As sessile organisms, plants have evolved with complex signaling networks to cope with the constantly changing environment. Receptor-like kinases （RLKs） and calcium both play important roles during the early stage of envi- ronmental responses in plants. RLKs are trans-membrane receptor kinases that sense extracellular signals, such as hormones, peptides, and pathogens, and trigger sig- nal transduction pathways leading to cellular responses （Osakabe et al., 2013）.

Excess phosphate(Pi)is stored into the vacuole through Pi transporters so that cytoplasmic Pi levels remain stable in plant *** hypothesized that the vacuolar Pi transporters may harbor a Pi-sensing mechanism so that they are activated to deliver Pi into the vacuole only when cytosolic Pi reaches a threshold high *** tested this hypothesis using Vacuolar Phosphate Transporter 1(VPT1),a SPX domain-containing vacuolar Pi transporter,as a *** studies have defined SPX as a Pi-sensing module that binds inositol polyphosphate signaling molecules(InsPs)produced at high cellular Pi *** showed here that Pi-deficient conditions or mutation of the SPX domain severely impaired the transport activity of *** further identified an auto-inhibitory domain in VPT1 that suppresses its transport *** together the results from detailed structure-function analyses,our study suggests that VPT1 is in the auto-inhibitory state when Pi status is low,whereas at high cellular Pi status InsPs are produced and bind SPX domain to switch on VPT1 activity to deliver Pi into the *** thus provides an auto-regulatory mechanism for VPT1-mediated Pi sensing and homeostasis in plant cells.

Ground state cooling of massive mechanical objects remains a difficult task restricted by the unresolved mechanical sidebands. We propose an optomechanically-induced-transparency cooling scheme to achieve ground state cooling of mechanical motion without the resolved sideband condition in a pure optomechanical system with two mechanical modes coupled to the same optical cavity mode. We show that ground state cooling is achievable for sideband resolution ωm/k as low as - 0.003. This provides a new route for quantum manipulation of massive macroscopic devices and high-precision measurements.

An introduced turbot population was used to establish families and to estimate genetic parameters of the offspring. However, there is a lack of pedigree information, and common environmental effects can be introduced when each full-sib family is raised in a single tank. Therefore, in the genetic evaluation, SSRs(simple sequence repeats) were used to reconstruct the pedigree and to calculate molecular relatedness between individuals, and the early mixed-family culture model was used to remove the impact of the common environmental effects. After100 d of early mixed culture, twenty SSRs were used to cluster 20 families and to calculate paired molecular relationships(n=880). Additive genetic matrices were constructed using molecular relatedness(MR) and pedigree reconstruction(PR) and were then applied to the same animal model to estimate genetic parameters. Based on PR, the heritabilities for body weight and body length were 0.214±0.124 and 0.117±0.141, and based on MR they were 0.101±0.031 and 0.102±0.034, respectively. Cross validation showed that the accuracies of the estimated breeding values based on MR(body weight and body length of 0.717±0.045 and 0.629±0.141, respectively) were higher than those of PR(body weight and body length of 0.692±0.052 and 0.615±0.060, respectively). The MR method ensure availability of all genotyped selection candidates, thereby improving the accuracy of the breeding value estimation.

ABSTRACT Calcium, as a ubiquitous second messenger, plays essential roles in tip-growing cells, such as animal neu-rons, plant pollen tubes, and root hairs. However, little is known concerning the regulatory mechanisms that code anddecode Ca2＋ signals in plants. The evidence presented here indicates that a calcium-dependent protein kinase, CPK32,controls polar growth of pollen tubes. Overexpression of CPK32 disrupted the polar growth along with excessive Ca2＋accumulation in the tip. A search of downstream effector molecules for CPK32 led to identification of a cyclic nucleotide-gated channel, CNGC18, as an interacting partner for CPK32. Co-expression of CPK32 and CNGC18 resulted in activationof CNGC18 in Xenopus oocytes where expression of CNGC18 alone did not exhibit significant calcium channel *** of CNGC18 produced a growth arrest phenotype coupled with accumulation of calcium in the tip, simi-lar to that induced by CPK32 overexpression. Co-expression of CPK32 and CNGC18 had a synergistic effect leading tomore severe depolarization of pollen tube growth. These results provide a potential feed-forward mechanism in whichcalcium-activated CPK32 activates CNGC18, further promoting calcium entry during the elevation phase of Ca2＋ oscilla-tions in the polar growth of pollen tubes.

