N-acetylserotonin alleviates retinal ischemia-reperfusion injury via HMGB1/RAGE/NF-κB pathway in rats

AIM:To observe the effects of N-acetylserotonin(naS)administration on retinal ischemia-reperfusion(RIR)injury in rats and explore the underlying mechanisms involving the high mobility group box 1(HMGB1)/receptor for advanced glycation end-products(RAGE)/nuclear factor-kappa B(NF-κB)signaling ***:A rat model of RIR was developed by increasing the pressure of the anterior chamber of the *** male Sprague Dawley were randomly divided into five groups:sham group(n=8),RIR group(n=28),RIR+naS group(n=28),RIR+FPS-ZM1 group(n=8)and RIR+naS+FPS-ZM1 group(n=8).The therapeutic effects of naS were examined by hematoxylin-eosin(H&E)staining,and retinal ganglion cells(RGCs)*** expression of interleukin 1 beta(IL-1β),HMGB1,RAGE,and nod-like receptor 3(NLRP3)proteins and the phosphorylation of nuclear factorkappa B(p-NF-κB)were analyzed by immunohistochemistry staining and Western blot *** expression of HMGB1 protein was also detected by enzyme-linked immunosorbent assay(ELISA).RESULTS:H&E staining results showed that naS significantly reduced retinal edema and increased the number of RGCs in RIR *** naS therapy,the HMGB1 and RAGE expression decreased significantly,and the activation of the NF-κB/NLRP3 pathway was antagonized along with the inhibition of p-NF-κB and NLRP3 protein ***,naS exhibited an anti-inflammatory effect by reducing IL-1β*** inhibitory of RAGE binding to HMGB1 by RAGE inhibitor FPS-ZM1 led to a significant decrease of p-NF-κB and NLRP3 expression,so as to the IL-1βexpression and retinal edema,accompanied by an increase of RGCs in RIR ***:naS may exhibit a neuroprotective effect against RIR via the HMGB1/RAGE/NF-κB signaling pathway,which may be a useful therapeutic target for retinal disease.

The microbial community and flavour substances of off-flavoured rice sour soup during storage

Fermented sour soup is a unique traditional sour food of the Miao and Dong ethnic groups in China. However， due to the commonly used traditional fermentation， the unstable fluctuation of microbial community often leads to some unpleasant inherent off-flavours in rice sour soup. In this study， the microbial composition and volatile flavour components of off-flavour and normal rice sour soup were examined by high-throughput sequencing (HTS) and headspace solid-phase microextraction coupled with gas chromatography-mass spectrometry (HS-SPME-GC-MS). Then， based on Pearson correlation coefficients， the correlations between fungi and bacteria and those between core microorganisms and key volatile flavour components were investigated. The dominant fungal genera included Candida， Dekkera， Pichia， Rhizopus， and Issatchenkia， whereas the dominant bacterial genera included Lactobacillus， Pectinatus， Bifidobacterium， Pseudomonas， and Acetobacter. Compared to normal rice sour soup， off-flavoured rice sour soup contained the significantly increased relative abundance of Candida and the significantly decreased relative abundance of Dkkera. In addition， 80 volatile flavour compounds detected in off-flavoured rice sour soup， mainly including esters， acids， and alcohols， were different from those detected in normal rice sour soup. Especially， some substances were only detected in off-flavoured rice sour soup， such as 1-hexanol (68-207 μg/kg)， butyric acid (20-91 μg/kg) and heptanoic acid (11-44 μg/kg)， which might be the main source of off-flavours. These results provide new ideas and strategies of removing or abating odour from fermented rice sour soup.

We revisit the γ-ray emission above 300 Me V towards the massive star-forming region of Orion B by adopting14 yr observations with the Fermi Large Area Telescope and utilizing the updated software *** extended γ-ray emission region around Orion B is resolved into two components(region Ⅰ and region Ⅱ).The γ-ray spectrum of region I agrees with the predicted γ-ray spectrum assuming the cosmic ray(CR)density is the same as that of Alpha Magnetic Spectrometer(AMS-02)measured ***γ-ray emissivity of region II appears to be deficit at low energy band(E<3 GeV).Through modeling we find that CR densities exhibit a significant deficit below 20 Ge V,which may be caused by a slow diffusion inside the dense *** is probably caused by an increased magnetic field whose strength increases with the gas density.

