statistical crackmechanics (Visco–SCRAM) model. Themethod for calculating the temperatureat the bottomof the explosive charge with respect to the bottomgap is described. Based on this combustionmodel,the temperature history of a Composition B (COMB) explosive charge in the presence of a bottom gap is obtainedduring the launch process of a 155-mm artillery. The simulation results show that the bottom gap thickness shouldbe no greater than 0.039 cm to ensure the safety of the COM B explosive charge in the launch environment. Thisconclusion is consistent with previous results and verifies the correctness of the proposed model. Ultimately, thispaper derives amathematical expression for themaximumtemperature of the COMB explosive chargewith respectto the bottomgap thickness (over the range of 0.00–0.039 cm), and establishes a quantitative evaluationmethod forthe launch safety of explosive charges.The research results provide some guidance for the assessment and detectionof explosive charge safety in complex launch environments.

Link Prediction Method Fusion with Local Structural Entropy for Directed Network

Link prediction utilizes accessible network information to complement or predict the network links.Similarity is an important prerequisite for link prediction which means links more likely occurs between two similar nodes. Existing methods utilize the similarity of nodes but neglect of network structure. However the link direction leads to a far more complex structure and contains more information useful than the undirected networks. Most classic methods are difficult to depict the distribution of the network structure with incidental direction so the similarity characteristics of the network structure itself are lost. In this respect, a new method of local structure entropy is proposed to depict the directed structural distribution characteristics, which can be used to evaluate the degree of local structural similarity of nodes and then applied to link prediction methods. Experimental results on 8 real directed networks show that this method is effective for both area under the receiver operating characteristic curve(AUC)and ranking-score measures, and improved predictive capacity of the baseline methodology.

Proteomics analysis of coronary atherosclerotic heart disease with different Traditional Chinese Medicine syndrome types before and after percutaneous coronary intervention

OBJECTIVE: To investigate the underlying protein molecular mechanisms of "Qi stagnation and blood stasis syndrome"(QS) and "Qi deficiency and blood stasis syndrome"(QD), as two subtypes of coronary artery disease(CAD) in Traditional Chinese Medicine(TCM),following percutaneous coronary intervention(PCI).METHODS: In this study, a total of 227 CAD patients with QS and 211 CAD patients with QD were enrolled; all participants underwent PCI. Label-free quantification proteomics were employed to analyze the changes in serum in two subtypes of CAD patients before and 6 months after PCI, aiming to elucidate the intervention mechanism of PCI in treating CAD characterized by two different TCM syndromes.RESULTS: Biochemical analysis revealed significant changes in tumor necrosis factor-α, high density lipoprotein cholesterol, blood stasis clinical symptoms observation, and Gensini levels in both patient groups post-PCI; Proteomic analysis identified 79 and 95 differentially expressed proteins in the QS and QD patient groups, respectively, compared to their control groups.complement C8 alpha chain, complement factor H,apolipoprotein H, apolipoprotein B, plasminogen,carbonic anhydrase 2, and complement factor Ⅰ were altered in both comparison groups. Furthermore,enrichment analysis demonstrated that cell adhesion and connectivity-related processes underwent changes in QS patients post-PCI, whereas lipid metabolism-related pathways, including the peroxisome proliferator-activated receptor signaling pathway and extracellular matrix receptor interaction, underwent changes in the QD group.The protein-protein interaction network analysis further enriched 52 node proteins, including apolipoprotein B,lipoprotein(a), complement C5, apolipoprotein A4,complement C8 alpha chain, complement C8 beta chain,complement C8 gamma chain, apolipoprotein H,apolipoprotein A-Ⅱ, albumin, complement C4-B,apolipoprotein C3, among others. Biliary dyskinesia is a relatively common gastrointestinal disease that is increas-ing in incidence as living standards improve.However,its underlying pathogenesis remains unclear,hindering the development of therapeutic drugs.Recently,“Expression and functional study of cholecystokinin-A receptors on the interstitial Cajal-like cells of the guinea pig common bile duct”demonstrated that cholecystokinin(CCK)regulates the contractile function of the common bile duct through interaction with the CCK-A receptor in interstitial Cajal-like cells,contributing to improving the academic understanding of biliary tract dynamics and providing emerging directions for the pathogenesis and clinical management of biliary dyskinesia.This letter provides a brief overview of the role of CCK and CCK-A receptors in biliary dyskinesia from the perspective of animal experiments and clinical studies,and discusses prospects and challenges for the clinical application of CCK and CCK-A receptors as potential therapeutic targets.

