The sensorimotor and histological aspects of peripheral neuropathies were already studied by our team in two rat models:the sciatic nerve crush and the Charcot-Marie-Tooth-1A disease.In this study,we sought to highlight and compare the protein signature of these two pathological situations.Indeed,the identification of protein profiles in diseases can play an important role in the development of pharmacological targets.In fact,Charcot-Marie-Tooth-1A rats develop motor impairments that are more severe in the hind limbs.Therefore,for the first time,protein expression in sciatic nerve of Charcot-Marie-Tooth-1A rats was examined.First,distal sciatic nerves were collected from Charcot-Marie-Tooth-1A and uninjured wild-type rats aged 3 months.After protein extraction,sequential window acquisition of all theoretical fragment ion spectra liquid chromatography and mass spectrometry was employed.445 proteins mapped to Swiss-Prot or trEMBL Uniprot databases were identified and quantified.Of these,153 proteins showed statistically significant differences between Charcot-Marie-Tooth-1A and wild-type groups.The majority of these proteins were overexpressed in Charcot-Marie-Tooth-1A.Hierarchical clustering and functional enrichment using Gene Ontology were used to group these proteins based on their biological effects concerning Charcot-Marie-Tooth-1A pathophysiology.Second,proteomic characterization of wild-type rats subjected to sciatic nerve crush was performed sequential window acquisition of all theoretical fragment ion spectra liquid chromatography and mass spectrometry.One month after injury,distal sciatic nerves were collected and analyzed as described above.Out of 459 identified proteins,92 showed significant differences between sciatic nerve crush and the uninjured wild-type rats used in the first study.The results suggest that young adult Charcot-Marie-Tooth-1A rats(3 months old)develop compensatory mechanisms at the level of redox balance,protein folding,myelination,and axonogenesis.These mechanisms seem insufficient

Osgood Schlatter Disease: Study of 2 Observations in Conakry, Guinea

Introduction: OSD is a growth osteochondrosis affecting the apophyseal cartilage of the anterior tibial tuberosity of the knee, 20 to 30% of young growing athletes suffer from this pathology, described by Osgood and Schlatter in 1903. Objective: To report 2 observations and review the literature. Observation 1: This was a 14-year-old Guinean teenager, Halpoular, living in Conakry, a right-handed footballer who presented with a painful swelling of the right knee that looked mechanical, whose father had HLA B27 positive ankylosing spondylitis. X-ray of the right knee showed fragmentation of the anterior tibial tuberosities, and ossicle with edema of the patellar tendons at stage 4 according to Ehrenbord and Lagergren (1961). Thus, the disease of OSD is retained, He received 120 mg of diclofenac, 20 mg omeprazole for 1 month and the cessation of sports activity, the evolution was favorably marked by the regression of pain. Observation 2: A 16-year-old girl, initials M.L.T, Guinean, residing in Coyah (Republic of Guinea), a handball player, consulted for intense pain (VAS: 8/10) in her knees after a violent shock received when she hit the post. The physical examination found a painful, soft swelling at the anterior tibial tuberosity of the right knee, the rest of the examination was normal. The X-ray of the right knee shows fragmentation of the anterior tibial tuberosity, and ossicle with edema of the patellar tendon at stage 4 according to Ehrenbord and Lagergren 1961, thus the OSD disease is retained, the treatment instituted was 120 mg of diclofenac, the cessation of sports activity which gives a good evolution (EVA at 2/10). Conclusion: OSD is a growth osteochondrosis affecting the apophyseal cartilage of the anterior tibial tuberosity of the knee in young growing athletes, resolving at rest.

