Recent landscape and trends for industry-sponsored pediatric clinical trials in China from 2013 to 2022

Importance: Pediatric medication is a challenging issue globally. Promoting trials of medications for children and implementing measures to encourage innovation for addressing unmet medical and health needs are important.Objective: To explore the recent landscape of pediatric clinical trials of new investigational drugs conducted by pharmaceutical enterprises in China from 2013 to 2022 to provide insight into pediatric drug development in the pharmaceutical industry and regulatory policy formulation.Methods: We performed a cross-sectional observational investigation of pediatric clinical trials registered from January 1, 2013, to December 31, 2022, on the Registration and Information Disclosure Platform for Drug Clinical Trials, the official registration platform established in 2013 for trials of new investigational drugs initiated by biopharmaceutical enterprises. Trials that included pediatric participants (under 18 years old) were retrieved, and their relevant characteristics were extracted and analyzed.Results: In total, 895 pediatric clinical trials were collected, accounting for 5.1% of the total registered clinical trials initiated prior to January 1, 2023. The overall average annual growth rate for the number of pediatric clinical trials was 12% (P < 0.001). Phase III trials accounted for the highest proportion (49.1%, 439). Of the 895 trials included, 736 (82.2%) were domestic trials, and 159 (17.8%) were international multicenter trials. In terms of tested drugs, investigations of biological products accounted for the largest proportion of trials (67.4%, 603). Among pediatric clinical trials, studies of vaccines accounted for the largest proportion of trials (41.0%, 367), followed by trials for rare diseases (17.2%, 154). Furthermore, geographical distribution analysis revealed that the largest and smallest numbers of trials were conducted in North China (35.7%, 320) and Northeast China (0.8%, 7), respectively.Interpretation: The growth trends for industry-sponsored clinical trials involving children il

French architectÉric Le Khanh has been strengthening Sino-French relationship with his works over two decades.IN 2021,archaeologists announced the results of their excavation in Fengtai District in southwest Beijing.They included ruins of the wall of an ancient city known as Zhongdu,the capital of the Jin Dynasty(1115-1234)for six decades before it fell to the Mongols.Subsequently,it was decided that a Jin Zhongdu City Wall Ruins Park would be built on the site,next to Beijing’s new Lize financial district,as part of the capital city’s urban renewal drive.French architect and urban plannerÉric Le Khanh was on the design team.“The protection project is part of a larger urban park and cultural facility that shows how history can be integrated into the central business district in a modern city,”Khanh told China Today.

Real-time Rescue Target Detection Based on UAV Imagery for Flood Emergency Response

Timely acquisition of rescue target information is critical for emergency response after a flood disaster.Unmanned Aerial Vehicles(UAVs)equipped with remote sensing capabilities offer distinct advantages,including high-resolution imagery and exceptional mobility,making them well suited for monitoring flood extent and identifying rescue targets during floods.However,there are some challenges in interpreting rescue information in real time from flood images captured by UAVs,such as the complexity of the scenarios of UAV images,the lack of flood rescue target detection datasets and the limited real-time processing capabilities of the airborne on-board platform.Thus,we propose a real-time rescue target detection method for UAVs that is capable of efficiently delineating flood extent and identifying rescue targets(i.e.,pedestrians and vehicles trapped by floods).The proposed method achieves real-time rescue information extraction for UAV platforms by lightweight processing and fusion of flood extent extraction model and target detection model.The flood inundation range is extracted by the proposed method in real time and detects targets such as people and vehicles to be rescued based on this layer.Our experimental results demonstrate that the Intersection over Union(IoU)for flood water extraction reaches an impressive 80%,and the IoU for real-time flood water extraction stands at a commendable 76.4%.The information on flood stricken targets extracted by this method in real time can be used for flood emergency rescue.

