Ethyl Lithospermate Reduces Lipopolysaccharide-Induced Inflammation through Inhibiting NF-κB and STAT3 Pathways in RAW 264.7 Cells and Zebrafish

Objective:To explore the anti-inflammatory effects of ethyl lithospermate in lipopolysaccharide(LPS)-stimulated RAW 264.7 murine-derived macrophages and zebrafish,and its underlying mechanisms.Methods:3-[4,5-dimethylthiazol-2-yl]-2,5-diphenyltetrazoliumbromide(MTT)assays were performed to investigate the toxicity of ethyl lithospermate at different concentrations(12.5–100μmol/L)in RAW 264.7 cells.The cells were stimulated with LPS(100 ng/mL)for 12 h to establish an inflammation model in vitro,the production of proinflammatory cytokines interleukin(IL)-6 and tumor necrosis factorα(TNF-α)were assessed by enzyme linked immunosorbent assay(ELISA).Western blot was used to ascertain the protein expressions of signal transducer and activator of transcription 3(STAT3),nuclear factor kappa B(NF-κB)p65,phospho-STAT3(p-STAT3,Tyr705),inhibitor of NF-κB(IκB)α,and phospho-IκBα(p-IκBα,Ser32),and confocal imaging was used to identify the nuclear translocation of NF-κB p65 and p-STAT3(Tyr705).Additionally,the yolk sacs of zebrafish(3 days post fertilization)were injected with 2 nL LPS(0.5 mg/mL)to induce an inflammation model in vivo.Survival analysis,hematoxylin-eosin(HE)staining,observation of neutrophil migration,and quantitative real-time polymerase chain reaction(qR T-PCR)were used to further study the anti-inflammatory effects of ethyl lithospermate and its probable mechanisms in vivo.Results:The non-toxic concentrations of ethyl lithospermate have been found to range from 12.5 to 100μmol/L.Ethyl lithospermate inhibited the release of IL-6 and TNF-α(P<0.05 or P<0.01),decreased IκBαdegradation and phosphorylation(P<0.05)as well as the nuclear translocation of NF-κB p65 and p-STAT3(Tyr705)in LPS-induced RAW 264.7 cells(P<0.01).Ethyl lithospermate also decreased inflammatory cells infiltration and neutrophil migration while increasing the survival rate of LPS-stimulated zebrafish(P<0.05 or P<0.01).In addition,ethyl lithospermate also inhibited the mR NA expression levels of of IL-6,TNF-α,IκBα,STAT3,and NF-κB in LPS-stimulated

Lipopolysaccharide(LPS)-induced inflammation causes massive threatening diseases,such as sepsis,acute lung injury and multiple organ dysfunction syndrome.Efficient treatment to prevent inflammation is crucial in LPS-induced inflammatory diseases.Heat-clearing Chinese medicines(CMs)have been used to ameliorate LPS-induced inflammation in China for centuries.Heat-clearing CMs regulate inflammatory pathways,thereby inhibiting the release of inflammatory factors.This review aimed to introduce promising heat-clearing CMs countering LPS-induced inflammation in the last 5 years,exploring the underlying molecular mechanisms.

Collective Molecular Machines: Multidimensionality and Reconfigurability

Molecular machines are key to cellular activity where they are involved in converting chemical and light energy into efficient mechanical work.During the last 60 years,designing molecular structures capable of generating unidirectional mechanical motion at the nanoscale has been the topic of intense research.Effective progress has been made,attributed to advances in various fields such as supramolecular chemistry,biology and nanotechnology,and informatics.However,individual molecular machines are only capable of producing nanometer work and generally have only a single functionality.In order to address these problems,collective behaviors realized by integrating several or more of these individual mechanical units in space and time have become a new paradigm.In this review,we comprehensively discuss recent developments in the collective behaviors of molecular machines.In particular,collective behavior is divided into two paradigms.One is the appropriate integration of molecular machines to efficiently amplify molecular motions and deformations to construct novel functional materials.The other is the construction of swarming modes at the supramolecular level to perform nanoscale or microscale operations.We discuss design strategies for both modes and focus on the modulation of features and properties.Subsequently,in order to address existing challenges,the idea of transferring experience gained in the field of micro/nano robotics is presented,offering prospects for future developments in the collective behavior of molecular machines.

