Relationship Between Leaf C/N Ratio and Insecticidal Protein Expression in Bt Cotton as Affected by High Temperature and N Rate

Expression of insecticidal protein for transgenic Bacillus thuringiensis （Bt） cotton is unstable and related to nitrogen metabolism. The objective of this study was to investigate the relationship between leaf carbon nitrogen ratio （C/N） and insecticidal efficacy of two Bt cotton cultivars. C/N ratio and Bt protein content were both measured at peak square period and peak boll period respectively under 5-7 d high temperature and different nitrogen fertilizer rates on the Yangzhou University Farm and the ludong Cotton Farm, China. All plants were grown in field. The results showed that the C/N ratio enhanced slightly and the Bt protein content remained stable at peak square period, but significant increases for the C/N ratio and decreases markedly for the leaf Bt protein concentration were detected at the peak boll period. The similar patterns at the two growth periods were found for the leaf C/N ratio and Bt protein content by different N fertilizer treatments. When nitrogen rate was from 0 to 600 kg ha-l, the C/N ratio was reduced by 0.017 and 0.006 for Sikang 1 and Sikang 3 at peak square period, compared to the 1.350 to 1.143 reduction for Sikang 1 and Sikang 3 at peak boll period, respectively. Correspondingly, the leaf Bt protein contents were bolstered by 2.6-11.8 and 26.9-36.9% at the two different growth periods, respectively. The results suggested that enhanced C/N ratio by high temperature and nitrogen application may result in the reduction of inseetiocidal efficacy in Bt cotton, especially in peak boll period.

Two new marasmone derivatives,1α,2β-dihydroxymarasmone（1） and 2β,3α-dihydroxymarasmone（2）,and one new norsesquiterpenoid,14-hydroxyoreadone（3）,were obtained from the fermentation broth of Agrocybe sp.,together with three known sesquiterpenes,agrocybolacton（4）,marasmene（5） and 14-hydroxymarasmene（6）.Their structures were elucidated by spectroscopic analyses,including 1D-and 2D-NMR experiments,high resolution electron spray ionization quadrupole time of flight（HR-ESI-Q-TOF） mass spectrometry,chemical correlation,X-ray single crystal diffraction and also compared with reported data.

2023年6月26日,国际可持续准则理事会正式发布《国际财务报告可持续披露准则第2号——气候相关披露》最终稿,该准则未来将可能成为全球气候相关披露的基准规范,相关披露要求也极可能在我国落地执行。对标我国目前气候风险管理和绿色金融业务发展现状,商业银行若执行该准则将会在数据采集、碳核算与统计、情景分析、气候目标等方面面临一定的挑战。因此,建议商业银行主动识别自身差距、提前部署准备工作、摸清碳数据“家底”、聘请外部机构协助开展情景分析、提升人才队伍专业能力并启动编制气候专项报告的先行先试工作。

The layer Hall effect describes electrons spontaneously deflected to opposite sides at different layers, which has been experimentally reported in the MnBi2Te4thin films under perpendicular electric fields. Here, we reveal a universal origin of the layer Hall effect in terms of the so-called hidden Berry curvature, as well as material design principles. Hence, it gives rise to zero Berry curvature in momentum space but non-zero layer-locked hidden Berry curvature in real space. We show that, compared to that of a trivial insulator, the layer Hall effect is significantly enhanced in antiferromagnetic topological insulators. Our universal picture provides a paradigm for revealing the hidden physics as a result of the interplay between the global and local symmetries, and can be generalized in various scenarios.

