A-to-I r.A editing is a r.cently discover.d pr.cess of post-tr.nscr.ption modification of pr.-mr.A by adenosine deamination that r.sults in the pr.duction of pr.-teins not inher.nt in the genome. The pr.sent study aimed to identify a r.le for.A-to-I r.A editing in the acute inflamma-tion. It was found that A-to-I edtiting activities of r.A edi-tases, ADAr., wer. upr.gulated in lung, spleen, thymus and lymph node of mouse dur.ng endotoxin stimulation. Impor.tantly, the number.of inosine in poly(A) r.A isolated fr.m mouse lung and spleen was significantly incr.ased in cor.e-lating with the induction of ADAr.?editing activity. The in vitr. synthesized r.A which did not contain inosine was edited by thymus extr.cts and the gener.tion of inosine was gr.atly incr.ased after.editing in LPS tr.ated thymus extr.ct. Take together. these data suggest that A-to-I r.A editing by ADAr. may play an impor.ant r.le in pathogenesis of in-flammation. The existence of high level of I-mr.A also sug-gests that mor. pr.tein isofor.s might be gener.ted fr.m a single gene via adenosine deamination by ADAr. dur.ng inflammator. str.ss.
