Background:The mechanisms underlying lesions of dopaminergic(DA)neurons,an essential pathology of Parkinson’s disease(PD),are largely unknown,although oxidative stress is recognized as a key *** have previously shown that the pro-oxidative aldehyde acrolein is a critical factor in PD pathology,and that acrolein scavenger hydralazine can reduce the elevated acrolein,mitigate DA neuron death,and alleviate motor deficits in a 6-hydroxydopamine(6-OHDA)rat *** such,we hypothesize that a structurally distinct acrolein scavenger,dimercaprol(DP),can also offer neuroprotection and behavioral ***:DP was used to lower the elevated levels of acrolein in the basal ganglia of 6-OHDA *** acrolein levels and related pathologies were measured by *** and behavioral effects of 6-OHDA injections and DP treatment were examined using the open field test and rotarod *** was assessed using mechanical allodynia,cold hypersensitivity,and plantar ***,the effects of DP were assessed in vitro on SK-N-SH dopaminergic cells exposed to ***:DP reduced acrolein and reversed the upregulation of pain-sensing transient receptor potential ankyrin 1(TRPA1)channels in the substantia nigra,striatum,and *** also mitigated both motor and sensory deficits typical of *** addition,DP lowered acrolein and protected DA-like cells in ***’s ability to upregulate TRPA1 was also verified in vitro using cell ***:These results further elucidated the acrolein-mediated pathogenesis and reinforced the critical role of acrolein in PD while providing strong arguments for anti-acrolein treatments as a novel and feasible strategy to combat neurodegeneration in *** the extensive involvement of acrolein in various nervous system illnesses and beyond,anti-acrolein strategies may have wide applications and broad impacts on human health.

Switchable carbamate coagulants to improve recycling ionic liquid from biomass solutions

A reversible amine-carbamate approach has been developed to reduce the use of antisolvents such as water in the coagulation of biopolymers from ionic liquid(IL)solution and thus improve the economy of IL *** and chitin were recovered from 1-ethyl-3-methylimidazolium acetate([C_(2)mim][OAc])solution by introducing the miscible amines triethanolamine(TEA),ethylenediamine(EDA),or butylamine(BA)and bubbling CO_(2) at 40℃ and atmospheric pressure through the solutions to form carbamate salts in situ which resulted in biopolymer *** gave the best results because of its low boiling point and low viscosity,which benefited both biopolymer recovery and IL *** films and fibers could be formed by extrusion of an MCC/[C_(2)mim]-[OAc]solution into a coagulating bath comprised of a 1:1 M mixture of[C_(2)mim][OAc]and butylammonium butylcarbamate(BA-carbamate).The cellulose,IL,amine,and CO_(2) were easily separated,although the cellulose recovered required some water washings to remove traces of *** to 96.4%of the[C_(2)mim][OAc]could be recovered,76.2%from the coagulation bath and 20.2%from the water *** recycled IL was suitable for another cycle of cellulose dissolution and extrusion and 84.6%of the IL used for the second cycle was *** further work is needed,not the least of which will be reducing the amount of water needed for washing steps,some promising features of this process point the way for new directions for more economically viable IL recycling processes in biomass treatment with ILs.

Targeting ERK1/2-calpain 1-NF-κB signal transduction in secondary tissue damage and astrogliosis after spinal cord injury

Neuronal damage, glial inflammation, and astrogliosis/astroglial scar formation are major secondary injury mechanisms that are significant contributors to functional deficits after spinal cord injury （SCI）. The objectives of the study were to evaluate the distinct roles of ERK2 vs. ERK1/2 and ERK1/2-calpain 1 -NF-r,B signal transduction in the tissue damage and astrogliosis/astroglial scar formation following SCI in rats. RNAi approaches, pharmacological intervention （U0126）, Western blot analysis, immunofluorescence analysis, and histological assessment were used to target ERK1/2-calpain 1-NF-KB signal transduction pathway for neuroprotection. Histological staining analysis demonstrated that selectively reducing pERK2 using ERK2 siRNA, but not inhibition of pERK1/2 with U0126, significantly reduced lesion volume and improved total tissue sparing, white matter sparing, and gray matter sparing in spinal cord two weeks after contusive SCI. An ERK1/2-calpain 1-NF-KB signal transduction pathway was involved in the astroglial scar formation after SCI. Blockade of ERK1/2 by U0126 decreased calpain 1 expression 4 h following SCI. Selective calpain 1 reduction by lentiviral shRNA attenuated astroglial NF-κB activity and astroglial scar formation after SCI in rats. Taken together, these results demonstrate the involvement of individual ERK2 and caipain 1 signaling pathways in tissue damage and astrogliosis/astroglial scar formation in animal models of SCI. Therefore, targeting individual ERK and its downstream signal transduction of calpain 1-NF-κB may provide greater potential as novel therapeutics for minimizing tissue damage and astroglial scar formation following SCI.