In vivo imaging reveals a synchronized correlation among neurotransmitter dynamics during propofol and sevoflurane anesthesia

General anesthesia is widely applied in clinical ***,the precise mechanism of loss of consciousness induced by general anesthetics remains ***,we measured the dynamics of five neurotransmitters,includingγ-aminobutyric acid,glutamate,norepinephrine,acetylcholine,and dopamine,in the medial prefrontal cortex and primary visual cortex of C57BL/6 mice through in vivo fiber photometry and genetically encoded neurotransmitter sensors under anesthesia to reveal the mechanism of general anesthesia from a neurotransmitter *** revealed that the concentrations of γ-aminobutyric acid,glutamate,norepinephrine,and acetylcholine increased in the cortex during propofol-induced loss of *** levels did not change following the hypnotic dose of propofol but increased significantly following surgical doses of propofol ***,the concentrations of the five neurotransmitters generally decreased during sevoflurane-induced loss of ***,the neurotransmitter dynamic networks were not synchronized in the non-anesthesia groups but were highly synchronized in the anesthetic *** findings suggest that neurotransmitter dynamic network synchronization may cause anesthetic-induced loss of consciousness.

Magnesium-based hydrides have been widely recognized as an appropriate choice for solid-state hydrogen ***,its undesirable thermodynamics and sluggish hydrogenation/dehydrogenation kinetics are major bottlenecks for its ***,a highly stable and highly dispersed Ni-based catalyst （Ni/Al2O3/GN） was fabricated to promote the hydrogen storage performance of MgH2via the electrostatic effect of NiAl-LDH/GN precursor with a co-calcination reduction ***2-5wt % Ni/Al2O3/GN exhibits excellent hydrogen storage performance,releasing about 5.7 wt % hydrogen in 3500 s at 250℃,and can reach a saturation hydrogen absorption of about 6.15 wt% in 3000 s at 100℃.Furthermore,it also shows low dehydrogenation apparent activation energy of 89.1 and 118.2 kJ·***,the catalyst ensures the stability of both the physical phase and structure during ball milling and cycling *** role of each phase in Ni/Al2O3/GN on the hydrogen storage performance of MgH2was also discussed through experiments and theoretical calculation,and the synergistic catalytic mechanism of Ni/Al2O3/GN was clearly *** work provides a unique perspective for the preparation of highly stable and highly dispersible catalysts.

MCSTransWnet:A new deep learning process for postoperative corneal topography prediction based on raw multimodal data from the Pentacam HR system

Variations, effectiveness and its associated factors of a nationwide web-based hypertension management training project in China: insights from a government-led campaign for 1.2 million lay health workers

OBJECTIVES To evaluate the effectiveness of a large-scale,web-based,in-service hypertension management training project among lay health workers(LHWs) at primary care health(PHC) settings in China,and to examine the factors contributing to the variations of *** We used data from a web-based national hypertension management training project implemented in 2018,it was designed to facilitate LHWs to learn,understand,and apply the relevant knowledge and skills in hypertension management through providing training courses by use of the web-based platform with unified *** LHWs were required to participate in the exams before and after training to acquire scores for the use of evaluating their performance of hypertension management *** first used descriptive analysis to present the variations of effectiveness in hypertension management knowledge among LHWs by important ***,we used multilevel logistic regression to examine the individual and regional factors contributing to the variations and quantify the magnitude of how these factors affected training *** There were 1,208,610 LHWs who completed training and were ***,the scores of LHWs increased significantly from 62.87 ± 21.14 out of 100 in the pre-test to 88.30 ± 11.31 in the post-test by 25.43(95% confidence interval[CI]:25.40-25.47).Training contents involved in antihypertensive medication showed the lowest score(54.36) in the pre-test and soared the most after training,up to 84.22 by 54.94%.Individual factors associated with disparities in the knowledge of hypertension management decreased substantially after training,which included sex,age,education,practice type,professional level,and hierarchy of working *** variations were shown at the provincial level,with the majority of them being explained by factors at the regional *** Accessible web-based training modality,government efforts,accompa

Immediate Effect of Electro-acupuncture on Endometrial Blood Flow in Patients with Recurrent Implantation Failure:A Randomized Controlled Trial
