Effect of fusion protein TAT and heme oxygenase-1 on liver sinusoidal endothelial cells apoptosis during preservation injury

Background Proteins or peptides can be directly transferred into cells when covalently linked to protein transduction domains （PTDs）. TAT is one of the most widely studied PTDs. The effect of fusion protein TAT and heme oxygenase-1 （HO-1） on liver sinusoidal endothelial cells （SECs） apoptosis during cold storage is unknown. The present study aimed to determine whether fusion protein TAT-HO-1 would transduce efficiently into liver during cold storage, and, if so, to determine whether TAT-HO-1 would attenuate SECs apoptosis during preservation injury in rat. Methods Livers of Sprague-dawley rats were harvested and randomly assigned to group 1 （HTK solution） and group 2 （HTK solution containing TAT-HO-1 fusion protein） according to the type of the preservation solution. The transduction efficiency of TAT-HO-1 was examined and the impairment of SECs was assessed during the period of cold storage followed by 1 hour of reperfusion. Results TAT-HO-1 can transduce efficiently into liver during cold storage. A significantly lower apoptotic index of SECs was observed in group 2, at 6, 12 and 18 hours of cold storage after 1 hour reperfusion, when compared with group 1. TAT-HO-1 reduced HA and ET levels in liver at each time point. Both Bcl-2 and Bax protein were expressed in hepatocytes and SECs at the periphery of the sinusoidal space. Moreover, higher Bcl-2 expression and lower Bax expression were observed in group 2. Conclusions TAT-HO-1 can transduce efficiently into rat livers and shows a protective effect on SECs by attenuating apoptosis during cold ischemia/reperfusion injury. Protein transduction will be a novel therapeutic strategy to reduce the risk of preservation injury in liver transplantation.

Protection of human islets from induction of apoptosis and improved islet function with HO-1 gene transduction

Background Islet transplantation represents an ideal therapeutic approach for treatment of type 1 diabetes but islet function and regeneration may be influenced by necrosis or apoptosis induced by oxidative stress and other insults. Heme oxygenase-1 （HO-1） is the rate-limiting enzyme in the catabolism of heme into biliverdin, releasing free iron and carbon monoxide. It has also been reported to be an antioxidant enzyme which can improve the function of grafted islets by cytoprotection via free radical scavenging and apoptosis prevention. In the present study, we investigated whether transduction of HO-1 genes into human islets with an adenovirus vector has cytoprotective action on islets cultured in vitro and discuss this method of gene therapy for clinical islet transplantation. Methods Cadaveric pancreatic islets were isolated and purified in vitro. Transduction efficiency of islets was determined by infecting islets with adenovirus vector containing the enhanced green fluorescent protein gene （Ad-EGFP） at multiplicities of infection （MOI） of 2, 5, 10, or 20. Newly isolated islets were divided into three groups： EGFP group, islets transduced with Ad-EGFP using MOI=20; HO-1 group, transduced with adenovirus vectors containing the human HO-1 gene using MOI=20; and control group, mock transduced islets. Insulin release after glucose stimulation of the cell lines was determined by a radioimmunoassay kit and the stimulation index was calculated. Flow cytometry was used to detect apoptotic cells in the HO-1 group and in the control group after induction by recombinant human tumor necrosis factor-α （rTNFα） and cycloheximide （CHX） for 48 hours. Results Adenovirus vectors have a high efficiency of gene transduction into adult islet cells. Transduction of islets with the Ad-EGFP was most successful at MOI 20, at which MOI fluorescence was very intense on day 7 after transduction and EGFP was expressed in cultured islet cells for more than four weeks in vitro. With abundant potassium resources and high capacity,potassium metal batteries(PMBs)present a compelling option for the next generation of energy storage ***,PMBs suffer from an unstable anode interface caused by uncontrolled dendrite growth,which results in unsatisfactory cyclability and safety *** investigations suggest that significant progress has been made in enhancing the interfacial stability of *** various effective strategies for stabilizing interfaces can ultimately be attributed to the regulation of the sluggish ion transfer kinetics and irregular deposition,i.e.,the arrangement of ion transport behaviors at the *** modulation of ions transport rate and ions deposition directions makes it possible to obtain a dendrite-free and smooth deposition ***,the influencing factors and action mechanism of K^(+)interface transport behaviors are discussed to understand the nature of material design for constructing stable anode interfaces,including regulating the solvation and desolvation structures,accelerating K^(+)transport kinetics and controlling K^(+)deposition *** addition,the deficiencies and prospects of the research on electrolyte,separators and designed electrode involved in the manufacturing and testing and ion transport process of PMBs are *** review is expected to provide some possible directions for constructing dendrite-free interfaces in advanced PMBs-related research and offer significant insights for prospective experimental research and commercial applications.

