Verteporfin fluorescence in antineoplastic-treated pancreatic cancer cells found concentrated in mitochondria

BACKGROUND Traditional treatments for pancreatic cancer(PC)are inadequate.Photodynamic therapy(PDT)is non-invasive,and proven safe to kill cancer cells,including PC.However,the mitochondrial concentration of the photosensitizer,such as verteporfin,is key.AIM To investigate the distribution of fluorescence of verteporfin in PC cells treated with antitumor drugs,post-PDT.METHODS Workable survival rates of PC cells(AsPC-1,BxPC-3)were determined with chemotherapy[doxorubicin(DOX)and gemcitabine(GEM)]and non-chemotherapy[sirolimus(SRL)and cetuximab(CTX)]drugs in vitro,with or without verteporfin,as measured via MTT,flow cytometry,and laser confocal microscopy.Reduced cell proliferation was associated with GEM that was more enduring compared with DOX.Confocal laser microscopy allowed observation of GEM-and verteporfin-treated PC cells co-stained with 4’,6-diamidino-2-phenylindole and MitoTracker Green to differentiate living and dead cells and subcellular localization of verteporfin,respectively.RESULTS Cell survival significantly dropped upon exposure to either chemotherapy drug,but not to SRL or CTX.Both cell lines responded similarly to GEM.The intensity of fluorescence was associated with the concentration of verteporfin.Additional experiments using GEM showed that survival rates of the PC cells treated with 10μmol/L verteporfin(but not less)were significantly lower relative to nil verte-porfin.Living and dead stained cells treated with GEM were distinguishable.After GEM treatment,verteporfin was observed primarily in the mitochondria.CONCLUSION Verteporfin was observed in living cells.In GEM-treated human PC cells,verteporfin was particularly prevalent in the mitochondria.This study supports further study of PDT for the treatment of PC after neoadjuvant chemotherapy.

Heterochronous multiple primary prostate cancer and lymphoma:A case report

BACKGROUND Multiple primary malignant tumors(MPMTs)are rare type of cancer,especially when solid tumors are the first and lymphoma is the second primary malignancy.We report a patient with heterochronous MPMTs consisting of prostate cancer and rectal diffuse large B-cell lymphoma(DLBCL).CASE SUMMARY We report a 77-year-old male patient diagnosed with prostate cancer who was treated with radiation therapy and one year of endocrine therapy with bicalutamide(50 mg per day)and an extended-release implant of goserelin(1/28 d).Seven years later,rectal DLBCL with lung metastases was found.CONCLUSION Although rare,the possibility of prostate cancer combined with a double primary cancer of DLBCL can provide a deeper understanding.

Late Cenozoic uplift of the Liupan Mountains:Evidence from the Neogene loess deposits

The Liupan Mountains,one of the important mountain ranges in western China,are located on the boundary between the northeastern Tibetan Plateau and the Ordos Block.The uplift history of the Liupan Mountains remains controversial.Loess deposits are good tracers of regional tectonic and geomorphic changes,because loess is sensitive to erosion and the formation and preservation of loess requires relatively flat highlands and relatively stable tectonic environments.We investigated the distribution of Neogene loess deposits on the western piedmont of the Liupan Mountains and examined a near-continuous loess section(Nanping section)on the piedmont alluvial highlands.Correlation of magnetic susceptibility stratigraphy with the QA-ⅠMiocene loess sequence dates this 56-m section covering the interval from~8.1 to 6.2 Ma.The lower boundary age of this section,together with previously reported Zhuanglang red clay(sand-gravel layers with intercalated loess during~9–8 Ma and near-continuous loess during~8–4.8 Ma)and Chaona red clay(~8.1–2.58 Ma),indicates that the Liupan Mountains were uplifted in the late Miocene(~9–8 Ma)and basically formed by~8 Ma,attesting to no intense mountain building since that time.In addition,based on the information from the Zhuanglang core and the QA-Ⅰsection,we infer that sizable parts of the Liupan Mountains were uplifted during the late Oligocene–early Miocene and did not experience intense uplift during~22–9 Ma.