Cambricon-QR:a sparse and bitwise reproducible quantized training accelerator

Quantized training has been proven to be a prominent method to achieve deep neural network training under limited computational resources.It uses low bit-width arithmetics with a proper scaling factor to achieve negligible accuracy loss.Cambricon-Q is the ASIC design proposed to efficiently support quantized training,and achieves significant performance improvement.However,there are still two caveats in the design.First,Cambricon-Q with different hardware specifications may lead to different numerical errors,resulting in non-reproducible behaviors which may become a major concern in critical applications.Second,Cambricon-Q cannot leverage data sparsity,where considerable cycles could still be squeezed out.To address the caveats,the acceleration core of Cambricon-Q is redesigned to support fine-grained irregular data processing.The new design not only enables acceleration on sparse data,but also enables performing local dynamic quantization by contiguous value ranges(which is hardware independent),instead of contiguous addresses(which is dependent on hardware factors).Experimental results show that the accuracy loss of the method still keeps negligible,and the accelerator achieves 1.61×performance improvement over Cambricon-Q,with about 10%energy increase.

As the search for effective treatments for COVID-19 continues,the high mortality rate among critically ill patients in Intensive Care Units(ICU)presents a profound challenge.This study explores the potential benefits of traditional Chinese medicine(TCM)as a supplementary treatment for severe COVID-19.A total of 110 critically ill COVID-19 patients at the Intensive Care Unit(ICU)of Vulcan Hill Hospital between Feb.,2020,and April,2020(Wuhan,China)participated in this observational study.All patients received standard supportive care protocols,with a subset of 81 also receiving TCM as an adjunct treatment.Clinical characteristics during the treatment period and the clinical outcome of each patient were closely monitored and analysed.Our findings indicated that the TCM group exhibited a significantly lower mortality rate compared with the non-TCM group(16 of 81 vs 24 of 29;0.3 vs 2.3 person/month).In the adjusted Cox proportional hazards models,TCM treatment was associated with improved survival odds(P<0.001).Furthermore,the analysis also revealed that TCM treatment could partially mitigate inflammatory responses,as evidenced by the reduced levels of proinflammatory cytokines,and contribute to the recovery of multiple organic functions,thereby potentially increasing the survival rate of critically ill COVID-19 patients.

Improved lightweight road damage detection based on YOLOV5

There is a problem of real-time detection difficulty in road surface damage detection. This paper proposes an improved lightweight model called GRC-YOLOv5s based on You Only Look Once Version5 (YOLOV5). Firstly， this paper fully utilized the convolutional neural network (CNN) + ghosting bottleneck (G＿bneck) architecture to reduce redundant feature maps. Afterwards， we upgraded the original upsampling algorithm to Content Aware Feature Recombination (CARAFE) and increased the receptive field. Finally， we replaced the Spatial Pyramid Pooling Fast (SPPF) module with the Basic Receptive Field Block (BasicRFB) pooling module and added dilated convolution. After comparative experiments， we can see that the number of parameters and model size of the improved algorithm in this paper have been reduced by nearly half compared to the YOLOv5s， the frame rate per second (FPS) has been increased by 3.25 times. The mean average precision(mAP@0.5～0.95) has increased by 8-17 percentage points compared to other lightweight algorithms.

Effect of heat treatment on microstructure and mechanical properties of selective laser-melted PH13-8Mo stainless steel

PH13-8Mo stainless steel powder with high sphericity and well fluidity was prepared by the plasma rotating electrode process.The formed parts with ultra-high purity were manufactured by selective laser melting.The tensile and impact mechanical properties of the printed parts under three heat treatment regimes were compared to those without heat treatment.The microstructure,grain orientation and phase composition were characterized by electron-backscatter diffraction,X-ray diffraction and transmission electron microscopy.The characterization results reveal that the addition of heat treatment makes the grains enlarge,and after different heat treatments,the average grain size from 1.51μm in the printed state increases to 2.78,3.09 and 2.06μm,respectively.The formed parts are mainly composed of martensite and retained austenite.Moreover,the NiAl and M23C precipitates form,which are the major strengthening phases of PH13-8Mo stainless steel.The optimal heat treatment process is 925℃×1 h water cooling(WC)+0℃×2 h air cooling(AC)+540℃×4 h AC.Using this heat treatment process,PH13-8Mo formed parts have the optimal comprehensive mechanical properties:the tensile strength,yield strength and impact energy KU2 are 1492 MPa,1432 MPa and 63 J,respectively.The strengthening and toughening via heat treatment are mainly attributed to dislocation strengthening,the formation of NiAl and M23Co precipitates and the change of volume fraction of the retained austenite.

