SMRT sequencing and ddPCR reveal the complexity of developmental trajectories and temporal dynamics of gut bifidobacterial communities in infants

Infant intestinal microbiome is closely linked with health and risk of disease. Bifidobacterium are important components of the infant gut and are known to confer various health effects on the host. However, few studies have described the precise composition and dynamics of early infant gut bifidobacterial communities. Thus, this was a pilot study aiming to describe the developmental trajectories and temporal dynamics of bifidobacterial communities in infants before 6 months of age. A total of 28 fecal samples from 4 infants(GF, ZZ, QM, TN, respectively)were collected and analyzed after 5, 15, 30, 60, 90, 120, 150, and 180 days of birth by a bifidobacteria-target method(based on single-molecule real-time sequencing of partial bifidobacterial rpsK genes)in conjunction with droplet digital polymerase chain reaction(ddPCR). The infant fecal microbiota comprised a total of 11 bifidobacterial species, including 4 major species, i.e., B. dentium(37.35%), B. catenulatum(32.04%), B. breve(22.24%), and B. animalis(8.02%). The infant microbiota showed highly individualized developmental trajectories. The leading species for GF was B. catenulatum, with a relatively stable developmental trajectory. In ZZ, B. breve was enriched, and the developmental trajectory was rather fluctuating. The most abundant species for QM and TN was B. dentium. The developmental trajectory of B. dentium in QM showed a trend of gradual decrease, whereas an opposite trend was seen in samples of TN. The results of ddPCR confirmed large variations in quantities of bifidobacteria between infants and suggested discordances in temporal dynamics of bifidobacterial communities during the first half year of infancy. In conclusion, our results suggested that the early infant gut bifidobacterial microbiota was highly complex and temporal dynamics, with individualized developmental trajectories, which should be considered in future research of infant gut microbiota.

Synthesis of dandelion-like three-dimensional nickel nanostructures via solvothermal route

The objectives of the present research are synthesizing three-dimensional （3D） nickel nanostructures and investigating their magnetic properties. Thus a template-free method was used to prepare 3D dandelion-like nickel nanostructures via reducing of nickel chloride with hydrazine hydrate in ethylene glycol solution at 100 ℃. The resulting Ni nanostructures were characterized by means of powder X-ray diffraction （XRD）, scanning electron microscopy （SEM）, transmission electron microscopy （TEM）, and selected-area electron diffraction （SAED）. And the magnetic properties of the 3D Ni nanostructures were measured as well. Results indicated that solvothermal process could be successfully used to prepare 3D dandelion-like nanostructures of Ni at a relatively mild temperature of 100℃. And the conclusions were made as follows： as-prepared Ni samples had obvious shape anisotropy and were composed of fine nanocrystallites, while they had significantly enhanced ferromagnetic properties than bulk Ni and Ni nanoparticles.

P2-type Na_(0.67)Ni_(0.33)Mn_(0.67)O_(2)is considered as a potential cathode material for sodium-ion batteries due to the merits of high voltage,low cost,and air ***,the unsatisfied cycling stability and rate performance caused by the destructive phase transition and side reactions hinder its practical ***,we present a feasible dual strategy of Mg^(2+)doping integrated with ZrO_(2)surface modification for P2-Na_(0.67)Ni_(0.33)Mn_(0.67)O_(2),which can well address the issues of phase transition and side reactions benefitting from the enhanced structural and interfacial ***,it exhibits a decent cycling stability with a capacity retention of 81.5%at 1 C and promising rate performance with a discharge capacity of 76.6 mA h g^(−1)at 5 *** in situ X-ray diffraction measurement confirms that the damaged P2-O2 phase transition is suppressed with better reversibility in high-voltage region,whereas the side reactions are inhibited due to the protective ZrO_(2)***,the full cell achieves an outstanding operating voltage of 3.57 V and a fabulous energy density of 238.91 W h kg^(−1)at 36.73 W kg^(−1),demonstrating great *** work is expected to provide a new insight for designing stable high-voltage cathode materials and high energy density full cells for sodium ion batteries.

Preparation and nucleation of spherical metallic droplet

The preparation and solidification of metallic droplets attract more and more attention for their signiifcance in both engineering and scientiifc ifelds. In this paper, the preparation and characterization of Sn-based al oy droplets using different methods such as atomization and consumable electrode direct current arc （CDCA） technique are reviewed. The morphology and structure of these droplets were determined by optical microscopy, X-ray diffraction （XRD） and scanning electron microscopy （SEM）. The solidiifcation behavior of single droplet was systematical y studied by means of scanning calorimetry （DSC）, and the nucleation kinetics was also calculated. In particular, the development of fast scanning calorimetry （FSC） made it possible to investigate the evolution of undercooling under ultrafast but control able heating and cooling conditions. The combination of CDCA technique and FSC measurements opens up a new door for quantitative studies on droplet solidiifcation, which is accessible to demonstrate some theories by experiments.

