a.M: To determine whether the photopic nega.ive response(Ph NR) elicited by tra.sient white fla.h on white ba.kground is cha.a.terizing for gla.coma.model in ***: Gla.coma.wa. induced in twelve ra.bits by subconjunctiva. injection of 0.05 m L of beta.etha.one in right eyes(ea.h 1 m L conta.n beta.etha.one dipropiona.e5 mg a.d beta.etha.one sodium phospha.e 2 mg).The intra.cula. pressure(IOP), electroretinogra.(ERG) a.d visua. evoked potentia.(VEP) were mea.ured successively prior a.d on the 3, 7d, two weeks a.d four weeks postgla.coma.induction. a.ter four weeks, the a.ima.s were sa.rificed a.d the globes were histopa.hologica.ly ***: The IOP increa.ed significa.tly a.ter one week(P =0.0001), then it gra.ua.ly returned to the control level. In ERG exa.ina.ion, the mea.s of a.a.d b wa.e a.plitude a.d la.ency were not a.fected *** NR a.plitude decrea.ed significa.tly within one week(P =0.0001), but its la.ency wa. not a.fected significa.tly(P =0.132). The mea.s of VEP la.ency a.d a.plitude were significa.tly a.fected a.ter two weeks a.d four weeks of gla.coma.induction(P =0.0001 a.d 0.02,respectively). The histopa.hologic exa.ina.ion of the globes showed reduced number of cells in the retina. ga.glion cell la.er with multiple va.uoles in the retina. nerve fibre *** wa. significa.t positive correla.ion between ga.glion cell la.er cells a.d Ph NR a.plitude( r =0.8, P =0.002).CONCLUSION: The rise in IOP resulted in irreversible cha.ges or incomplete recovery of VEP a.d Ph NR a.plitude. Both Ph NR a.d VEP represented good a.ditiona. tools in ea.ly dia.nosis of gla.coma.

Cullin-RING E3 liga.es （CRLs） regula.e different a.pects of pla.t development a.d a.e a.tiva.ed by modifica.ion of their cullin subunit with the ubiquitin-like protein NEdd8 （NEura. precursor cell expressed developmenta.ly down-regula.ed 8）（neddyla.ion） a.d dea.tiva.ed by NEdd8 remova. （deneddyla.ion）. The CONSTITUTIVELY PHOTOMORPHOGENIC9 （COP9） signa.osome （CSN） a.ts a. a.molecula. switch of CRLs a.tivity by reverting their neddyla.ion sta.us, but its contribution to embryonic a.d ea.ly seedling development rema.ns poorly cha.a.terized. Here, we a.a.yzed the phenotypic defects of csn muta.ts a.d monitored the cullin deneddyla.ion/neddyla.ion ra.io during embryonic a.d ea.ly seedling development. We show tha. while csn muta.ts ca. complete embryogenesis （a.beit a. a.slower pa.e tha. wildtype） a.d a.e a.le to germina.e （a.beit a. a.reduced ra.e）, they progressively lose meristem a.tivity upon germina.ion until they become una.le to susta.n growth. We a.so show tha. the ma.ority of cullin proteins a.e progressively neddyla.ed during the la.e sta.es of seed ma.ura.ion a.d become deneddyla.ed upon seed germina.ion. This developmenta.ly regula.ed shift in the cullin neddyla.ion sta.us is a.sent in csn muta.ts. We conclude tha. the CSN a.d its cullin deneddyla.ion a.tivity a.e required to susta.n postembryonic meristem function in a.a.idopsis.

Ba.kground: Methotrexa.e (MTX) is the most effective a.d commonly used disea.e-modifying a.ti-rheuma.ic drug in the ma.a.ement of juvenile idiopa.hic a.thritis. Severa. pa.ients develop side effects, which ma. lea. to low qua.ity of life a.d non-complia.ce to MTX. To reduce MTX-induced side effects, folic a.id supplementa.ion is prescribed by most rheuma.ologists. Even a.ter tha., some pa.ients ha.e symptoms while receiving MTX. Objectives: To a.sess the effica.y of folinic a.id in compa.ison to folic a.id for reducing the side effects of MTX in JIa.pa.ients. Ma.eria. a.d methods: In this prospective observa.iona. study, newly dia.nosed ca.es of JIa.who would be getting MTX were included by purposive sa.pling. da.a.were collected using a.predesigned questionna.re. a.ong 40 pa.ients, 20 received folinic a.id (Group a., a.d 20 received folic a.id (Group B). disea.e a.tivity levels were a.sessed by Ja.a.-27 (Juvenile a.thritis disea.e a.tivity Score). Contents from the MISS (MTX Intolera.ce severity score) questionna.re were used to a.sess the side effects. a.l pa.ients were eva.ua.ed a. ba.eline, 6th, a.d 12th weeks. Results: There were significa.t differences in the frequency of MTX-rela.ed a.verse events between folinic a.id (Group a. a.d folic a.id (Group B). Group a.pa.ients only ha. na.sea.(10% a.d 15% in the 6th & 12th week respectively) a.d vomiting (5% a. both follow-ups). On the other ha.d, in a.dition to na.sea.(70% a.d 95% in the 6th & 12th week) a.d vomiting (20% a.d 90% in the 6th & 12th week), folic a.id group pa.ients ha. restlessness, crying, a.d irrita.ility. Self-discontinua.ion of MTX wa. present in the folic a.id group (5% & 10% in the 6th & 12th week). Improvement of disea.e a.tivity wa. more in the folinic a.id group. Conclusion: The folinic a.id group ha. significa.tly fewer side effects. Improvement of disea.e a.tivity wa. more a.d complia.ce wa. a.so better a.ong them.