Epidemiological, Clinical Progress Aspects of Congenital Heart Disease with Neonatal Revelation at the Mother-Child Hospital of Bingerville (HME) Concerning 98 Cases from January 2021 to December 2022 (Côte d’Ivoire)

Introduction The distribution of congenital heart disease (CHD) in sub-Saharan Africa is highly imprecise and varies from one region to another due to the inequality of diagnostic facilities. The aim of this study was to determine the in-hospital prevalence of congenital heart disease in children at the Mother-Child hospital of Bingerville (HME) by specifying the diagnostic, therapeutic and evolutionary aspects. Materials and methods We conducted a retrospective, descriptive, cross-sectional study at HME of Bingerville from January 2021 to December 2022. All newborns with congenital heart disease confirmed by echocardiography were included in the study. Results Of 656 admissions to the neonatology department over the study period, congenital heart disease accounted for 14.9% (98/656) of cases. In our series, 76.7% were diagnosed before the 1st week of life, with a mean chronological age of 5.18 days and extremes of 0 and 46 days. There were as many male patients (50%) as female (50%), i.e. a sex ratio of 1. These newborns were premature in 60.2% of cases, with a mean and median gestational age of 34 weeks’ amenorrhea. Most were left-right shunts (90.8%). Persistent ductus arteriosus (PDA) (48.9%) predominated, followed by atrial septal defect (38.7%), ventricular septal defect (13.3%), common trunk artery (CTA) (3.1%) and open septal pulmonary atresia (OSPA) (1%) as the primary cyanogenic heart disease. Pulmonary arterial hypertension (PAH) (50%) was primary in 38.8% and secondary (61.2%). The mortality rate was 30.6%, and all CTA patients died (100%), with a significant statistical relationship (p = 0.027). Progression under treatment was marked by clinical stabilization (68/98) in 69.4% of cases. Conclusion: Congenital heart disease is relatively common at the Bingerville HME. Access to echocardiography should be facilitated in neonatology departments for rapid diagnosis and optimal management of congenital heart disease in newborns.

背景:维持性血液透析患者(Maintenance Hemodialysis MHD)第一死亡原因为心脑血管疾病,而血管内皮细胞的损伤坏死为其主要临床特征,但是内皮细胞凋亡的发病机制尚未清楚。本研究观察了 MHD合并心血管疾病患者血浆外泌体(Serum-derived exosomes Serum-exo)体外对主动脉内皮细胞(Human Aortic Endothelial Cells HAECs)凋亡的影响机制。方法:为了研究MHD患者血浆由来外泌体,体外对内皮细胞系凋亡的信号传导机制,本实验采用以下8个研究步骤进行探索:1.取3名MHD合并心血管疾病患者血浆及3名正常人血浆。通过透射电子显微镜,纳米颗粒跟踪分析仪以及Western blot分析,观察和鉴定血浆由来的外泌体;2.通过患者血浆与HAECs进行共培养,观察患者血浆对HAECs细胞的凋亡影响;3.通过miRNAs sequencing及RT-qPCR,比较正常人组与维持性血液透析组血浆微小RNA表达的差异;4.提取外泌体后,通过RT-qPCR,观察外泌体组(Serum-exo)、外泌体+抑制剂组(serum-exo+GW4869(外泌体抑制剂))及Mock组,观察血浆外泌体对HAECs细胞内miR-30a-5p水平的影响;用Western blot分析细胞凋亡分子Caspase-3,8;用流式细胞仪,观察以上三组细胞凋亡率变化;5.通过设计 miR-30a-5p 的 mimic sense 及 anti-sense,上调或下调 miR-30a-5p 表达,进一步观察miR-30a-5p对HAECs细胞凋亡的影响;6.为了观察miR-30a-5p是否通过镁离子受体 Transient Receptor Potential Melastatin 7(TRPM7)起作用;用生物素标记的miR-30a-5p为探针,采用双荧光素酶报告分析及用RT-qPCR法,探测TRPM7表达;设计了 TRPM7siRNA,上调或/和下调HAECs细胞中miR-30a-5p和TRPM7,观察上调/下调对HAECs细胞凋亡的影响;为了进一步评估miR-30a-5p调节HAECs上皮细胞凋亡是否通过TRPM7,观察一系列不同转染细胞组别:NC+Control Vector;miR-30a-5p mimic+Control Vector;NC+TRPM7 OE;miR-30a-5p mimic+TRPM7 OE。7.我们假设:miR-30a-5p通过调节TRPM7的表达影响细胞内镁离子浓度变化,从而影响细胞内氧化应激,影响细胞凋亡。为了证实该假设,(1)设计了 HAECs上皮细胞与serum-exo和 serum-exo+ GW4869 共培养,通过 Mg2+indicator Mag-indo-1 测定镁离子在 HAECs细胞中浓度的变化;(2)通过上调或下调HAECs细胞中镁离子的浓度,用DCFH-DA fluorescent probe staining 测定 HAECs 细胞中的 Reactive Oxygen Species(ROS)浓度的变化;通过DHE intensity观察ROS的time course;用Western blot分析细胞凋亡分子Caspase-3,8的表达;用流式细胞仪观察细胞凋亡率改变,最终观察HAECs细胞中镁离子浓度的改变对ROS浓度及细胞凋亡的影响。8.为了进一步证明miR-30a-5p是通过TRPM7来调节镁离子介导的ROS诱导的HAECs上皮细胞的凋亡,我们设计了(1)在细胞外H202(50 μM)存在的情况下,和在改变miR-30a-5p或TRPM7水平情况下,以及镁离子有(10 mM)或无(0.1 mM)的情况下,用DHE强度测定细胞内ROS相对的浓度的实验;(2)转染细胞组别:NC+control vector细胞组;miR-30a-5p mimic+control vector 细胞组;miR-30a-5p mimic+control vector+NAC 细胞组;NC+TRPM7 OE细胞组;NC+TRPM7 OE+H2O2细胞组,观察各组细胞调亡发生率的改变。结果:本研究的一系列实验结果如下:1.分离和鉴定了 MHD患者中血浆存在外泌体。透视电镜和纳米颗粒跟踪分析观察结果为:外泌体直径分布于30到150 nm,Western blot分析发现该外泌体表达CD63和CD9蛋白,且分别于对照组比较,有显著差异(P<0.05,P<0.01)。2.血浆中RT-qPCR测定结果:与正常人血浆比较,MHD患者血浆中miR-30a-5p含量较高(P<0.001)。3.对3名MHD患者及正常人血浆miRNAs sequencing及RT-qPCR结果发现:有5种miRNA在MHD患者血浆中较高,其中miR-30a-5p水平最高(P<0.001与正常

