Blueberry anthocyanins extract attenuates oxidative stress and angiogenesis on an in vitro high glucose-induced retinopathy model through the miR-33/GLCCI1 axis

Background:Diabetes retinopathy(DR)is a complication of diabetes that affects patients’*** studies have found blueberry anthocyanins extract(BAE)can inhibit the progression of DR,but its mechanism is not completely ***:To study the role of BAE in diabetes retinopathy,we treated human retinal endothelial cells(HRCECs)with 30 mM high glucose to simulate the microenvironment of diabetes retinopathy and used BAE to intervene the in vitro high glucose-induced retinopathy *** cell viability and apoptosis rates were examined by Cell Counting Kit 8(CCK-8)assay and flow cytometry *** binding sites between miR-33 and glucocorticoid-induced transcript 1(GLCCI1)were assessed by luciferase reporter *** neovascularization and oxidative stress contribute to diabetic *** tubule formation assay was applied to detect the retinal *** oxidative stress in the HRCECs was manifested by the reactive oxygen species(ROS)level,the malondialdehyde(MDA)level,and the superoxide dismutase(SOD)***:Compared with HRCECs cells cultured under normal conditions,high glucose(HG)can induce oxidative stress in HRCRCs,specifically manifested in the increase of ROS and MDA levels,and the decrease of SOD *** relieved the tubule formation in n the *** also relieved the ROS and MDA levels and increased the SOD *** reporter assay revealed that GLCCI1 is a target molecule downstream of *** HRCEC,BAE significantly inhibited the expression of miR-33 induced by ***-33 mimic inhibited the BAE’s effects on oxidative stress and angiogenesis in an in vitro high glucose-induced retinopathy ***:BAE alleviated the oxidative stress and microangiogenesis of HRCEC by regulating the miR-33/GLCCI1 axis.

Flavone from Zhongjiefeng(Herba Sarcandrae Glabrae) inhibits platelet apoptosis in immune-induced bone marrow failure through mitochondrial pathway

OBjECTIVE: To investigate the effect of Flavone from Zhongjiefeng(Herba Sarcandrae Glabrae) on the platelet number in immune-induced bone marrow failure(BMF) and its mechanism of mitochondrial apoptotic ***: Immune-induced BMF model, established in mice, was randomly divided into four groups: normal control group without BMF, BMF control group, cyclosporine(CSA) group and flavone group(n = 10 in each group). Mice were given0.027 g/kg cyclosporine or 0.2 g/kg flavone lavage daily in either the cyclosporine or flavone group respectively. Platelet count, mitochondrial transmembrane potential(ΔΨm), cytochrome C(Cyt C), phosphatidylserine(PS), changes of calcium ion(Ca^(2+)),and protein expression of mitochondrial apoptotic pathway including B-cell lymphoma-2(bcl-2) Homologous Antagonist-Killer Protein(Bak), bcl-2-associated X protein(Bax), caspase-3, caspase-8, and caspase-9 were examined and ***: Compared with the normal control group, the BMF group had significantly lower levels of platelet count, ΔΨm, and expressions of caspase family proteins as well as higher levels of Cyt C, PS,Ca^(2+), and expressions of Bak and Bax(all P < 0.05).Compared with the BMF group, the CSA and flavone groups had significantly higher ΔΨm and expressions of caspase family proteins(all P < 0.05)whereas the levels of Cyt C, PS, Ca^(2+), and expressions of Bak and Bax were reduced(all P < 0.05).More importantly, the flavone group had higher levels of Cyt C, Ca^(2+)and expressions of Bak and Bax compared with the CSA group(all P < 0.05), while the levels of PS and caspase family proteins were reduced(all P < 0.05).CONCLUSION: Flavone from Zhongjiefeng(Herba Sarcandrae Glabrae) significantly increases the platelet number and prevents its apoptosis through mitochondrial pathway.