钩吻总碱对肺腺癌细胞增殖和凋亡作用的研究

目的以人肺腺癌细胞(A549、SPCA1)为研究对象,通过在培养液中加入不同浓度的钩吻总碱(TAG)研究其抗肿瘤作用。方法采用加热回流、萃取等方法提取TAG,利用薄层色谱法鉴定TAG的提取,通过Incucyte S3活细胞动态分析系统拟合不同浓度的TAG作用A549、SPCA1细胞的细胞融合度并观察A549、SPCA1细胞的形态,采用CCK-8法、集落形成实验检测细胞增殖,Hoechst33258染色、Rhodamine123染色检测细胞凋亡,碘化丙啶单染法检测细胞周期,Western blotting检测凋亡指标蛋白Bax、Bcl-2、Caspase-3蛋白表达。结果经薄层色谱法鉴定成功提取TAG。Incucyte S3活细胞动态分析系统拟合出TAG作用A549、SPCA1细胞的EC50值分别是99.2、82.0μg/mL,由此确定TAG作用A549、SPCA1细胞的低、中、高浓度依次为50、100、150μg/mL,且细胞拍照观察到随着药物浓度增加细胞融合度下降、细胞数目减少。CCK-8法和集落形成实验结果表明TAG抑制A549、SPCA1细胞增殖(P <0.05)。Hoechst 33258细胞核染色实验发现给药组细胞出现核碎裂;流式细胞术检测Rhodamine123探针发现给药组出现荧光信号强度增加,检测细胞周期发现TAG使A549、SPCA1细胞周期阻滞在G_2/M期;Western blot结果显示TAG诱导Bcl-2蛋白表达下调,Bax表达上调、Caspase-3蛋白切割活化增加,通过细胞染色实验、流式细胞术及Western blotting表明TAG促进A549、SPCA1细胞凋亡(P <0.05)。结论 TAG抑制肺腺癌细胞增殖,诱导细胞凋亡。

Geochemical compositions can be used to determine the tectonic setting of sedimentary basins,while where the link of source to sink is no longer preserved,detrital zircon age patterns can aid in resolving the original basin setting.The metasedimentary Legoupil Formation,located at Cape Legoupil and the Schmidt Peninsula,could give a hint for the tectonic evolution of Antarctic Peninsula.In this contribution,we constrain the sedimentary provenance of the Legoupil Formation through geochemistry and detrital zircon U-Pb geochronology.The petrography and geochemical features indicate that the provenance of the Legoupil Formation could be felsic rocks.Detrital zircon grains record a steady supply of Permian and Ordovician material into the Legoupil Formation.The youngest concordant zircon ages of 262 Ma suggest that the depositional time of Legoupil Formation is no older than Late Permian.The detrital zircon age spectrum of Legoupil Formation suggests that the Legoupil Formation sediments should be derived from regional sources endemic to western Gondwana prior to its breakup.Together with the previous studies,geochemistry and detrital zircons reflect an active continental margin tectonic setting and the detrital zircon spectra of Legoupil Formation are similar to the ones deposited in forearc tectonic setting.

Objective:To explore the mechanism by which icariin alleviates viral myocarditis.Methods:CVB3-induced cardiomyocytes were used as an in vitro model of viral myocarditis to assess the effects of icariin treatment on cell viability,inflammation,and apoptosis.Moreover,the effects of icariin on ferroptosis and TLR4 signaling were assessed.After AC16 cells were transfected with TLR4 overexpression plasmids,the role of TLR4 in mediating the regulatory effect of icariin in viral myocarditis was investigated.Results:Icariin significantly elevated cell viability and reduced inflammatory factors TNF-α,IL-1β,IL-6,and IL-18.Flow cytometry revealed that icariin decreased apoptosis rate,and the protein expression of Bax and cleaved caspase 3 and 9 in CVB3-induced cardiomyocytes.Additionally,it suppressed ferroptosis including lipid peroxidation and ferrous ion,as well as the TLR4 signaling.However,TLR4 overexpression abrogated the modulatory effects of icariin.Conclusions:Icariin mitigates CVB3-induced myocardial injury by inhibiting TLR4-mediated ferroptosis.Further animal study is needed to verify its efficacy.

This paper studies the optimal portfolio allocation of a fund manager when he bases decisions on both the absolute level of terminal relative performance and the change value of terminal relative performance comparison to a predefined reference point. We find the optimal investment strategy by maximizing a weighted average utility of a concave utility and an Sshaped utility via a concavification technique and the martingale method. Numerical results are carried out to show the impact of the extent to which the manager pays attention to the change of relative performance related to the reference point on the optimal terminal relative performance.
