Relationship between AQP4 expression and structural damage to the blood-brain barrier at early stages of traumatic brain injury in rats

Background Although some studies have reported that aquaporin-4 （AQP4） plays an important role in the brain edema after traumatic brain injury （TBI）, little is known about the AQP4 expression in the early stage of TBI, or about the correlation between the structural damage to the blood-brain barrier （BBB） and angioedema. The aim of this project was to investigate the relationship between AQP4 expression and damage to the BBB at early stages of TBI. Methods One hundred and twenty healthy adult Wistar rats were randomly divided into two groups： sham operation group （SO） and TBI group. The TBI group was divided into five sub-groups according to the different time intervals： 1, 3, 6, 12, and 24 hours. The brains of the animals were taken out at different time points after TBI to measure brain water content. The cerebral edema and BBB changes in structure were examined with an optical microscopy （OM） and transmission electron microscopy （TEM）, and the IgG content and AQP4 protein expression in traumatic brain tissue were determined by means of immunohistochemistry and Western blotting. The data were analyzed with SPSS 13.0 statistical software. Results In the SO group, tissue was negative for IgG, and there were no abnormalities in brain water content or AQP4 expression. In the TBI group, brain water content significantly increased at 6 hours and peaked at 24 hours following injury. IgG expression significantly increased from 1 to 6 hours following injury, and remained at a high level at 24 hours. Pathological observation revealed BBB damage at 1 hour following injury. Angioedema appeared at 1 hour, was gradually aggravated, and became obvious at 6 hours. Intracellular edema occurred at 3 hours, with the presence of large glial cell bodies and mitochondrial swelling. These phenomena were aggravated with time and became obvious at 12 hours. AIM: To investigate the effect of human apolipoprotein B mRNA-editing enzyme catalytic-polypeptide 3G (APOBEC3G) and its N-terminal or C-terminal cytosine deaminase domain-mediated antiviral activity against hepatitis B virus (HBV) in vitro and in vivo. METHODS: The mammalian hepatoma cells hepG2 and huH7 were cotransfected with APOBEC3G and its N-terminal or C-terminal cytosine deaminase domain expression vector and 1.3-fold-overlength HBV DNA as well as the linear monomeric HBV of genotype B and C. For in vivo study, an HBV vector-based mouse model was used in which APOBEC3G and its N-terminal or C-terminal cytosine deaminase domain expression vectors were co-delivered with 1.3-fold-overlength HBV DNA via high-volume tail vein injection. Levels of hepatitis B virus surface antigen (HBsAg) and hepatitis B virus e antigen (HBeAg) in the media of the transfected cells and in the sera of mice were determined by *** expression of hepatitis B virus core antigen (HBcAg) in the transfected cells was determined by Western blot analysis. Core-associated HBV DNA was examined by Southern blot analysis. Levels of HBV DNA in the sera of mice as well as HBV core-associated RNA in the liver of mice were determined by quantitative PCR and quantitative RT-PCR analysis, respectively. RESULTS: human APOBEC3G exerted an anti-HBV activity in a dose-dependent manner in hepG2 cells, and comparable suppressive effects were observed on genotype B and C as that of genotype A. Interestingly, the N-terminal or C-terminal cytosine deaminase domain alone could also inhibit HBV replication in hepG2 cells as well as huh7 cells. Consistent with in vitro results, the levels of HBsAg in the sera of mice were dramatically decreased, with more than 50 times decrease in the levels of serum HBV DNA and core-associated RNA in the liver of mice treated with APOBEC3G and its N-terminal or C-terminal cytosine deaminase domain as compared to the controls. 目的探讨水通道蛋白4（aquaporins4，AQP4）在早期创伤性脑水肿组织中的表达与组织病理改变，以及与血脑屏障（blood—brain barrier，BBB）结构破坏的相关性。方法健康成年Wistar大鼠120只，制作脑创伤模型，按随机数字表法分为假手术组和创伤组，创伤组又分为创伤1h组、3h组、6h组、12h组、24h组5个亚组，每组20只。分别测定脑含水量，病理观察脑水肿情况及BBB结构变化，免疫组织化学及免疫印迹实验检测脑组织IgG和AQP4表达。结果假手术组脑组织结构正常，IgG染色阴性，脑含水量及AQP4表达未见异常。创伤组的脑含水量从创伤6h后明显增加，至24h最高；IgG从创伤1h后即明显增加至6h，之后维持高水平至24h。病理观察显示，脑创伤后1h即出现BBB破坏，红细胞从血管内漏出，组织间隙增宽，呈现血管源性水肿，并随时间延长而逐渐加重；创伤后6h较明显。创伤后3h开始出现细胞内水肿，胶质细胞胞体增大，胞浆淡染或空泡样变，线粒体肿胀，亦随时间延长而逐渐加重，在6h较为明显。创伤后12h除了上述两种水肿并存之外，开始出现组织坏死、炎性细胞浸润及小胶质细胞增生，24h较明显。AQP4蛋白在1h开始降低，6h降至最低点，12h开始回升，呈“V”形变化。结论以BBB破坏为特征的血管源性水肿是早期脑创伤的主要病理改变，继而出现细胞内水肿并存的混合性水肿，随时间延长两种水肿逐渐加重。早期脑创伤时AQP4表达下调，而当细胞内水肿参与时AQP4表达上调。