Impacts of electrochemical disinfection on the viability and structure of the microbiome in secondary effluent water

Electrochemical disinfection(ECD)is a promising disinfection technique for wastewater reclamation;however,the impacts of ECD on the microbiome in secondary effluent wastewater remain unknown.In this study,Propidium monoazide-qPCR(PMA-qPCR)and the plate count method were used to evaluate the inactivation performance,and the PMA-16S rRNA gene sequences of living cells were targeted to study the microbiome.A discrepancy was found between PMA-qPCR and the plate count method in the evaluation of cell count,with increases of 1.5 to 2.2 orders of magnitude in the disinfection rate after 150 s of disinfection.However,the cell count recovered and occasionally exceeded original levels within 3 d after disinfection.Biodiversity was suppressed after ECD,but the microbiome after 150 s disinfection retained a higher level of evenness and stability in the community with a median Shannon index(>3.7).Pathogenic bacteria remained high in relative abundance even after 150 s of 25 V disinfection,but the biofilm-forming population was effectively suppressed by ECD.The co-occurrence network revealed a centralized and fragile network as disinfection persisted,demonstrating the destabilizing effects of ECD on the microbiome.Functional pathways for cell membrane synthesis and organic compound degradation were enriched after ECD.The reaction of the microbiome after ECD was similar to other disinfection techniques in terms of community structure.

Failure evaluation and control factor analysis of slope block instability along traffic corridor in Southeastern Tibet

The instability of slope blocks occurred frequently along traffic corridor in Southeastern Tibet(TCST),which was primarily controlled by the rock mass structures.A rapid method evaluating the control effects of rock mass structures was proposed through field statistics of the slopes and rock mass structures along TCST,which combined the stereographic projection method,modified M-JCS model,and limit equilibrium theory.The instabilities of slope blocks along TCST were then evaluated rapidly,and the different control factors of instability were analyzed.Results showed that the probabilities of toppling(5.31%),planar(16.15%),and wedge(35.37%)failure of slope blocks along TCST increased sequentially.These instability modes were respectively controlled by the anti-dip joint,the joint parallel to slope surface with a dip angle smaller than the slope angle(singlejoint),and two groups of joints inclined out of the slope(double-joints).Regarding the control effects on slope block instability,the stabilization ability of doublejoints(72.7%),anti-dip joint(67.4%),and single-joint(57.6%)decreased sequentially,resulting in different probabilities of slope block instability.Additionally,nearby regional faults significantly influenced the joints,leading to spatial heterogeneity and segmental clustering in the stabilization ability provided by joints to the slope blocks.Consequently,the stability of slope blocks gradually weakened as they approached the fault zones.This paper can provide guidance and assistance for investigating the development characteristics of rock mass structures and the stability of slope blocks.

Selective photosynthesis of Z-olefins through crystalline metal-organic cage-initiated expeditious cascade reactions

The semi-hydrogenation of alkyne to form Z-olefins with high conversion and high selectivity is still a huge challenge in the chemical industry.Moreover,flammable and explosive hydrogen as the common hydrogen source of this reaction increases the cost and danger of industrial production.Herein,we connect the photocatalytic hydrogen evolution reaction and the semihydrogenation reaction of alkynes in series and successfully realize the high selective production of Z-alkenes using low-cost,safe,and green water as the proton source.Before the cascade reaction,a series of isomorphic metal–organic cage catalysts(Co_(x)Zn_(8−x)L_(6),x=0,3,4,5,and 8)are designed and synthesized to improve the yield of the photocatalytic hydrogen production.Among them,Co_(5)Zn_(3)L_(6) shows the highest photocatalytic activity,with a H_(2) generation rate of 8.81 mmol g^(−1) h^(−1).Then,Co_(5)Zn_(3)L_(6) is further applied in the above tandem reaction to efficiently reduce alkynes to Z-alkenes under ambient conditions,which can reach high conversion of>98%and high selectivity of>99%,and maintain original catalytic activity after multiple cycles.This“one-pot”tandem reaction can achieve a highly selective and safe stepwise conversion from water into hydrogen into Z-olefins under mild reaction conditions.