Dynamics of surface charge and electric field distributions on basin-type insulator in GIS/GIL due to voltage polarity reversal

In this study,a simulation model of surface charge accumulation has been *** model considers three accumulation ways,*** conduction within the gas,through insulator volume and along the insulator *** generation,diffusion,drift and recombination of charge carriers are also taken into *** on it,the influence of polarity reversal,reversal time on surface charge and electric field distribution on a basin-type insulator are *** polarity of the surface charges and the direction of the electric field change after the voltage polarity *** the preload voltage is equal to reversal voltage,the surface charge and the electric field distributions at steady state before and after voltage polarity reversal are all the same with opposite sign,and not affected by the reversal ***,the time to reach the steady state varies with different reversal *** steady-state surface charge and electric field increased with the rise of reversal *** transient normal and tangential electric field would not exceed the value of the steady state,which means voltage polarity reversal has no additional influence on insulation *** research can provide guidance to the design and manufacture of DC GIS/GIL.

Synthesis and Characterization of (E)-5-[2-(N-hexylcarbazolyl)vinyl] furan Derivatives with Acceptor Groups

A new series of (E)-5-[2-(N-hexylcarbazolyl)vinyl]furan chromophores with various accepters have been synthesized by the Knoevenagel condensation of (E)-5-[2-(N- hexylcarbazolyl) vinyl]-2-furaldehydes with malononitrile, 1,3-diethyl-2-thiobarbituric acid, or 3-phenyl-5-isoxazol one, respectively. They are characterized by H-1-NMR, FT- IR, UV-VIS, MS and elemental analysis, and have shown strong solvatochromism and high thermal stability.

The attenuation of oscillatory thermo-capillary convection in the oxide melt by a transverse magnetic field

The effect of a transverse magnetic field on the oscillatory thermocapillary con- vection in the NaBi(WO4)2 melt was studied by using the in-situ observation system. The oscillation was attenuated when the 60 mT magnetic field was applied, as shown by the decrease in the amplitude and the frequency. Furthermore, the os- cillation under smaller temperature difference was stabilized after the magnetic field was applied. The magnetic effect could be due to the Lorentz force generated by the interaction between motional ions and the vertical magnetic field. The ionic conductivities were measured to demonstrate the effect of the magnetic field. The solid ionic electrical conductivity increases with the temperature rise, and the melt ionic electrical conductivity was measured to be about 2.0×10-4 -1cm-1. Experi- mental results manifest that the effect of the magnetic field on anions and cations in the melt makes the flow change to the direction normal to the applied field, so the flow is more orderly and the oscillation is suppressed.

Design, synthesis and in vitro antitumor evaluation of novel diaryl urea derivatives bearing sulfonamide moiety

A novel series of diaryl urea derivatives bearing sulfonamide moiety have been designed and synthesized. Their in vitro antitumor effect against human cancer cell lines MX-1, A375, HepG2, Ketr3 and HT-29 was screened and evaluated by the standard MTT assay with sorafenib as the positive control. Some of the compounds showed significant inhibitory activity against multiple cell lines compared to sorafenib. In particular, 2,6-dimethyl-4-{6-[3-（4-chloro-3-（trifluoromethyl）phenyl）urealnaphthalen- 2-yllsulfonyl morpholine （10d） was found to be the most potent against A375, HepG2 and Ketr3 with ICs0 values of 0.65-0.97 μmol/L, which were 5-20-fold more potent than sorafenib. Compound 10d emerged as a valuable lead for further optimization.