Pig H3K4me3,H3K27ac,and gene expression profiles reveal reproductive tissue-specific activity of transposable elements

Regulatory sequences and transposable elements(TEs)account for a large proportion of the genomic sequences of species;however,their roles in gene transcription,especially tissue-specific expression,remain largely unknown.Pigs serve as an excellent animal model for studying genomic sequence biology due to the extensive diversity among their wild and domesticated populations.Here,we conducted an integrated analysis using H3K27ac ChIP-seq,H3K4me3 ChIP-seq,and RNA-seq data from 10 different tissues of seven fetuses and eight closely related adult pigs.We aimed to annotate the regulatory elements and TEs to elucidate their associations with histone modifications and mRNA expression across different tissues and developmental stages.Based on correlation analysis between mRNA expression and H3K27ac and H3K4me3 peak activity,results indicated that H3K27ac exhibited stronger associations with gene expression than H3K4me3.Furthermore,1.45%of TEs overlapped with either the H3K27ac or H3K4me3 peaks,with the majority displaying tissue-specific activity.Notably,a TE subfamily(LTR4C_SS),containing binding motifs for SIX1 and SIX4,showed specific enrichment in the H3K27ac peaks of the adult and fetal ovaries.RNA-seq analysis also revealed widespread expression of TEs in the exons or promoters of genes,including 4688 TE-containing transcripts with distinct development stage-specific and tissue-specific expression.Of note,1967 TE-containing transcripts were enriched in the testes.We identified a long terminal repeat(LTR),MLT1F1,acting as a testis-specific alternative promoter in SRPK2(a cell cycle-related protein kinase)in our pig dataset.This element was also conserved in humans and mice,suggesting either an ancient integration of TEs in genes specifically expressed in the testes or parallel evolutionary patterns.Collectively,our findings demonstrate that TEs are deeply embedded in the genome and exhibit important tissue-specific biological functions,particularly in the reproductive organs.

Optical grade (Gd_(0.95-x)lu_(x)Eu_(0.05))_(3)Al_(5)O_(12) ceramics with near-zero optical loss: Effects of lu^(3+) doping on structural feature, microstructure evolution, and far-red luminescence

A chemical co-precipitation strategy was employed to synthesize a series of(Gd_(0.95−x)lu_(x)Eu_(0.05))_(3)Al_(5)O_(12)(x=0.1−0.95)powder phosphors,followed by vacuum sintering to achieve transparent garnet ceramic phosphors.The density functional theory indicated lu_(3)Al_(5)O_(12) was formed in priority compared with Gd_(3)Al_(5)O_(12) during solid-phase reaction.Upon high-temperature sintering,the lu^(3+)substitution for Gd^(3+)suppressed point mass diffusion leading to a smaller grain size.The in-line transmittances of bulk specimens with x=0.1,0.3,0.5,0.7,and 0.95 nm were~83.5%,80.1%,68.8%,73.7%,and 82.2%at 710 nm(Eu^(3+)emission center),respectively,among which the sample of x=0.1 exhibited the optical grade with near-zero optical loss in agreement with the defect-free single crystal(~100%of the theoretical transmittance).The resulting particle and ceramic materials both presented characteristic Eu^(3+)emission arising from 5D0→7FJ(J=1−4)transition,where the dominant far-red emission at~710 nm arising from ^(5)D_(0)→^(7)F_(4) transition overlapped with the absorption of phytochrome(PFR).The photoluminescence excitation and photoluminescence intensities of(Gd_(0.95−x)lu_(x)Eu_(0.05))_(3)Al_(5)O_(12) powders and ceramics generally increased at a higher Gd^(3+)/lu^(3+)ratio.lu^(3+)dopants delayed the fluorescence lifetime while the bulk samples had shorter lifetime compared to the particle counterparts.The transparent(Gd_(0.85)lu_(0.1)Eu_(0.05))3Al_(5)O_(12) ceramic phosphor exhibited good thermal stability with a high thermal quenching temperature above 533 K.The designed ceramic phosphor-converted light-emitting diode had a saturation injection current of 435 mA and a current-dependent color rendering index.More importantly,our report marked the developmental stage of transparent ceramic materials towards zero optical loss.

Catalytic four-component radical carbonylation of unactivated alkenes has recently been recognized as a robust protocol for rapid construction of various structurally diverse carbonyl compounds.Given the significance of fluorine-containing groups,this reaction class has been extensively applied to assembly of a variety of perfluoroalkyl carboxylic acid derivatives by transition metal catalysis.Herein,we report a visible-light-driven radical relay 1,2-perfluoroalkylation aminocarbonylation of unactivated alkenes using CO gas as carbonyl source and 4CziPN as organic photocatalyst.A wide range of alkenes and amines were well tolerated,providing the valuableβ-perfluoroalkylated amides with generally good yields and high chemoselectivity.