Effect of low temperature in the development cycle of Lucilia sericata (Meigen) (Diptera, Calliphoridae): implications for the minimum postmortem interval estimation

Knowledge of necrophagous insects' developmental data is necessary for the forensic entomologist to estimate a reliable minimum postmortem interval (PMImin).Among the most represented necrophagous species,Lucilia sericata (Diptera,Calliphoridae) is particularly interesting.It is regularly identified in samples,with a predominance in summer,and is commonly used by analysts of our entomology department (Institut de Recherche Criminelle de la Gendarmerie Nationale) to estimate the PMImin with the accumulated degree days (ADD) method.This method requires the mathematical lower thermal threshold to be known.This value dictates the quality of the applied ADD method but cannot be considered as fixed,especially when insect development occurs at temperatures close to the biological threshold.In such conditions,it is necessary to study the influence of such temperatures on development rate,as well as the consequences of estimating the period of first oviposition on cadavers,when using the ADD method.Seven replicate rearings were conducted at six different temperatures: 30 ℃,24 ℃,18 ℃,15 ℃,12 ℃ and 10 ℃.Time of development and time of emergence were recorded.The effect of low temperature on the development cycle and the reliability of the ADD method under this entire temperature spectrum were studied using different linear regression models.Calculated durations of total insect time development and experimental rearing duration were then compared.A global linear model cannot be used on the whole temperature spectrum experienced by L.sericata without resulting in an overestimation at some temperatures.We found a combination of two linear regression models to be suitable for the estimation of the total development time,depending on the temperature experienced by L.sericata.This approach allowed us to obtain a variation lower than 2% at 12 ℃ and 10 ℃ between the calculated duration and experimental duration of development.In comparison,the results obtained with a global model show a variation higher than 3% at 12 ℃ and 10%at 10

