目的基于磷脂酰肌醇3激酶(PI3K)/蛋白激酶B(Akt)/核因子κB(NF-κB)信号通路探讨槲皮素对心肾综合征(CRS)大鼠心肾功能的保护作用及机制。方法取SD大鼠,采用左冠状动脉前降支结扎结合肾急性缺血再灌注的方法构建CRS模型,将造模成功的大鼠随机分为模型组、槲皮素低剂量组(35 mg/kg)、槲皮素高剂量组(70 mg/kg)、槲皮素高剂量+740Y-P组(70 mg/kg槲皮素+3.5 mg/kg PI3K/Akt/NF-κB信号通路激活剂740Y-P),每组12只;另取12只正常大鼠设为假手术组。每天给药1次,连续14 d。给药结束后,检测大鼠心功能指标[左心室射血分数(LVEF)、舒张末期容积(EDV)、等容舒张时间(IVRT)]与肾功能指标[血尿素氮(BUN)、血肌酐(Scr)、24 h尿蛋白];观察大鼠心肾组织纤维化情况;检测大鼠血清与心肾组织中炎症指标[白细胞介素1β(IL-1β)、肿瘤坏死因子α(TNF-α)]水平及心肾组织中PI3K/Akt/NF-κB信号通路相关蛋白表达。结果与假手术组比较,模型组大鼠BUN、24 h尿蛋白、Scr水平,心肾组织的胶原容积分数,血清与心肾组织中IL-1β、TNF-α水平,心肾组织中PI3K、Akt、NF-κB p65蛋白磷酸化水平均显著升高(P<0.05),LVEF、IVRT、EDV均显著降低(P<0.05)。与模型组比较,槲皮素低、高剂量组大鼠上述指标均显著改善(P<0.05),且高剂量组的改善效果更好(P<0.05)。740Y-P逆转了高剂量槲皮素对各指标的改善作用(P<0.05)。结论槲皮素可减轻CRS大鼠心肾纤维化和功能损伤,其作用机制可能与抑制PI3K/Akt/NF-κB信号通路激活有关。

Black water aggregation （BWA） in Taihu Lake is a disaster for the lake environment. It is a phenomenon resulting from water environmental deterioration and eutrophication caused by accumulation of pollutants in the lake, according to research on the water quality, pollutants of BWA, and occurrence mechanisms of BWA. Dead algae are the material base of BWA, the polluted sediment is an important factor for the formation of BWA, and hydrological and meteorological conditions such as sun light, air temperature, wind speed, and water flow are the other factors that may lead to the formation of BWA. Thioether substances such as dimethyl trisulfide are the representative pollutants of BWA. Parameters such as chlorophyll-a, DO, pH, and water temperature are sensitive indicators of BWA. Measures such as algae collection, ecological dredging, pollution control, and water diversion from the Yangtze River to the lake, are effective, and strengthening aeration is an emergency measure to control BWA.

Based on a genetically modified radioresistant bacteria Deinococcus radiodurans, we constructed a real time whole cell biosensor to monitor radioactivity and genotoxicity in highly radioactive environ-ment. The enhanced green fluorescence protein (eGFP) was fused to the promoter of the crucial DNA damage-inducible recA gene from D. radiodurans, and the consequent DNA fragment (PrecA-egfp) car-ried by plasmid was introduced into D. radiodurans R1 strain to obtain the biosensor strain DRG300. This engineered strain can express eGFP protein and generate fluorescence in induction of the recA gene promoter. Based on the correlation between fluorescence intensity and protein expression level in live D. radiodurans cells, we discovered that the fluorescence induction of strain DRG300 responds in a remarkable dose-dependent manner when treated with DNA damage sources such as gamma radiation and mitomycin C. It is encouraging to find the widely detective range and high sensitivity of this re-constructed strain comparing with other whole cell biosensors in former reports. These results suggest that the strain DRG300 is a potential whole cell biosensor to construct a detective system to monitor the biological hazards of radioactive and toxic pollutants in environment in real time.

Based on the transmission line theory of the buried metallic structures, the concerned harmful effects to the buried oil and natural gas pipelines caused by the power line short-circuit fault are further discussed. A closed-form expression of the induced voltage caused by the short-circuit fault of the ultra-high voltage power （UHVAC） transmission line is given. The transmission line model of the buried pipeline is set up and a set of formulas for calculating induced voltage on the pipeline and the parameters of the buried pipeline in actual environment condition are given. At last, the characteristic of the harm^hl effects on the buried pipeline from the power line short-circuit fault are discussed.

目的研究抑制Ras同源基因（Ras homolog gene, Rho）/Rho相关卷曲螺旋形成蛋白激酶（Rhoassociated coiled-coil forming protein kinase, ROCK）通路对心肌素（myocardin, MYOCD）参与的气道平滑肌细胞增殖与迁移的影响及分子机制。方法体外构建腺病毒载体Ad-ZsGreen-shRNA-hROCK1感染人气道平滑肌细胞，将细胞随机分为4组：ROCK1基因沉默对照（shNC）组、shNC+花生四烯酸（arachidonic acid, AA;Rho/ROCK通路激活剂）组、ROCK1基因沉默（shROCK1）组、shROCK1+AA组（各组n=3）。采用实时荧光定量聚合酶链反应法、Western blot法检测ROCK1、MYOCD mRNA和蛋白表达水平，ELISA法检测球状肌动蛋白和丝状肌动蛋白水平，免疫荧光染色法及细胞划痕实验检测细胞增殖及迁移情况。结果与shNC+AA组相比，shROCK1+AA组ROCK1、MYOCD mRNA和蛋白表达水平降低，球状肌动蛋白、丝状肌动蛋白表达降低，细胞增殖、细胞迁移能力降低（P<0.05）。结论抑制Rho/ROCK通路致气道平滑肌细胞增殖及迁移能力受抑，其机制可能与MYOCD的下调有关。[中国当代儿科杂志，2024,26 (9):974-981]

Biodegradation of Complex Bacteria on Phenolic Derivatives in River Water

Objective To isolate, incubate, and identify 4-chlorophenol-degrading complex bacteria, determine the tolerance of these bacteria to phenolic derivatives and study their synergetic metabolism as well as the aboriginal microbes and co-metabolic degradation of mixed chlorophenols in river water. Methods Microbial community of complex bacteria was identified by plate culture observation techniques and Gram stain method. Bacterial growth inhibition test was used to determine the tolerance of complex bacteria to toxicants. Biodegradability of phenolic derivatives was determined by adding 4-chlorophenol-degrading bacteria in river water. Results The complex bacteria were identified as Mycopiana, Alcaligenes, Pseudomonas, and Flavobacterium. The domesticated complex bacteria were more tolerant to phenolic derivatives than the aboriginal bacteria from Qinhuai River. The biodegradability of chlorophenols, dihydroxybenzenes and nitrophenols under various aquatic conditions was determined and compared. The complex bacteria exhibited a higher metabolic efficiency on chemicals than the aboriginal microbes, and the final removal rate of phenolic derivatives was increased at least by 55% when the complex bacteria were added into river water. The metabolic relationship between dominant mixed bacteria and river bacteria was studied. Conclusion The complex bacteria domesticated by 4-chlorophenol can grow and be metabolized to take other chlorophenols, dihydroxybenzenes and nitrophenols as the sole carbon and energy source. There is a synergetic metabolism of most compounds between the aboriginal microbes in river water and the domesticated complex bacteria, 4- chlorophenol-degrading bacteria can co-metabolize various chlorophenols in fiver water.