BACKGROUND Cronkhite-Canada syndrome(CCS)is a rare nonhereditary disease characterized by chronic diarrhoea,diffuse gastrointestinal polyposis and ectodermal *** lethality of CCS can be up to 50%if it is untreated or if treatment is delayed or *** than 35%of the patients do not achieve long-term clinical remission after corticosteroid administration,with relapse occurring during or after the cessation of glucocorticoid *** optimal strategy of maintenance therapy of this disease is *** SUMMARY A 47-year-old man presented to the hospital with a 3-mo history of frequent watery diarrhoea,accompanied by macular skin pigmentation that included the palms and soles,and onychodystrophy of the fingernails and *** and colonoscopy revealed numerous polyps in the stomach and *** other possibilities were ruled out by a series of examinations,CCS was diagnosed and treated with *** patient took prednisone for more than 1 year before achieving complete resolution of his symptoms and endoscopic *** patient was then given prednisone 5 mg/d for 6 mo of maintenance *** clinical improvement and polyp regression,prednisone was *** mo after the discontinuation of prednisone,the diarrhoea and gastrointestinal polyps ***,the patient was given the same dose of prednisone,and complete remission was achieved *** It is necessary to extend the duration of prednisone maintenance therapy for *** is still effective when readministered after *** endoscopy at intervals of 1 year or less is recommended to assess mucosal disease activity.

Columnar to Equiaxed Transition During Solidification of Small Ingot by Using Electric Current Pulse

A new approach to applying the electric current pulse （ECP） with parallel electrodes to the promotion of the transition from columnar crystal to equiaxed crystal and the improvement of macrosegregation was introduced. The ECP was applied to different stages of the solidification. The results showed that the application of the ECP in both the initial stage （the thickness of solidified shell reached 2 mm approximately） and the late stage （the thickness of solidified shell reached 14 mm approximately） of solidification can promote the columnar to equiaxed transition （CET）. The analysis showed that during solidification, a large number of nuclei around the upper surface fell off due to ECP, which subsequently showered on the melt and impinged the growth front of the columnar crystal. Therefore, the CEToccurred. In addition, this method was also employed to influence the solidification process of bearing steel, and the results showed that the structure was changed from columnar crystal to equiaxed crystal, indicating that ECP can enhance the homogeneity of structure and composition of bearing steel.

Repair of long segmental ulnar nerve defects in rats by several different kinds of nerve transposition

Multiple regeneration of axonal buds has been shown to exist during the repair of peripheral nerve injury, which confirms a certain repair potential of the injured peripheral nerve. Therefore, a systematic nerve transposition repair technique has been proposed to treat severe peripheral nerve injury. During nerve transposition repair, the regenerated nerve fibers of motor neurons in the anterior horn of the spinal cord can effectively grow into the repaired distal nerve and target muscle tissues, which is conducive to the recovery of motor function. The aim of this study was to explore regeneration and nerve functional recovery after repairing a long-segment peripheral nerve defect by transposition of different donor nerves. A long-segment(2 mm) ulnar nerve defect in Sprague-Dawley rats was repaired by transposition of the musculocutaneous nerve, medial pectoral nerve, muscular branches of the radial nerve and anterior interosseous nerve(pronator quadratus muscle branch). In situ repair of the ulnar nerve was considered as a control. Three months later, wrist flexion function, nerve regeneration and innervation muscle recovery in rats were assessed using neuroelectrophysiological testing, osmic acid staining and hematoxylin-eosin staining, respectively. Our findings indicate that repair of a long-segment ulnar nerve defect with different donor nerve transpositions can reinnervate axonal function of motor neurons in the anterior horn of spinal cord and restore the function of affected limbs to a certain extent.

Highly reversible Zn anode enabled by anticatalytic carbon layer with suppressed hydrogen evolution

Aqueous zinc-ion batteries(AZIBs)have emerged as a promising high-efficiency energy storage system due to the high energy density,low-cost and environmental ***,the practical application of AZIBs is severely restricted by the challenges faced by the Zn anode,which include uncontrollable dendrite growth,corrosion and hydrogen evolution ***,a simple and convenient physical vapor deposition(PVD)method is reported for fabricating uniform graphite as a protection layer on the surface of Zn *** high conductivity graphite layer on Zn anode(denoted as Zn@C)not only benefits the uniform distribution of the electric field,but also provides numerous Zn nucleation sites to regulate and navigate Zn-ion stripping/plating ***,the graphite layer with a poor catalytic activity endows the Zn@C anode with a highly suppressed hydrogen ***,a hydrogen and dendrite free anode is achieved with artificial anticatalytic carbon layer on Zn anode,exhibiting a high reversibility and excellent cycling stability over 2600 h at the current density of 5 mA·cm^(-2)with a capacity of 2.5 mAh·cm^(-2)and longtime cycling stability for assembled full *** work strategically designs the properties of the artificial interface layer to effectively address various challenges simultaneously,which presents insights for the future development of high-performance rechargeable AZIBs.