An efficient method for the site-specific 99mTc labeling of nanobody

Recently, there has been a lot of interest by using nanobodies（heavy chain-only antibodies produced naturally from the Camelidae） as targeting molecules for molecular imaging, especially for the nuclear medicine imaging. A radiolabeled method that generates a homogeneous product is of utmost importance in radiotracer development for the nuclear medicine imaging. The conventional method for the radiolabeling of nanobodies is non-specifically, which conjugates the radioisotope chelating group to the side chain ε-amine group of lysine or sulfhydryl of cysteine of nanobodies, with a shortcoming of produce of the heterogeneous radiotracer. Here we describe a method for the site-specific radioisotope;99m;Tc labeling of nanobodies by transpeptidase Sortase A. The radiolabeling process includes two steps: first step, NH;2;-GGGGK（HYNIC）-COOH peptide（GGGGK = NH;2;-Gly-Gly-Gly-Gly-Lys-COOH, HYNIC =6-hydrazinonicotinyl） was labeled with;99m;Tc to obtain GGGGK-HYNIC-;99m;Tc; second step, Sortase A catalyzes the formation of a new peptide bond between the peptide motif LPETG（NH;2;-Leu-Pro-GluThr-Gly-COOH） expressed C-terminally on the nanobody and the N-terminal of GGGGK-HYNIC-;99m;*** a simple purification process, homogeneous single-conjugated and stable;99m;Tc-labeled nanobodies were obtained in >50% yield. This approach demonstrates that the Sortase A-mediated conjugation is a valuable strategy for the development of site-specifically;99m;Tc-labeled nanobodies.

Weeds are mainly spread by weed seeds being mixed with agricultural and forestry crop seeds,grain,animal hair,and other plant products,and disturb the growing environment of target plants such as crops and wild native *** accurate and efficient classification of weed seeds is important for the effective management and control of ***,classification remains mainly dependent on destructive sampling-based manual inspection,which has a high cost and rather low *** considered that this problem could be solved using a nondestructive intelligent image recognition ***,on the basis of the establishment of the image acquisition system for weed seeds,images of single weed seeds were rapidly and completely segmented,and a total of 47696 samples of 140 species of weed seeds and foreign materials ***,six popular and novel deep Convolutional Neural Network(CNN)models are compared to identify the best method for intelligently identifying 140 species of weed *** these samples,33600 samples are randomly selected as the training dataset for model training,and the remaining 14096 samples are used as the testing dataset for model *** and GoogLeNet emerged from the quantitative evaluation as the best *** has strong classification accuracy and efficiency(low time consumption),and GoogLeNet has the best classification accuracy.A suitable CNN model for weed seed classification could be selected according to specific identification accuracy requirements and time costs of *** research is beneficial for developing a detection system for weed seeds in various *** resolution of taxonomic issues and problems associated with the identi-fication of these weed seeds may allow for more effective management and control.

Dynamic Mechanical Behavior and Adiabatic Shear Bands of Ultrafine Grained Pure Zirconium

Dynamic compression tests were carried out to investigate dynamic mechanical behavior and adiabatic shear bands in ultrafine grained(UFG)pure zirconium prepared by equal channel angular pressing(ECAP)and rotary *** cylindrical specimens were deformed dynamically on the split Hopkinson pressure bar(SHPB)at different strain rates of 800 to 4000s^-1 at room *** temperature distribution of the shear bands was estimated on the basis of temperature rise of uniform plastic deformation stage and thermal diffusion *** results show that the true stress-true strain curves of UFG pure zirconium are concave upward trend of strain in range of 0.02-0.16 due to the effects of strain hardening,strain rate hardening and thermal *** formation of the adiabatic shear bands is the main reason of UFG pure zirconium failure.A large number of micro-voids are observed in the adiabatic shear bands,and the macroscopic cracks develop from the micro-voids *** fracture surface of UFG pure zirconium exhibits quasi cleavage fracture with the characteristic features of shear dimples and river *** highest temperature within the shear bands of UFG pure zirconium is about 592 K.

The acceleration saltation of the traditional S-type acceleration model in the speed planning of the NURBS curve will result in the vibration and flexible impact of the machine *** will affect the surface quality of the *** high speed smooth S-type acceleration and deceleration model deals with flexible impact,but the calculation is *** at the above problems,the traditional S-type acceleration and deceleration model is improved to make the jerk change linearly at a certain slope to reduce the flexible *** the speed planning,it is needed to find the arc length and curvature of each point on the NURBS curve with a tiny step,and to determine the speed sensitivity point on the curve *** to the speed sensitive point,the NURBS curve is *** attribute parameters of each section are determined by adaptive speed ***,the speed planning can be performed on the NURBS curve according to the speed characteristics *** simulation results show that the algorithm can effectively reduce the flexible impact,improve the machining precision and efficiency,and simplify the classification of speed characteristics.
