Silk-based nerve guidance conduits with macroscopic holes modulate the vascularization of regenerating rat sciatic nerve

Peripheral nerve injuries induce a severe motor and sensory deficit. Since the availability of autologous nerve transplants for nerve repair is very limited, alternative treatment strategies are sought, including the use of tubular nerve guidance conduits(tngCs). However, the use of tngCs results in poor functional recovery and central necrosis of the regenerating tissue, which limits their application to short nerve lesion defects(typically shorter than 3 cm). given the importance of vascularization in nerve regeneration, we hypothesized that enabling the growth of blood vessels from the surrounding tissue into the regenerating nerve within the tngC would help eliminate necrotic processes and lead to improved regeneration. In this study, we reported the application of macroscopic holes into the tubular walls of silk-based tngCs and compared the various features of these improved silk^(+) tngCs with the tubes without holes(silk^(–) tngCs) and autologous nerve transplants in an 8-mm sciatic nerve defect in rats. Using a combination of micro-computed tomography and histological analyses, we were able to prove that the use of silk^(+) tngCs induced the growth of blood vessels from the adjacent tissue to the intraluminal neovascular formation. A significantly higher number of blood vessels in the silk^(+) group was found compared with autologous nerve transplants and silk^(–), accompanied by improved axon regeneration at the distal coaptation point compared with the silk^(–) tngCs at 7 weeks postoperatively. In the 15-mm(critical size) sciatic nerve defect model, we again observed a distinct ingrowth of blood vessels through the tubular walls of silk^(+) tngCs, but without improved functional recovery at 12 weeks postoperatively. Our data proves that macroporous tngCs increase the vascular supply of regenerating nerves and facilitate improved axonal regeneration in a short-defect model but not in a critical-size defect model. A stability-indicating LC–MS/MS method for zidovudine: Identification,characterization and toxicity prediction of two major acid degradation products

Zidvovudine(AZT) is a nucleoside analogue reverse transcriptase inhibitor(nRTI), a class of anti-retroviral drug. A stability-indicating assay method for AZT was developed in line with ICH guideline. Successful separation of AZT and its degradation products was achieved by gradient elution mode on reverse phase C_(18) column using 10 mM ammonium acetate: acetonitrile as the mobile phase at 0.8 mL/min flow rate, 25 μL injection volume, 30 °C column temperature and 285 nm detection wavelength. Two major acid degradation products were identified and characterized by liquid chromatography–electrospray ionization mass spectrometry(LC–ESI/MS/MS) and accurate mass measurements. The probable mechanisms for the formation of degradation products were identified based on a comparison of the fragmentation pattern of the [M + H]^+ions of AZT and its degradation products. One of the degradation products, DP-1, was isolated by semi-preparative high performance liquid chromatography(HPLC) using Waters XBridge Prep C_(18)(250 mm×10 mm, 5 μm).Degradation products showed higher toxicity compared to the drug in some models assessed by TOPKAT software. The method validation was performed with respect to robustness, specificity, linearity, precision and accuracy as per ICH guideline Q2(R1).

Metabolic syndrome is a cluster of several clinical conditions characterized by insulin-resistance and high cardiovascular risk. non-alcoholic fatty liver disease is the liver expression of the metabolic syndrome, and insulin resistance can be a frequent comorbidity in several chronic liver diseases, in particular hepatitis C virus infection and/or cirrhosis. Several studies have demonstrated that insulin action is not only relevant for glucose control, but also for vascular homeostasis. Insulin regulates nitric oxide production, which mediates to a large degree the vasodilating, antiinflammatory and antithrombotic properties of a healthy endothelium, guaranteeing organ perfusion. The effects of insulin on the liver microvasculature and the effects of IR on sinusoidal endothelial cells have been studied in animal models of non-alcoholic fatty liver disease. The hypotheses derived from these studies and the potential translation of these results into humans are critically discussed in this review.

Wearable gas sensors that are lightweight, portable, and inexpensive have great potential application in the real-time detection of human health and environmental monitoring. In this work, we fabricated flexible fiber gas sensors with single-walled carbon nanotube （SWCnT）, multi-walled carbon nanotube （MWCnT）, and ZnO quantum dot-decorated SWCnT （SWCnTs@ZnO） sensing elements. These flexible fiber gas sensors could be operated at room temperature to detect target gases with good sensitivity and recovery time. They also exhibited superior long-term stability, as well as good device mechanical bending ability and robustness. Integrating these flexible gas sensors into face masks, the fabricated wearable smart face masks could be used to selectively detect CaHsOH, HCHO, and nH3 by reading the corresponding LEDs with different colors. Such face masks have great potential application in the Internet of Things and wearable electronics.

Co,ni,and Zn iso-reticular coordination polymers of 2,5-dihydroxyterphthalic acid(H_(2)dot)can be rapidly obtained under room temperature and aqueous conditions using either sodium or ammonium hydroxide as the bases to deprotonate the *** use of metal bromides and tetrabutylammonium hydroxide(TBAOH)as the base allows the green synthesis of ionic metal-organic frameworks(MOFs),characterized by X-ray diffraction(XRD),Fourier transform infrared(FTIR),scanning electron microscopy(SEM),thermo gravimetric analysis(TgA),H-nuclear magnetic resonance(nMR),elemental analysis,and X-ray photoelectron spectroscopy(XPS),and employed in the pressure,solvent,and additive-free CO_(2)fixation to epoxides by its cycloaddition to epichlorohydrin,exhibiting a cooperative behavior between the MOF and the ionic liquid formed within the framework,achieving TOns of the confined tetrabutylammonium bromide(TBABr)up to 700.

Solvent-free aerobic oxidation of ethylbenzene over supported ni catalysts using molecular oxygen at atmospheric pressure

We investigated the aerobic oxidation of ethylbenzene in the absence of solvent or any additive carried out over ni on different types of supports namely SiO2, hydroxyapatite, SBA-15, and USY Zeolites. The oxidation of ethylbenzene activities was measured in a round bottom flask immersed in oil bath at known reaction temperature. The physicochemical characteristics of the catalysts were examined by BET surface area, XRD, FT-IR and the oxidation activities were correlated with the acidities of the catalysts obtained by TPD of nH3. It was observed that both hydroxyapatite and USY （13% na2O） supported ni catalysts displayed higher ethylbenzene conversion and 80% selectivity towards acetophenone.

Update on endoscopic ultrasound-guided liver biopsy

Endoscopic ultrasound guided liver biopsy(EUS-LB)has emerged as a minimally-invasive alternative to the traditional(percutaneous or transjugular)liver biopsy techniques for the diagnosis of liver parenchymal ***,EUS-LB combines the advantages of percutaneous and transjugular liver biopsy in addressing focused sampling in addition to measuring portal ***,EUS-LB facilitates access to both the lobes of the liver which is not considered with the traditional percutaneous liver *** studies have compared EUS-LB with conventional liver biopsy and reported comparable diagnostic yield,increased acquisition of complete portal tracts,and longer specimen length as compared to the traditional ***-LB is associated with lesser post-procedural pain and shorter recovery time,while providing lower risk of complications when compared to traditional liver *** in needle types,needle sizes and suction techniques have aimed at further optimizing the EUS-LB *** review article updates current literature with focus on the variations in the technique and equipment used for EUS-LB,and compares EUS-LB with traditional methods of liver biopsy.