GPa-level pressure-induced enhanced corrosion resistance in TiZrTaNbSn biomedical high-entropy alloy

TiZrTaNb-based high-entropy alloys(HEAs)are research frontier of biomedical materials due to their high hardness,good yield strength,excellent wear resistance and corrosion resistance.Sn,as an essential trace element in the human body that plays a significant role in physiological process.It has stable chemical properties and a low elastic modulus.In this study,a new material,TiZrTaNbSn HEAs,was proposed as a potential biomedical alloy.The Ti_(35)Zr_(25)Ta_(15)Nb_(15)Sn_(10)biomedical high-entropy alloys(BHEAs)were successfully prepared through an arc melting furnace and then remelted using a German high-temperature and high-pressure apparatus under GPa-level(4 GPa and 7 GPa).The precipitation behavior of the needle-like HCP-Zr_(5)Sn_(3)phase that precipitates discontinuously at the grain boundary was successfully controlled.The phase constitution,microstructure,and corrosion resistance of the alloy were studied.The results show that the needle-like HCP-Zr_(5)Sn_(3)phase is eliminated and the(Zr,Sn)-rich nano-precipitated phase is precipitated in the microstructure under high pressure,which leads to the narrowing of grain boundaries and consequently improves the corrosion resistance of the alloy.In addition,the formation mechanisms of(Zr,Sn)-rich nanoprecipitates in BHEAs were discussed.More Zr and Sn dissolve in the matrix due to the effect of high pressure,during the cooling process,they precipitate to form a(Zr,Sn)-rich nano-precipitated phase.

Poly(3-hexylthio) thiophene Field-effect Transistor Device Performance: Impact of the Content of Hexylthio Side Chain on Backbone

Regioregular poly(3-hexylthio)thiopene(P3HTT) has emerged tremendous potential in organic electronic applications due to the strong noncovalent interactions from the sulfur atom linked to thiophene. However, P3HTT generally exhibits low charge mobility mostly due to poor solution processability attributed to dense arrangement of hexylthio side chain in polymer, which led to strong noncovalent interactions among sulfur atoms. To balance the nonvalent interaction and aggregation for P3HTT, herein, we systematically study the effect of hexylthio side chain content in polymer backbone on the structure and properties. A series of regioregular P3HTT-based homopolymers(P3HTT, P3HTT-50,P3HTT-33 and P3HTT-25) were prepared via Kumada catalyst transfer polycondensation method from a set of mono-, bi-, ter-and quarterthiophenes containing different contents of hexylthio side chain. The DFT calculation shows the planarity of polymers backbone could be improved through reducing the density of hexylthio side chain in polymer mainchain. And significant changes in their crystallinity, aggregation and optical properties were observed with the content of hexylthio side chain reducing. The P3HTT-33 displayed the highest field-effect transistor hole mobility of 2.83×10^(-2) cm^(2)·V^(-1)·s^(-1) resulting from a balance between the crystallinity and planarity. This study demonstrates modulating the content of hexylthio side chain in P3HTT is an effective strategy to optimize the opto-electronic properties of polymer obtaining excellent semiconductor device performance.

High energy density in ultra-thick and flexible electrodes enabled by designed conductive agent/binder composite

Thick electrodes can increase incorporation of active electrode materials by diminishing the proportion of inactive constituents,improving the overall energy density of batteries.However,thick electrodes fabricated using the conventional slurry casting approach frequently exhibit an exacerbated accumulation of carbon additives and binders on their surfaces,invariably leading to compromised electrochemical properties.In this study,we introduce a designed conductive agent/binder composite synthesized from carbon nanotube and polytetrafluoroethylene.This agent/binder composite facilitates production of dry-process-prepared ultra-thick electrodes endowed with a three-dimensional and uniformly distributed percolative architecture,ensuring superior electronic conductivity and remarkable mechanical resilience.Using this approach,ultra-thick LiCoO_(2)(LCO) electrodes demonstrated superior cycling performance and rate capabilities,registering an impressive loading capacity of up to 101.4 mg/cm^(2),signifying a 242% increase in battery energy density.In another analytical endeavor,time-of-flight secondary ion mass spectroscopy was used to clarify the distribution of cathode electrolyte interphase(CEI) in cycled LCO electrodes.The results provide unprecedented evidence explaining the intricate correlation between CEI generation and carbon distribution,highlighting the intrinsic advantages of the proposed dry-process approach in fine-tu ning the CEI,with excellent cycling performance in batteries equipped with ultra-thick electrodes.

Boosted Electrocatalytic Glucose Oxidation Reaction on Noble-Metal-Free MoO_(3)-Decorated Carbon Nanotubes

Electrocatalytic glucose oxidation reaction(GOR)has attracted much attention owing to its crucial role in biofuel cell fabrication.Herein,we load MoO_(3)nanoparticles on carbon nanotubes(CNTs)and use a discharge process to prepare a noblemetal-free MC-60 catalyst containing MoO_(3),Mo_(2)C,and a Mo_(2)C–MoO_(3)interface.In the GOR,MC-60 shows activity as high as 745μA/(mmol/L cm^(2)),considerably higher than those of the Pt/CNT(270μA/(mmol/L cm^(2)))and Au/CNT catalysts(110μA/(mmol/L cm^(2))).In the GOR,the response minimum on MC-60 is as low as 8μmol/L,with a steady-state response time of only 3 s.Moreover,MC-60 has superior stability and anti-interference ability to impurities in the GOR.The better performance of MC-60 in the GOR is attributed to the abundant Mo sites bonding to C and O atoms at the MoO_(3)–Mo_(2)C interface.These Mo sites create active sites for promoting glucose adsorption and oxidation,enhancing MC-60 performance in the GOR.Thus,these results help to fabricate more effi cient noble-metal-free catalysts for the fabrication of glucose-based biofuel cells.

