Rota.or cuff tea.s a.e frequent shoulder problems tha. a.e usua.ly dea.t with surgica. repa.r. Despite improved surgica. techniques, the tendon-to-bone hea.ing ra.e is unsa.isfa.tory due to difficulties in restoring the delica.e tra.sitiona. tissue between bone a.d tendon. It is essentia. to understa.d the molecular mecha.isms tha. determine this fa.lure. The study of the molecular environment during embryogenesis a.d during norma. hea.ing a.ter injury is key in devising stra.egies to get a.successful repa.r. Mesenchyma. stem cells(MSC) ca. differentia.e into different mesoderma. tissues a.d ha.e a.strong pa.a.rine, a.ti-inflamma.ory, immunoregulatory a.d a.giogenic potentia.. Stem cell thera.y is thus a.potentia.ly effective thera.y to enha.ce rota.or cuff hea.ing. Promising results ha.e been reported with the use of a.tologous MSC of different origins in a.ima. studies: they ha.e shown to ha.e better hea.ing properties, increa.ing the a.ount of fibroca.tilage forma.ion a.d improving the orienta.ion of fibroca.tilage fibers with less immunologic response a.d reduced lymphocyte infiltra.ion. a.l these cha.ges lea. to a. increa.e in biomecha.ica. strength. This effect is concentra.iondependent, only pa.tia.ly rcversible a.d the degree of inhibition increa.es with time. The synthesis of DNa.a.d RNa.is inhibited in pa.a.lel by increa.ing concentra.ions of the chemica.
