Role of TRIM33 in Wnt signaling during mesendoderm differentiation

Tripartite motif 33(TRIM33), a member of the transcription intermediate factor 1(TIF1) family of transcription cofactors,mediates transforming growth factor-beta(TGF-β) signaling through its PHD-Bromo cassette in mesendoderm differentiation during early mouse embryonic development. However, the role of the TRIM33 RING domain in embryonic differentiation is less clear. Here, we report that TRIM33 mediates Wnt signaling by directly regulating the expression of a specific subset of Wnt target genes, and this action is independent of its RING domain. We show that TRIM33 interacts with β-catenin, a central player in Wnt signaling in mouse embryonic stem cells(mESCs). In contrast to previous reports in cancer cell lines, the RING domain does not appear to function as the E3 ligase for β-catenin, since neither knockout nor overexpression of TRIM33 had an effect on β-catenin protein levels in mESCs. Furthermore, we show that although TRIM33 seems to be dispensable for Wnt signaling through a reporter assay, loss of TRIM33 significantly impairs the expression of a subset of Wnt target genes, including Mixl1,in a Wnt signaling-dependent manner. Together, our results indicate that TRIM33 regulates Wnt signaling independent of the E3 ligase activity of its RING domain for β-catenin in mESCs.

To improve the separation efficiency of air dense medium fluidized beds tor dry coal preparation, a gasvibro fluidized bed has been proposed in which magnetic powder is used as the heavy medium. Pressure fluctuations in the gas-vibro fluidized bed were investigated using time- and frequency-domain analysis methods. The relationship between pressure fluctuations, bubble behavior, and separation efficiency was established. The low amplitude of the standard deviation, the power spectral density （PSD）, the incoherent-output PSD, and the high amplitude of the coherent-output PSD, which corresponds to the bubble behavior in the bed, were improved for coal preparation. The coal ash content was reduced from 42.5570 to 16.54% by using the gas-vibro fluidized bed.

The direct generation of passively Q-switched lasers at a green wavelength has rarely been investigated in the *** this Letter,we demonstrate a passively Q-switched praseodymium-doped yttrium lithium fluoride green laser at 522 nm using CdTe/CdS quantum dots as a saturable absorber.A maximum average output power of 33.6 m W is achieved with the shortest pulse width of 840 *** corresponding pulse energy and peak power reached 0.18μJ and 0.21 W,*** the best of our knowledge,this is the first demonstration in regard to a quantum dots saturable absorber operating in the green spectral region.

Traditionally, complex engineering applications （CEAs）, which consist of numerous components （software） and require a large amount of computing resources, usu- ally run in dedicated clusters or high performance computing （HPC） centers. Nowadays, Cloud computing system with the ability of providing massive computing resources and cus- tomizable execution environment is becoming an attractive option for CEAs. As a new type on Cloud applications, CEA also brings the challenges of dealing with Cloud resources. In this paper, we provide a comprehensive survey of Cloud resource management research for CEAs. The survey puts forward two important questions： 1） what are the main chal- lenges for CEAs to run in Clouds？ and 2） what are the prior research topics addressing these challenges？ We summarize and highlight the main challenges and prior research topics. Our work can be probably helpful to those scientists and en- gineers who are interested in running CEAs in Cloud envi- ronment.

In this paper,an operation model for distribution systems with energy storage(ES)is proposed and solved with the aid of machine *** model considers ES applications with uncertainty *** also considers ES applications for economy and security *** the special features of ES operations under day-ahead decision mechanisms of distribution systems,an ES operation scheme is designed for transferring uncertainties to later hours through ES to ensure the secure operation of distribution *** a result,uncertainties from different time intervals are assembled and may counteract each other,thereby alleviating the *** different ES applications rely on ES flexibility(in terms of charging and discharging)and interact with each other,by coordinating different ES applications,the proposed operation model achieves efficient exploit of ES *** shorten the computation time,a long short-term memory recurrent neural network is used to determine the binary variables corresponding to ES *** proposed operation model then becomes a convex optimization problem and is solved ***,the solving efficiency is greatly improved while ensuring the satisfactory use of ES flexibility in distribution system operation.

Deformation mechanisms of nanograined and submicron-grained pure cobalt processed by means of high strain rate shear deformation at cryogenic temperatures were *** analysis revealed a transition of governing deformation mechanism from deformation twinning and dislocation slip in submicron-grains to and dislocations slip in *** tests illustrated that the Hall-Petch relation slope changes consequently with the transition of deformation mechanism.

Up to 1.5wt%of Cr(Ⅲ)salts(CrCl_(3),and Cr_(2)O_(3))and Cr(Ⅵ)salts(Na_(2)CrO_(4),and CaCr_(2)O_(7))were incorporated into red mud-based geopolymers,*** solidification/stabilization,compressive strength,and durability of the Cr-containing geopolymers were *** experimental results indicate that the red mud-based geopolymer could effectively solidify/stabilize different types of Cr salts with solidification/stabilization rates of above 99.61%.Geopolymers are environmentally safe when the dosage of CaCr_(2)O_(7)is≤1.0wt%,or the dosage of CrCl_(3),Cr_(2)O_(3),and Na_(2)CrO_(4)is≤1.5wt%,*** effects of Cr salts on the compressive strength varies with the type and content of Cr *** freeze-thaw cycle is more destructive to geopolymer properties than sulfate attack or acid rain *** solidification/stabilization of Cr is mainly attributed to the following reasons:a)The chemical binding of Cr is related to the formation of Cr-containing hydrates(eg,magnesiochromite((Mg,Fe)(Cr,Al)_(2)O_(4)))and doping into N-A-S-H gel and C-A-S-H gel framework;b)The physical effect is related to the encapsulation by the hydration products(e g,N-A-S-H gel and C-A-S-H gel).This study provides a reference for the treatment of hazardous Cr-containing wastes by solid waste-based geopolymers.

Fatty acyl reductases(FARs)are key enzymes that participate in sex pheromone biosynthesis by reducing fatty acids to fatty *** typically harbor numerous FAR gene family *** FAR genes are involved in the biosynthesis of sex pheromones in moths,the key FAR gene of Spodoptera litura remains *** this work,we predicted 30 FAR genes from the *** genome and identified a domain duplication within gene SlitFAR3,which exhibited high and preferential expression in the sexually mature female pheromone glands(PGs)and a rhythmic expression pattern during the scotophase of sex pheromone *** molecular docking of SlitFAR3,as predicted using a 3D model,revealed a co-factor NADPH binding cavity and 2 substrate binding *** expression in yeast cells combined with comprehensive gas chromatography indicated that the SlitFAR3 gene could produce fatty alcohol *** study is the first to focus on the special phenomenon of FAR domain duplication,which will advance our understanding of biosynthesis-related genes from the perspective of evolutionary biology.

Titanium dioxide(TiO_(2))was recently employed to apply onto road surfaces to degrade the harmful compounds from vehicle ***,it remains a challenging task to find a highly compatible pavement type for TiO_(2)application to achieve durable and efficient air-purifying *** study proposed to coat TiO_(2)particles onto semi-flexible pavement surface and tried to investigate an optimum coating *** coating methods,including direct mixing TiO_(2)(MT)with asphalt mixture,spraying dry TiO_(2)(ST)coating and watersolution-based TiO_(2)(WT)coating on semi-flexible pavement *** achieve this objective,semi-flexible samples were prepared to evaluate and compare the performances of three coating methods by employing resistance to wearing,NO removal efficiency tests and residual texture depth *** was found that the ST method not only provided better NO degrading efficiency but also improved the resistance to wearing than the other two methods.
