Single-cell mass spectrometry studies of drug metabolism heterogeneity and primary resistance to gefitinib in non-small cell lung cancer cells

Patients with epidermal growth factor receptor(EGFR)wild-type non-small cell lung cancer(NSCLC)often show primary resistance to gefitinib *** is thus necessary to study the metabolism of gefitinib in NSCLC cells to comprehensively reveal the reasons for the primary resistance of ***,we develop a platform for studying drug metabolism heterogeneity based on single-cell mass spectrometry(sDMH-scMS)by integrating living-cell electrolaunching ionization MS(ILCEI-MS)and high-performance liquid chromatography-MS(HPLC-MS)analysis,and the primary resistance of NSCLC cells to gefitinib was studied using this *** ILCEI-MS analysis showed that approximately 11.9%of NSCLC single cells contained the gefitinib metabolite M11;HPLC-MS detection diluted the intensity of M11 in subpopulations and concealed the heterogeneity of drug metabolism in tumor single *** intensity of gefitinib in EGFR wild-type A549 cells was markedly lower than in mutant PC9 cells,and the intensity of gefitinib metabolites was significantly higher than in PC9 cells,suggesting that the primary resistance of NSCLC cells is related to gefitinib ***,the combination of gefitinib and the drug-metabolizing enzyme inhibitorα-naphthoflavone was shown to overcome the primary resistance of the NSCLC ***,the results of this study are expected to be applicable for clinical drug resistance diagnosis and treatment at the single-cell level.

Nitrate(NO_(3)^(−))electroreduction reaction(NO_(3)^(−)RR)provides an attractive and sustainable route for NO_(3)^(−)pollution mitigation or energy-saved ammonia(NH3)*** this work,high-quality B and Fe co-doped Co_(2) P hollow nanocubes(B/Fe-Co_(2) P HNCs)are successfully synthesized though simultaneous boronation-phosphorization treatment,which reveal outstanding selectivity,activity,stability for the NO_(3)^(−)to NH_(3) conversion in neutral electrolyte because of big surface area,fast mass transport,superhydrophilic surface,and optimized electronic structure.B/Fe-Co_(2) P HNCs can achieve the high NH3 yield rate(22.67 mg h^(−1) mg_(cat)^(−1))as well as Faradaic efficiency(97.54%)for NO_(3)^(−)RR,greatly outperforming most of non-precious metal based NO_(3)^(−)RR electrocatalysts.

Synergistic chemotherapy/PTT/oxygen enrichment by multifunctional liposomal polydopamine nanoparticles for rheumatoid arthritis treatment

Amultifunctional liposomal polydopamine nanoparticle(MPM@Lipo)was designed in this study,to combine chemotherapy,photothermal therapy(PTT)and oxygen enrichment to clear hyperproliferating inflammatory cells and improve the hypoxic microenvironment for rheumatoid arthritis(RA)***@Lipo significantly scavenged intracellular reactive oxygen species and relieved joint hypoxia,thus contributing to the repolarization of M1 macrophages into M2 ***,MPM@Lipo could accumulate at inflammatory joints,inhibit the production of inflammatory factors,and protect cartilage in vivo,effectively alleviating RA progression in a rat adjuvant-induced arthritis ***,upon laser irradiation,MPM@Lipo can elevate the temperature to not only significantly obliterate excessively proliferating inflammatory cells but also accelerate the production of methotrexate and oxygen,resulting in excellent RA treatment ***,the use of synergistic chemotherapy/PTT/oxygen enrichment therapy to treat RA is a powerful potential strategy.

The increasing use of renewable energy in the power system results in strong stochastic disturbances and degrades the control performance of the distributed power *** this paper,a novel multi-agent collaborative reinforcement learning algorithm is proposed with automatic optimization,namely,Dyna-DQL,to quickly achieve an optimal coordination solution for the multi-area distributed power *** proposed Dyna framework is combined with double Q-learning to collect and store the environmental *** can iteratively update the agents through buffer replay and real-time *** the environmental data can be fully used to enhance the learning speed of the *** mitigates the negative impact of heavy stochastic disturbances caused by the integration of renewable energy on the control *** are conducted on two different models to validate the effectiveness of the proposed *** results demonstrate that the proposed Dyna-DQL algorithm exhibits superior stability and robustness compared to other reinforcement learning algorithms.