The second(O2)observational campaign of gravitational waves(GWs)organized by the LIGO/Virgo Collaborations has led to several breakthroughs such as the detection of GW signals from merger systems involving black holes or neutrons *** O2,14 GW alerts were sent to the astronomical community with sky regions mostly covering over hundreds of square *** them,six were finally confirmed as real astrophysical *** 2013,a new set of ground-based robotic telescopes called Ground-based Wide Angle Camera system(GWAC)project and its pathfinder mini-GWAC has been developed to contribute to the various challenges of multi-messenger and time domain *** GWAC system is built up in the framework of the ground-segment system of the SVOM mission that will be devoted to the study of the multi-wavelength transient sky in the next *** O2,only the mini-GWAC telescope network was fully *** to the wide field of view and fast automatic follow-up capabilities of the mini-GWAC telescopes,they were adept to efficiently cover the sky localization areas of GW event *** this paper,we present the mini-GWAC pipeline we have set up to respond to GW alerts and we report our optical follow-up observations of eight GW alerts detected during the O2 *** observations provided the largest coverage of the GW localization areas with a short latency made by any optical *** found tens of optical transient candidates in our images,but none of those could be securely associated with any confirmed black hole-black hole merger *** on this first experience and the near future technical improvements of our network system,we will be more competitive in detecting the optical counterparts from some GW events that will be identified during the upcoming O3 run,especially those emerging from binary neutron star mergers.

This study aimed to assess the efficacy and safety of the traditional Chinese medicine, Niaoduqing particles, for delaying renal dysfunction in patients with stage 3b-4 ***： The present study was a prospective, randomized, double-blind, placebo-controlled, naulticentcr clinical trial. Frorn May 2013 to December 2013,300 CKD patients with an estimated glomerular filtration rate （eGFR） between 20 and 45 ml ＂rain ~＂ 1.73 m 2, aged 18-70 years were recruited from 22 hospitals in 11 Chinese provinces. Patients were randomized in a 1：1 ratio to either a test group, which was administered Niaoduqing particles 5 g thrice daily and 10 g before bedtime for 24 weeks, or a control group, which was administered a placebo using the same methods. The primary endpoints were changes in baseline serum creatinine （Scr） and eGFR after completion of treatment. The primary endpoints were analyzed using Student＇s t-test or Wilcoxon＇s rank-sum test. The present study reported results based on an intention-to-treat （ITT） analysis. Results： A total of 292 participants underwent the ITT analysis. At 24 weeks, the median （interquartile range） change in Scr was 1.1 （-13.0-24.1） and 11.7 （-2.6-42.9） p, mol/L for the test and control groups, respectively （Z = 2.642, P = 0.008）, and the median change in eGFR was -0.2 （-4.3-2.7） and -2.2 （-5.7-0.8） ***-1·1.73 m-2, respectively （Z = -2.408, P = 0.016）. There were no significant differences in adverse events between the groups. Conclusions： Niaoduqing particles safely and effectively delayed CKD progression in patients with stage 3b-4 CKD. This traditional Chinese medicine may be a promising alternative medication for patients with moderate-to-severe renal dysfunction.

Background MicroRNAs （miRNAs） control gene expression by destabilizing target transcripts and inhibiting their translation. Aberrant expression of miRNAs has been described in many human diseases, including schizophrenia. However, the effects on miRNA expression in response to antipsychotic treatment in peripheral circulation have not been thoroughly examined. Methods Using quantitative real-time PCR （qRT-PCR）, We quantified the expression of seven candidate miRNAs in plasma samples of 40 first-episode schizophrenics before and after antipsychotic treatment. The patients were all treated with risperidone and achieved remission in 1 year. Results Compared with the baseline, the expression levels of miR-365 and miR-520c-3p were significantly down- regulated after 1 year of risperidone treatment （P 〈0.001）. There were no significant correlations between the clinical symptoms and the expression levels of these two miRNAs （P 〉0.05）. Conclusions This study analyzed possible circulating miRNAs in response to antipsychotic monotherapy for schizophrenia, the further mechanism need to be confirmed.

Objective: To follow up the participants of the randomized clinical trial "Efficacy and Safety of Niaoduqing Particles(尿毒清颗粒) for Delaying Moderate-to-Severe Renal Dysfunction", and assess the long-term effects of Niaoduqing Particles on delaying the progression of renal dysfunction. Methods: Participants, who had previously been randomly assigned to receive Niaoduqing Particles or placebo for 24 weeks(146 cases in each group), were invited to follow-up and all were administered Niaoduqing Particles 5 g thrice daily and 10 g before bedtime for 24 weeks. The primary endpoints were changes in baseline serum creatinine(Scr) and estimated glomerular filtration rate(e GFR) after completion of the open-label treatment period. Results: After the double-blind period, the median(interquartile range) changes in Scr were 1.1(–13.0–24.1) and 11.7(–2.6–42.9) μmol/L for the Niaoduqing Particle and placebo groups, respectively(P=0.008), and the median changes in e GFRs were –0.2(–4.3–2.7) and –2.21(–5.7–0.8) mL·min^(-1)·1.73 m^(-2), respectively(P=0.016). There were significant differences in the double-blind period changes in renal function between groups. After the open-label period, the median changes in Scr were 9.0(–10.0–41.9) and 17.5(–6.0–50.0) μmol/L for the Niaoduqing Particle and placebo groups according to baseline grouping, respectively(P=0.214), and the median changes in eGFRs were –2.3(–6.4–1.9) and –3.7(–7.5–1.1) mL·min^(-1)·1.73 m^(-2), respectively(P=0.134). There were no statistical differences in the open-label period changes in renal function between groups. The eGFR reduction of participants who accepted Niaoduqing Particle treatment for 48 weeks was projected to 2.5 m L·min^(-1)·1.73 m(-2) per year. Conclusions: Niaoduqing Particles appear to have long-term efficacy for patients with moderate-to-severe renal dysfunction. Although there was no statistical difference, the early use of Niaoduqing Paticles seems to ameliorate the worsening of renal function.