Mechanical behaviors of mylonitic granite and granitic protomylonite in a deep ductile shear zone

For projects near the tectonic belt,mylonite of varying metamorphic degrees may be *** matrix proportion of rock reflects its internal microscopic characteristics,thus it is beneficial for engineering geology to study the effect of the matrix proportion on the mechanical properties and rupture behaviors of *** of mylonitic granite and granitic protomylonite with varying matrix proportions were obtained from a ductile shear zone for a series of uniaxial compression and acoustic emission(AE)*** results showed that with the increase in matrix proportion,the average strength and elastic modulus of the samples increased,and the rock sample with the largest matrix proportion exhibited the maximum peak stress of 244.42 MPa,which was 45.86%greater than the average peak stress of the rock samples with the smallest matrix *** the rock samples with larger matrix proportion,their mechanical parameters exhibited greater dispersion and the large-scale appearance of AE events occurred earlier,showing a relatively gradual failure *** samples had larger accumulated AE parameter values and greater degree of *** contrast,for samples with smaller matrix proportions,the large-scale appearance of AE events occurred close to the peak stress,indicating that the occurrence of damage and fractures was centralized and *** samples had lower accumulated AE parameter values and fewer cracks after ***,for the rock samples with more matrix proportion,the average variance of the b-value was 1.1,which was lower than that of rock samples with the smallest matrix proportion(the average variance of the b-value was 3.7).Furthermore,it can be predicted that under certain stress,the failure depth around a tunnel is generally smaller when the strength of rock samples with larger matrix proportion is greater.

Elemental doping is an effective strategy to enhance photocatalytic activity and extend the light absorption range of single-component *** this work,a series of La-doped CeO2nanorods（La-CeO2-x） with La content of 1 wt%-15 wt% are synthesized by a simple hydrothermal method and further used as photocatalyst for sulfamerazine（SMR） *** prepared La-CeO2-x nanorods exhibit a great improvement in electron-hole pair migration and visible-light response due to the synergistic effect of abundant oxygen vacancies and heterogeneous elements（La）.Consequently,La-CeO2-x exhibited excellent visible-light photocatalytic performances and chemical stability for SMR degradation,the La-CeO2-5 sample achieved the highest SMR degradation rate of 81%,which was 3.4 times higher than that of the original ***,three possible degradation pathways of SMR in La-CeO2photocatalytic reactions were proposed by liquid chromatography-mass spectrometry ***,density functional theory calculations were carried out to provide an in-depth understanding of the structure-performance *** its excellent properties and better photocatalytic performance,this study demonstrates that La doping in CeO2is an effective way to increase oxygen vacancy and improve the photochemical properties of photocatalysts.

Ethanol extract of Abelmoschus manihot suppresses endoplasmic reticulum stress in contrast-induced nephropathy

Modern conflicts demand substantial physical and psychological exertion,often resulting in fatigue and diminished combat or operational *** exoskeletons have been developed recently to address these challenges,presenting various limitations that affect their operational or everyday *** article evaluates the performance of a dual-purpose passive ankle exoskeleton developed for the reduction of metabolic costs during walking,seeking to identify a force element that could be applied to the target *** on the 6-min walk test,twenty-nine subjects participated in the study using three different force *** results indicate that it is possible to reduce metabolic expenditure while using the developed ***,the comfort and range of motion results verify the exoskeleton's suitability for use in uneven terrain and during extended ***,the choice of the force element should be tailored to each user,and the control system should be adjustable to optimise the exoskeleton's performance.