中国式现代化与中华民族现代文明的建设,是同一个历史过程的两个方面。在中国式现代化展开过程中,中华民族现代文明日益生成和发展。中国式现代化既是对具体现代化诸模式的超越,也是对一般现代化的超越,是一种超越现代性的新现代化。中华民族现代文明是中国特色社会主义文明,由中国共产党领导、中国人民创造,以中国特色社会主义制度为内核,奋力实现全体社会成员的美好生活,是“文化”之人的文明,从而成为一种超出其他现代文明的更高现代文明,构成人类文明的一种崭新形态,在内容上展开为相互协调的高度的物质文明、政治文明、精神文明、社会文明和生态文明。相对于中华传统文明,中华民族现代文明要求并表现出更为高度的统一性、和平性尤其是包容性与创新性,在兼收并蓄、综合创造中通达更高发展境界。

马克思生息资本思想需要进一步澄明。在表面上,生息资本仿佛不需要做任何事就能从产业资本处得到利息,但其真正运动是分割产业资本剥削到的剩余价值。无产阶级的剩余劳动构成利息的真正源泉。源于既一致又对立的利益,生息资本同产业资本既合作又斗争,构成工业资本主义体系的重要环节,展现了产业资本主导的权力架构。生息资本在特殊运动过程中呈现出拜物教性、疯狂性,亦蕴含一定的社会性。生息资本突出的拜物教性,由物性与神秘性两大向度结合而成。生息资本运动的外部表象脱离其内在实质,撇开所有中介环节与过程,直接从G到G',从而成为物神。此种物神形态必然使资产阶级及其代言人形成物神观念。物神形态与物神观念相互强化,确立起强烈的拜物教。生息资本拜物教的形成,意味着资本拜物教的极致化和最终完成。它使人错误理解资本的典型形态与资本主义生产的总体结构,掩盖资本主义的剥削,削弱反抗和变革意识。马克思以本质—现象透视法、关系性思维、矛盾分析法与历史性思维,注重穿透表象尤其假象,洞见深层本质,并依据真实本质反思虚假现象,根本性地澄明了生息资本,为在新的时代条件下科学理解进而妥善处理生息资本及其发展形式,提供了有力的思想基础。

本文运用顶空固相微萃取-气相色谱-质谱联用技术(HS-SPME-GC-MS)研究了不同发育阶段沙枣花(花蕾、全开和凋谢)、叶片和幼枝的挥发性成分,并采用主成分分析(PCA)、正交偏最小二乘法判别分析(OPLS-DA)等方法筛选出差异代谢物。结果表明,沙枣挥发油以酯类、烃类、醇类和醛类物质为主,主要成分为肉桂酸乙酯、β-罗勒烯和4-己烯-1-醇乙酸酯等。代谢组学分析共筛选出17种差异代谢物。其中沙枣花蕾中的特征成分为α-橙花叔醇、反式-2-己烯醇、十一醛和β-罗勒烯;沙枣花全开时的特征成分为肉桂酸乙酯和安息香酸乙酯;凋谢后沙枣花的特征成分为癸醛、庚醛、植酮和2-甲基丁酸乙酯;沙枣叶片的特征成分为4-己烯-1-醇乙酸酯和叶醇而沙枣幼枝的特征成分为壬醛、顺式-3-己烯基异戊酸酯、苯乙醇和3-己烯-1-醇。沙枣花、叶片和幼枝挥发性成分的代谢组学分析,可为沙枣精油的综合开发利用提供一定理论依据。

GaN-based metal-insulator-semiconductor high-electronmobility-transistors (MISHEMTs) have many excellent performances compared with the Si and GaA s counterparts,and are prime candidates for applications in communication base stations, radars, and satellites. Most of the current GaNRF transistors for the below Ka-band applications adopt a conventional AlGaN/GaN heterostructure with a typical Al-composition from 0.25 to 0.3 for the barrier layer. For higher frequency applications, the thin barrier AlN/GaN heterojunction or InA lN/GaN heterojunction is preferred [1].