Genetic Parameters of Body Length and Response to Selection for Growth Across Four Generations of Artemia sinica

To investigate the genetic components of growth in the brine shrimp Artemia sinica, we estimated the genetic parameters of body length and the response to selection using a fully pedigreed population of A. sinica. The base population was generated from four wild founder populations. We tested 4160 offspring in 360 families over four generations for growth and survival performance. Across four generations, we produced full-and half-sib families with nested mating, where two dams were mated to the same sire. Individual body length was measured for each nauplius at day 20 post-hatching. Heritability of body length was estimated across four generations with the restricted maximum likelihood method. The heritability of body length in A. sinica was low(0.14 ± 0.05), and the common environmental effect was 0.14 ± 0.02. We estimated the response to selection for body length by calculating the difference in the mean breeding values between different generations. The accumulated genetic gain in body length was 278.94 μm after three generations of selection. This low response to selection was probably caused by the low heritability of body length, small sample size, and the low selection intensity(50%). The results suggest that A. sinica selective breeding programs must be changed to generate any substantial, sustainable genetic increases in body length. We suggest that optimal genetic gains could be achieved by introducing wild strains into the nuclear breeding population to increase genetic variation, and by increasing the size of the breeding population to allow for increased selection intensity.

ABA Signaling in Guard Cells Entails a Dynamic Protein-Protein Interaction Relay from the PYL-RCAR Family Receptors to Ion Channels

Plant hormone abscisic acid （ABA） serves as an integrator of environmental stresses such as drought to trig-ger stomatal closure by regulating specific ion channels in guard cells. We previously reported that SLAC1, an outward anion channel required for stomatal closure, was regulated via reversible protein phosphorylation events involving ABA signaling components, including protein phosphatase 2C members and a SnRK2-type kinase （OST1）. In this study, we reconstituted the ABA signaling pathway as a protein-protein interaction relay from the PYL/RCAR-type receptors, to the PP2C-SnRK2 phosphatase-kinase pairs, to the ion channel SLAC1. The ABA receptors interacted with and inhibited PP2C phosphatase activity against the SnRK2-type kinase, releasing active SnRK2 kinase to phosphorylate, and activate the SLAC1 channel, leading to reduced guard cell turgor and stomatal closure. Both yeast two-hybrid and bimolecular fluorescence complementation assays were used to verify the interactions among the components in the pathway. These biochemical assays demonstrated activity modifications of phosphatases and kinases by their interaction partners. The SLAC1 channel activity was used as an endpoint readout for the strength of the signaling pathway, depending on the presence of different combinations of signaling components. Further study using transgenic plants overexpressing one of the ABA receptors demonstrated that changing the relative level of interacting partners would change ABA sensitivity.

Establishment of microsatellite-based triplex PCR for parentage analysis of Chinese shrimp Fenneropenaeus chinensis

Through exploring the microsatellite primers from the random genome sequences of Chinese shrimp （Fenneropenaeus chinensis）, some microsatellite primers were obtained with rich polymorphic genetic information, and a triplex PCR was established using three primers （RS1101, RS0683 and H081 primers）. By adjusting the final concentration of Mg^2＋, dNTP and primers, and using a touch-town PCR program, the optimum amplification parameters of PCR system were obtained, which could successfully amplify the three primers in a PCR reaction. In the denatured PAGE gel, the amplified DNA fragments of three primers RS1 101,RS0683 and H081 could be easily identified each other. For the triplex PCR system, the PPE （probabilities of paternity exclusion） is 0.967 9,and the DP （discrimination power） is 0.999 *** the triplex PCR to test ten individuals of a parentage and their parents, an individual was excluded from the parentage in all of the three microsatellite loci, which might be mixed into the parentage for some unknown reason such as factitious misplay. The triplex PCR will be of great practical value in identifying the parentages of F. chinensis.