Seed exudates influence the behavior of soil organisms,but howthis occurs remains unclear,particularly for multicellular animals.Here we show that compounds associated with Arabidopsis seed-coat mucilage regulate the behavior of soil-borne animals,specifically root-knot nematodes (RKNs).Infective RKN J2 larvae actively travel toward Arabidopsis seeds through chemotaxis.Analysis of Arabidopsis mucilage mutants demonstrated that the attraction of RKNs toArabidopsis seeds requires the synthesis and extrusion of.seed-coat mucilage.Extracted mucilage alone is not sufficient to attract RKNs,but seed-surface carbohydrates and proteins are required for this process.These findings suggest that the RKN chemoattractant is synthesized de novo upon mucilage extrusion but may be highly unstable.RKNs attracted by thismucilage-dependent mechanism can infect the emerging seedling.However,the attraction signal from seedling roots likely acts independently of the seed-coat signal and may mask the attraction to seed-coat mucilage after germination.Multiple RKN species are attracted byArabidopsis seeds,suggesting that this mechanism is conserved in RKNs.These findings indicate that seed exudate can regulate the behavior of multicellular animals and highlight the potential roles of seed-coat mucilage in biotic interactions with soil microorganisms.

The high cytotoxic activity of Vγ9Vδ2 T lymphocytes against tumor cells makes them useful candidates in anticancer therapies.However,the molecular mechanism of their activation by phosphoantigens(PAgs)is not completely known.Many studies have depicted the mechanism of Vγ9Vδ2 T-cell activation by PAg-sensed accessory cells,such as immune presenting cells or tumor cells.In this study,we demonstrated that pure resting Vγ9Vδ2 T lymphocytes can self-activate through exogenous PAgs,involving their TCR and the butyrophilins BTN3A1 and BTN2A1.This is the first time that these three molecules,concurrently expressed at the plasma membrane of Vγ9Vδ2 T cells,have been shown to be involved together on the same and unique T cell during PAg activation.Moreover,the use of probucol to stimulate the inhibition of this self-activation prompted us to propose that ABCA-1 could be implicated in the transfer of exogenous PAgs inside Vγ9Vδ2 T cells before activating them through membrane clusters formed byγ9TCR,BTN3A1 and BTN2A1.The self-activation of Vγ9Vδ2 T cells,which leads to self-killing,can therefore participate in the failure ofγδT cell-based therapies with exogenous PAgs and should be taken into account.

NTR/NRX Define a New Thioredoxin System in the Nucleus of Arabidopsis thaliana Cells

Thioredoxins （TRX） are key components of cellular redox balance, regulating many target proteins through thiol/disulfide exchange reactions. In higher plants, TRX constitute a complex multigenic family whose members have been found in almost all cellular compartments. Although chloroplastic and cytosolic TRX systems have been largely studied, the presence of a nuclear TRX system has been elusive for a long time. Nucleoredoxins （NRX） are potential nuclear TRX found in most eukaryotic organisms. In contrast to mammals, which harbor a unique NRX, angiosperms gen- erally possess multiple NRX organized in three subfamilies. Here, we show thatArabidopsis thaliana has two NRXgenes （AtNRX1 and AtNRX2）, respectively, belonging to subgroups I and III. While NRX1 harbors typical TRX active sites （WCG/ PPC）, NRX2 has atypical active sites （WCRPC and WCPPF）. Nevertheless, both NRXl and NRX2 have disulfide reduction capacities, although NRX1 alone can be reduced by the thioredoxin reductase NTRA. We also show that both NRX1 and NRX2 have a dual nuclear/cytosolic localization. Interestingly, we found that NTRA, previously identified as a cytosolic protein, is also partially localized in the nucleus, suggesting that a complete TRX system is functional in the nucleus. We show that NRX1 is mainly found as a dimer in vivo. nrxl and nrx2 knockout mutant plants exhibit no phenotypic perturbations under standard growth conditions. However, the nrxl mutant shows a reduced pollen fertility phenotype, suggesting a specific role of NRX1 at the haploid phase.