Dear Editor,This letter studies the input-to-state stability(ISS)for a class of impulsive switched systems,where uncertain impulse-switching moments are involved.The robustness of ISS with respect to the perturbations of the occurrence time of impulse-switching moments is revealed by several less conservative dwell-time conditions for the uncertain impulse-switching moments combined with Lyapunov conditions.Moreover,the Lyapunov conditions have multiple coefficients at discrete time so as to handle the hybrid effect of impulse-switching moments,and the case that time derivative of Lyapunov function is indefinite is also taken into account.Finally,a numerical example is proposed to illustrate the effectiveness of the theoretical results.

Establishment of a transformation system in close relatives of wheat under the assistance of TaWOX5

Species closely related to wheat are important genetic resources for agricultural production,functional genomics studies and wheat improvement.In this study,a wheat gene related to regeneration,TaWOX5,was applied to establish the Agrobacterium-mediated transformation systems of Triticum monococcum,hexaploid triticale,and rye(Secale cereale L.)using their immature embryos.Transgenic plants were efficiently generated.During the transformation process,the Agrobacterium infection efficiency was assessed by histochemical staining forβ-glucuronidase(GUS).Finally,the transgenic nature of regenerated plants was verified by polymerase chain reaction(PCR)-based genotyping for the presence of the GUS and bialaphos resistance(bar)genes,histochemical staining for GUS protein,and the QuickStix strip assay for bar protein.The transformation efficiency of T.monococcum genotype PI428182 was 94.4%;the efficiencies of four hexaploid triticale genotypes Lin456,ZS3297,ZS1257,and ZS3224 were 52.1,41.2,19.4,and 16.0%,respectively;and the transformation efficiency of rye cultivar Lanzhou Heimai was 7.8%.Fluorescence in situ hybridization(FISH)and genomic in situ hybridization(GISH)analyses indicated that the GUS transgenes were integrated into the distal or near centromere(proximal)regions of the chromosomes in transgenic T.monococcum and hexaploid triticale plants.In the transgenic hexaploid triticale plants,the foreign DNA fragment was randomly integrated into the AABB and RR genomes.Furthermore,the transgene was almost stably inherited in the next generation by Mendel’s law.The findings in this study will promote the genetic improvement of the three plant species for grain or forage production and the improvement of cereal species including wheat for functional genomics studies.

Non-B-form DNA is associated with centromere stability in newly-formed polyploid wheat

Non-B-form DNA differs from the classic B-DNA double helix structure and plays a crucial regulatory role in replication and transcription.However,the role of non-B-form DNA in centromeres,especially in polyploid wheat,remains elusive.Here,we systematically analyzed seven non-B-form DNA motif profiles(A-phased DNA repeat,direct repeat,G-quadruplex,inverted repeat,mirror repeat,short tandem repeat,and Z-DNA)in hexaploid wheat.We found that three of these non-B-form DNA motifs were enriched at centromeric regions,especially at the CENH3-binding sites,suggesting that non-B-form DNA may create a favorable loading environment for the CENH3 nucleosome.To investigate the dynamics of centromeric non-B form DNA during the alloploidization process,we analyzed DNA secondary structure using CENH3 ChIP-seq data from newly formed allotetraploid wheat and its two diploid ancestors.We found that newly formed allotetraploid wheat formed more non-B-form DNA in centromeric regions compared with their parents,suggesting that non-B-form DNA is related to the localization of the centromeric regions in newly formed wheat.Furthermore,non-B-form DNA enriched in the centromeric regions was found to preferentially form on young LTR retrotransposons,explaining CENH3's tendency to bind to younger LTR.Collectively,our study describes the landscape of non-B-form DNA in the wheat genome,and sheds light on its potential role in the evolution of polyploid centromeres.