RNA-binding proteins(RBPs)-RNA networks have contributed to cancer development.Circular RNAs(circRNAs)are considered as protein recruiters;nevertheless,the patterns of circRNA-protein interactions in colorectal cancer(CRC)are still lacking.Processing bodies(PBs)formed through liquid-liquid phase separation(LLPS)are membrane-less organelles(MLOs)consisting of RBPs and RNA.Previous evidence suggests a connection between PBs dynamics and cancer progression.Despite the increasingly acknowledged crucial role of RBPs and RNA in the accumulation and maintenance of MLOs,there remains a lack of specific research on the interactions between PBs-related RBPs and circRNAs in CRC.Herein,we identify that MEX-3 RNA binding family member A(MEX3A),frequently upregulated in CRC tissues,predicts poorer patient survival.Elevated MEX3A accelerates malignance and inhibits autophagy of CRC cells.Importantly,MEX3A undergoes intrinsically disordered regions(IDRs)-dependent LLPS in the cytoplasm.Specifically,circMPP6 acts as a scaffold to facilitate the interaction between MEX3A and PBs proteins.The MEX3A/circMPP6 complex modulates PBs dynamic and promotes UPF-mediated phosphodiesterase 5A(PDE5A)mRNA degradation,consequently leading to the aggressive properties of CRC cells.Clinically,CRC patients exhibiting high MEX3A expression and low PDE5A expression have the poorest overall survival.Our findings reveal a collaboration between MEX3A and circMPP6 in the regulation of mRNA decay through triggering the PBs aggregation,which provides prognostic markers and/or therapeutic targets for CRC.

Micro defects formation and dynamic response analysis of steel plate of quasi-cracking area subjected to explosive load

As the protective component,steel plate had attracted extensive attention because of frequently threats of explosive loads.In this paper,the evolution of microstructure and the mechanism of damage in the quasi-cracking area of steel plate subjected to explosive load were discussed and the relationships between micro defects and dynamic mechanical response were revealed.After the explosion experiment,five observation points were selected equidistant from the quasi-cracking area of the section of the steel plate along the thickness direction,and the characteristics of micro defects at the observation points were analyzed by optical microscope(OM),scanning electron microscope(SEM) and electron backscattered diffraction(EBSD).The observation result shows that many slip bands(SBs) appeared,and the grain orientation changed obviously in the steel plate,the two were the main damage types of micro defects.In addition,cracks,peeling pits,grooves and other lager micro defects were appeared in the lower area of the plate.The stress parameters of the observation points were obtained through an effective numerical model.The mechanism of damage generation and crack propagation in the quasicracking area were clarified by comparing the specific impulse of each observation point with the corresponding micro defects.The result shows that the generation and expansion of micro defects are related to the stress area(i.***.the upper compression area,the neutral plane area,and the lower tension area).The micro defects gather and expand at the grain boundary,and will become macroscopic damage under the continuous action of tensile stress.Besides,the micro defects at the midpoint of the section of the steel plate in the direction away from the explosion center(i.***.the horizontal direction) were also studied.It was found that the specific impulse at these positions were much smaller than that in the thickness direction,the micro defects were only SBs and a few micro cracks,and the those decreased with the increase of the distance from the e

Solar-driven CO2conversion into hydrocarbon fuels is a sustainable approach to synchronously alleviating the energy crisis and achieving net CO2emissions.However,the dependence of the conversion process on solar illumination hinders its practical application due to the intermittent availability of sunlight at night and on cloudy or rainy days.Here,we report a model material of Pt-loaded hexagonal tungsten trioxide（Pt/h-WO3） for decoupling light and dark reaction processes,demonstrating the sustainable CO2conversion under dark conditions for the first time.In such a material system,hydrogen atoms can be produced by photocatalytic water splitting under solar illumination,stored together with electrons in the h-WO3through the transition of W6+to W5+and spontaneously released to trigger catalytic CO2reduction under dark conditions.Furthermore,we demonstrate using natural light that CH4production can persist at night and on rainy days,proving the accomplishment of all-weather CO2conversion via a sustainable way.
