目的对1个Stargardt病家系进行临床特征与遗传学分析,并探讨三磷酸腺苷结合盒家族亚科A成员4(ABCA4)基因突变在Stargardt病中的致病性。方法先证者因视力下降于2021年5月就诊于济南市第二人民医院,对先证者及家系成员进行详细的眼科检查,根据Stargardt病临床诊断标准评估确诊先证者为Stargardt病。提取先证者及家系其他成员基因组DNA,对先证者DNA进行全外显子测序寻找致病的突变基因。通过PolyPhen-2、SIFT、MutationTaster等网站分析突变位点危害性,利用Sanger测序验证并确认致病突变,进行同源物种保守性分析及三维蛋白结构功能预测以分析其致病性。结果眼科检查表明先证者双眼视力下降、黄斑萎缩并伴有“牛眼征”,其他家系成员均正常。通过先证者的全外显子测序分析和家系成员及正常对照的Sanger测序验证,ABCA4基因的复合杂合突变(c.215G>A和c.6563T>C)与该家系患者表型共分离。SIFT、PolyPhen-2和MutationTaster网站预测这2个突变均有害,保守性分析和三维蛋白结构模型预测表明该突变导致蛋白的结构发生改变,影响蛋白质功能。结论ABCA4基因的复合杂合突变(c.215G>A和c.6563T>C)为该Stargardt病家系携带的致病突变。

Reduction of the radar cross-section(RCS) is the key to stealth technology. To improve the RCS reduction effect of the designed checkerboard metasurface and overcome the limitation of thinlayer plasma in RCS reduction technology, a double-layer-plasma-based metasurface—composed of a checkerboard metasurface, a double-layer plasma and an air gap between them—was investigated. Based on the principle of backscattering cancellation, we designed a checkerboard metasurface composed of different artificial magnetic conductor units;the checkerboard metasurface can reflect vertically incident electromagnetic(EM) waves in four different inclined directions to reduce the RCS. Full-wave simulations confirm that the doublelayer-plasma-based metasurface can improve the RCS reduction effect of the metasurface and the plasma. This is because in a band lower than the working band of the metasurface, the RCS reduction effect is mainly improved by the plasma layer. In the working band of the metasurface,impedance mismatching between the air gap and first plasma layer and between first and second plasma layers cause the scattered waves to become more dispersed, so the propagation path of the EM waves in the plasma becomes longer, increasing the absorption of the EM waves by the plasma. Thus, the RCS reduction effect is enhanced. The double-layer-plasma-based metasurface can be insensitive to the polarization of the incoming EM waves, and can also maintain a satisfactory RCS reduction band when the incident waves are oblique.

某型飞机横向操纵时副翼和扰流板会耦合偏转实现飞机的滚转运动,现有气动力参数辨识方法无法单独识别出副翼和扰流板的操纵导数。基于飞机控制律设计逻辑和滚转运动时结构的受载特点,提出一种基于飞行实测载荷的副翼操纵导数识别方法。通过实测机动飞行中机翼各测载剖面的结构载荷并结合剖面外结构质量与加速度分布,提出了机动飞行中机翼气动载荷的测量方法;结合飞机横向操纵瞬时滚转力矩平衡方程和操纵面偏转逻辑,推导了滚转改出时副翼操纵导数识别过程,提出了基于机翼实测载荷的副翼操纵导数识别方法;基于前述方法开展了副翼操纵导数识别,并分析了不同马赫数对副翼操纵导数的影响。研究表明,基于飞行实测载荷能够进行副翼操纵导数识别,而且同一马赫数下副翼操纵导数识别结果集中度较好,随着马赫数的增加,副翼操纵效能将会降低。飞行载荷实测结果可用于气动参数辨识工作中。

Objective: Triple-negative breast cancer(TNBC) is a heterogeneous and aggressive cancer. Although our previous study classified primary TNBC into four subtypes, comprehensive longitudinal investigations are ***: We assembled a large-scale, real-world cohort comprised of 880 TNBC patients [465 early-stage TNBC(e TNBC) and 415 metastatic TNBC(m TNBC) patients] who were treated at Fudan University Shanghai Cancer Center. The longitudinal dynamics of TNBC subtypes during disease progression were elucidated in this patient cohort. Comprehensive analysis was performed to compare primary and metastatic lesions within specific TNBC ***: The recurrence and metastasis rates within 3 years after initial diagnosis in the e TNBC cohort were 10.1%(47/465). The median overall survival(OS) in the m TNBC cohort was 27.2 months [95% confidence interval(CI), 24.4±30.2 months], which indicated a poor prognosis. The prognostic significance of the original molecular subtypes in both e TNBC and m TNBC patients was confirmed. Consistent molecular subtypes were maintained in 77.5% of the patients throughout disease progression with the mesenchymal-like(MES) subtype demonstrating a tendency for subtype transition and brain metastasis. Additionally, a precision treatment strategy based on the metastatic MES subtype of target lesions resulted in improved progression-free survival in the FUTURE ***: Our longitudinal study comprehensively revealed the clinical characteristics and survival of patients with the original TNBC subtypes and validated the consistency of most molecular subtypes throughout disease progression. However, we emphasize the major importance of repeat pathologic confirmation of the MES subtype.

Effects of Different Fertilization Treatments on Soil, Leaf Nutrient and Fruit Quality of Citrus grandis var. longanyou

The five-year-old “Longanyou” trees were used as the experimental material to study the effects of different fertilization treatments. The nutrient contents in soil and leaves, fruit yield and quality were determined, and then the correlations were analyzed. The results showed that: 1) The soil nutrient contents of 0 - 20 cm depth were more than the 20 - 40 cm, and the trends of nutrient contents of the 0 - 20 cm soil layers were as follows: treatment 2 (T2) > treatment 3 (T3) > treatment 4 (T4) > treatment 1(T1) > control (CK). However, the 20 - 40 cm depth had not significant difference between different treatments, but T2, T4 and T3 were higher than T1 and CK. It indicated that the soil effective nutrient content increased in T2 and T3. 2) Compared with the control, the content of K and B elements was improved obviously in leaves with the increase of organic manure application. The contents of P (1.60 g·kg-1), B (26.00 mg·kg-1) and Mg (1.18 g·kg-1) were the highest, and other nutrients contents were also higher, indicating that T2 could effectively improve the leaves’ nutrient contents. 3) The fruit yield per plant was the highest in T2 (95.40 kg plant-1), and the single fruit weight, total sugar, sugar and acid ratio, vitamin C were also the highest, but titratable acid was lower. It indicated that T2 effectively improved fruit yield and quality. 4) There were positive correlations between multiple factors of soil nutrients and the quality index, such as fruit peel thickness, total sugar, solid acid ratio, sugar and acid ratio, Vc content and single yield etc. There was significant correlation between K, B, Zn, Fe contents and fruit yield and quality index, and the contents of B, Zn and Fe in leaves were significantly correlated with soil nutrient, indicating that the contents of K, B, Zn, Fe in soil and leaf were closely related to fruit yield and quality. In sum, the T2 was the best fertilization scheme for orchard management practice of “Longanyou”.