A mechanics model is developed for the contact radius of stamps with pyramid tips in transfer *** is important to the realization of reversible control of adhesion,which has many important applications,such as climbing robots,medical tapes,and transfer printing of *** contact radius is shown to scale linearly with the work of adhesion between the stamp and the contacting surface,and inversely with the plane-strain modulus of the stamp. It also depends on the cone angle and tip radius of the stamp,but is essentially independent of details of the tip geometry.

Using Tillandsia recurvate (Ball Moss) as a Biological Indicator to Monitor Air Pollution and Retain Oil Pollution

Air pollution is defined as the presence of a substance in the atmosphere that is harmful to human health, living things, and/or has a negative impact on the environment. A plant such as Tillandsia recurvata, ball moss, could be used as an inexpensive biological indicator for urban pollution. The purpose of this research was to determine if ball moss could be used as a biological indicator of urban pollution and retain oil pollution. Multiple sites were identified and grouped by vehicular traffic frequency (counts) using the Louisiana State Department of Transportation and Development (LaDOTD) traffic data to randomly select five low (0.0 - 7000), and five medium/high frequency (7001 to >14,000) traffic counts in locations within Baton Rouge, La. city limits. Differential analysis determined that harvested ball moss tissue levels from areas with low traffic (<0.05 level) contained lower S concentrations than plants tested from high traffic counts. In a second study, dried Tillandsia recurvata plant tissue accumulated greater oil weight than absorbent paper towels. Tillandsia recurvata absorbed and/or retained oil at a greater ratio of oil than its own mass. Therefore, the results of each experiment indicated that Tillandsia recurvata may successfully function as a biological indicator and serve as an oil retentionist on a small-scale test. Further research is needed on a larger-scale area to confirm the efficacy of ball mosses for controlling water pollution in-situ.

Characterization and transcriptional analyses of cDNAs encoding three trypsin- and chymotrypsin-like proteinases in CrylAb-susceptible and CrylAb-resistant strains of sugarcane borer, Diatraea saccharalis

Diatraea saccharalis is a major corn borer pest. Midgut serine proteinases are essential for insect growth and development. Alteration of midgut proteinases is re- sponsible for Bt resistance development in some species. To clone midgut trypsin and chymotrypsin cDNAs and to test if the CrylAb resistance in D. saccharalis is associ- ated with changes in midgut proteinases, total midgut tryptic and chymotryptic activities, cDNA sequences, and gene expressions of three trypsin and three chymotrypsin genes were comparatively examined between Cry 1 Ab-susceptible （Cry 1 Ab- S S） and Cry 1 Ab-resistant （Cry 1 Ab-RR） strains. Full-length cDNAs encoding three trypsin- and three chymotrypsin- like proteinases were sequenced from CrylAb-SS and CrylAb-RR larvae. These cDNAs code for active forms of midgut serine proteinases with all fimctional motifs, includ- ing signal peptide, conserved His-Asp-Ser for the catalytic triad, three pairs of cysteines for disulfide bridge configurations, and conserved substrate specificity determination residues. In general, cDNA and putative protein sequences are highly similar between CrylAb-SS and CrylAb-RR strains, except for a few nucleotide and predicted amino acid substitutions, whose function need to be further clarified. Total trypsin and chymotrypsin activities were also similar between CrylAb-SS and CrylAb-RR strains. Transcriptional levels of the trypsin and chymotrypsin genes had numerical difference between Cry 1 Ab-SS and CrylAb-RR strains, but the difference was not statistically significant. Data suggest that the development of CrylAb resistance in D. saccharalis was not significantly as- sociated with these trypsins and chymotrypsins. Results clarified the role of six midgut proteinases and provided a foundation for continuing examination of potential involvement of other midgut proteinases in Bt resistance development and other important biochemical processes.