Early prediction and prevention of infected pancreatic necrosis

Approximately 20%-30%of patients with acute necrotizing pancreatitis develop infected pancreatic necrosis(IPN),a highly morbid and potentially lethal complication.Early identification of patients at high risk of IPN may facilitate appropriate preventive measures to improve clinical outcomes.In the past two decades,several markers and predictive tools have been proposed and evaluated for this purpose.Conventional biomarkers like C-reactive protein,procalcitonin,lymphocyte count,interleukin-6,and interleukin-8,and newly developed biomarkers like angiopoietin-2 all showed significant association with IPN.On the other hand,scoring systems like the Acute Physiology and Chronic Health Evaluation II and Pancreatitis Activity Scoring System have also been tested,and the results showed that they may provide better accuracy.For early prevention of IPN,several new therapies were tested,including early enteral nutrition,anti-biotics,probiotics,immune enhancement,etc.,but the results varied.Taken together,several evidence-supported predictive markers and scoring systems are readily available for predicting IPN.However,effective treatments to reduce the incidence of IPN are still lacking apart from early enteral nutrition.In this editorial,we summarize evidence concerning early prediction and prevention of IPN,providing insights into future practice and study design.A more homo-geneous patient population with reliable risk-stratification tools may help find effective treatments to reduce the risk of IPN,thereby achieving individualized treatment.

Structural transformation from Waugh-type to Keggin-type polyoxomolybdate-based crystalline material for photo/electrocatalysis

An inorganicDorganic hybrid crystalline material (Si Mo12O40)[Cu（2,2'-bipy）2]2·2H2O （Complex 1,2,2'-bipy=2,2'-bipyridine） was synthesized for the first time by using 2,2'-bipy and transition metal copper to convert Waugh-type polyoxometalate[Mn Mo9O32]6-into a Keggin polyoxometalate structure[Si Mo12O40]4-under hydrothermal conditions in glass vials.Single crystal tests and a series of characterizations were carried out on Complex 1.The structure of Complex 1 is composed of[Si Mo12O40]4-and[CuII（2,2'-bipy）2]2+.The five-coordinated Cu is connected to two 2,2'-bipy through CuDN bonds,forming an approximately square structure.Astonishingly,Complex 1 exhibited good photocatalytic performance for methylene blue degradation and electrocatalytic nitrite reduction properties simultaneously.

Tuning mechanical behaviors of highly entangled hydrogels with the random distribution of mobile entanglements

Highly entangled hydrogels exhibit excellent mechanical properties,including high toughness,high stretchability,and low hysteresis.By considering the evolution of randomly distributed entanglements within the polymer network upon mechanical stretches,we develop a constitutive theory to describe the large stretch behaviors of these hydrogels.In the theory,we utilize a representative volume element(RVE)in the shape of a cube,within which there exists an averaged chain segment along each edge and a mobile entanglement at each corner.By employing an explicit method,we decouple the elasticity of the hydrogels from the sliding motion of their entanglements,and derive the stress-stretch relations for these hydrogels.The present theoretical analysis is in agreement with experiment,and highlights the significant influence of the entanglement distribution within the hydrogels on their elasticity.We also implement the present developed constitutive theory into a commercial finite element software,and the subsequent simulations demonstrate that the exact distribution of entanglements strongly affects the mechanical behaviors of the structures of these hydrogels.Overall,the present theory provides valuable insights into the deformation mechanism of highly entangled hydrogels,and can aid in the design of these hydrogels with enhanced performance.

Integrating machine learning and CALPHAD method for exploring low-modulus near-β-Ti alloys

Traditional theoretical and empirical calculation methods can guide the design of β-and metastable β-alloys for bio-titanium. However, it is still difficult to obtain novel near-β-Ti alloys with low modulus. This study developed a method that combines machine learning with calculation of phase diagrams(CALPHAD) to facilitate the design of near-β-Ti alloys. An elastic modulus database of Ti–Nb–Zr–Mo–Ta–Sn system was constructed first, and then three features(the electron to atom ratio, mean absolute deviation of atom mass, and mean electronegativity) were selected as the key factors of modulus by performing a three-step feature selection. With these features, a highly accurate model was built for predicting the modulus of near-β-Ti alloys. To further ensure the accuracy of modulus prediction, machine learning with the elastic constants calculated was leveraged by CALPHAD database. The root mean square error of the well-trained model can be as low as 6.75 GPa. Guided by the prediction of machine learning and CALPHAD, three novel near-β-Ti alloys with elastic modulus below 50 GPa were successfully designed in this study. The best candidate alloy(Ti–26Nb–4Zr–4Sn–1Mo–Ta) exhibits an ultra-low modulus(36.6 GPa) after cold rolling with a thickness reduction of 20%. Our method can greatly save time and resources in the development of novel Ti alloys, and experimental verifications have demonstrated the reliability of this method.
